Three varieties of Arachis hypogeae, GG 11, GG 20 and GG 24, were compared for resistance against A. niger. GG 20 showed the least disease severity. Infection with A. niger resulted in a rapid increase in NADPH oxidase, Glutathione reductase (GR) and salicylic acid in all the three varieties, indicating hyper increase of reactive oxygen species (ROS) and activation of phenyl propanoid pathway. Ferric reducing antioxidant power value was found to be decreasing due to infection in all the three varieties, confirming the role of ROS in pathogenesis. Since A. niger was found to cause pathogenesis by oxidative stress, the treatment of zinc was given as an antioxidant and its effect was studied. The application of zinc inhibited NADPH oxidase and GR activity in the control as well as in the infected GG 11 and GG 24 varieties but induced in the tolerant variety GG 20. Because zinc treatment could control the ROS in GG 11 and GG 24 varieties, disease severity was reduced but in GG 20 variety, zinc treatment aggravated ROS levels and also the disease severity. The protein profile of GG 20 in comparison to GG 11 and GG 24 varieties revealed one oligomeric protein of 110 kD as one of the responsible factors for its resistance. Total oil and its iodine value were found little higher in GG 20 variety than in other two varieties. It was found that the control of ROS could control the A. niger infection in Arachis hypogeae.  相似文献

Abstract: In this study we examined the effect on oligodendroglial survival of exogenous cystine deprivation. Oligodendroglia isolated from mixed glial primary cultures derived from brains of 1-day-old rats, and then grown for 3 days, were markedly dependent on extracellular cystine for survival. The EC50 values for cystine for a 24-h exposure ranged from 2 to 65 µ M . After 6 h of cystine deprivation, the cellular glutathione level decreased to 21 ± 13% of the control. Free radical scavengers (α-tocopherol, ascorbate, idebenone, and N-tert -butyl-α-phenylnitrone) were protective against cystine deprivation but had no effect on the glutathione level. An iron chelator, desferrioxamine mesylate, also was protective. These findings suggest that intracellular hydroxyl radicals are important for this toxicity. In contrast to the observations in 3-day-old cultures, the dependence on exogenous cystine for cell viability was not observed consistently in oligodendroglia cultured for 6 days before the onset of cystine deprivation. Several observations suggested that this loss of cystine dependence was due to a diffusible factor. Sensitivity to the toxicity of cystine deprivation in day 6 cultures increased as the volume of medium was increased from 0.3 to 2 ml. Furthermore, preincubation of cystine-depleted medium with astrocyte cultures eliminated the toxicity of the cystine deprivation. HPLC assay of the conditioned cystine-depleted medium showed no significant change in cystine or cysteine concentration. We conclude that oligodendroglia are highly susceptible to cystine deprivation in day 3 cultures and that this susceptibility is due to the accumulation of intracellular free radicals in the setting of glutathione depletion. The resistance of day 6 oligodendroglial cultures is caused at least in part by a diffusible factor.  相似文献

In order to clarify the mechanisms of selenocysteine incorporation into glutathione peroxidase, some evidence to show the in vitro conversion of phosphoseryl-tRNA to selenocysteyl-tRNA is reported. [3H]Phosphoseryl-tRNA was incubated in a reaction mixture composed of SeO2, glutathione and NADPH in the presence of selenium-transferase partially purified. Analyses of amino acids on the product tRNA showed that a part (4%) of [3H]phosphoseryl-tRNA was changed to [3H]selenocysteyl-tRNA. The conversion from seryl-tRNAsu or major seryl-tRNAIGA was not found. Selenium-transferase was essential for the conversion. [3H]Selenocysteine, liberated from the tRNA, was modified with iodoacetic acid. The product was confirmed to be carboxymethyl-selenocysteine by two-dimensional TLC. Selenocysteyl-tRNAsu should be used to synthesize glutathione peroxidase by co-translational mechanisms.  相似文献

The effects of two glycosylated whey hydrolysates (GWH-Gal A and GWH-Gal B) on glutathione (GSH) and related antioxidant enzymes in SGC-7901 cells were evaluated. Two whey glycosylated hydrolysates promoted an increase in reduced glutathione (GSH) in normal SGC-7901 cells. GSH, glutathione peroxidase (GPx), γ-glutamine cysteine synthetaase (γ-GCS), and catalase (CAT) at 1.0 and 2.0 mg/mL in normal SGC-7901 cells were higher in the GWH-Gal A group than in the GWH-Gal B group (P < 0.05). Compared with GWH-Gal B, GWH-Gal A more strongly inhibited decreases in intracellular GSH, GPx, γ-GCS, CAT, and superoxide dismutase (SOD) in H2O2-induced SGC-7901 cells. Compared with GWH-Gal B, GWH-Gal A at 1.0 and 2.0 mg/mL effectively inhibited increases in lactate dehydrogenase (LDH) and malondialdehyde (MDA) in H2O2-induced SGC-7901 cells (P < 0.05). Therefore, GSH content and related antioxidant enzyme activity levels (GPx, γ-GCS, CAT, SOD) in both normal and H2O2-induced SGC-7901 cells were considerably stronger in the GWH-Gal A group than in the GWH-Gal B group.  相似文献

Abstract: Prostaglandin H-E isomerase (EC 5.3.99.3) was purified from human brain cytosol. Purification was by ammonium sulfate fractionation, diethylaminoethyl-Sephar-ose chromatography, gel filtration on a BioGel P-100 column, GSH-agarose chromatography, and MonoQ chromatography. The activity was eluted in two peaks from the MonoQ column, which were designated peaks 1 and 2. The molecular weights of peaks 1 and 2, determined by gel filtration, were 42,000 and 44,000, respectively. On sodium dodecyl sulfate-polyacrylamide gel electrophoresis, peak 1 showed two bands at the molecular weights of 24,500 and 25,000, and peak 2 showed a single band at the molecular weight of 25,000, results suggesting that both were dimeric proteins. The pI values of both enzymes were ∼5.4. The enzymes catalyzed selective conversion of prostaglandin H2 to prostaglandin E2. The K m values for prostaglandin H2 of peaks 1 and 2 were 147 and 308 μ M , respectively, and the V max values were 380 and 720 nmol/min/mg of protein, respectively. GSH was required for the catalysis of both enzymes, and no other sulfhydryl compounds could support the reaction. A part of glutathione S -transferase (EC 2.5.1.18) was copurified with peaks 1 and 2 of prostaglandin H-E isomerase. Prostaglandin H-E isomerase activity of peak 2 enzyme was competitively inhibited by 1-chloro-2,4-dinitrobenzene, a substrate of glutathione S -transferase. These results suggested that prostaglandin H-E isomerases in human brain cytosol were identical with anionic forms of glutathione S -transferase.  相似文献

The content of polyunsaturated fatty acids, the activities of superoxide dismutase (SOD), glutathione peroxidase, glutathione reductase, and catalase, and the concentration of reduced glutathione were measured in cerebral microvessels isolated from rat brain. Polyunsaturated fatty acids, mainly arachidonic, linoleic, and docosahexaenoic acids, accounted for 32% of total fatty acids in cerebral microvessels. Whereas total SOD activity in the microvessels was slightly lower than that found in cerebrum and cerebellum, glutathione peroxidase and glutathione reductase activities were twice as high and catalase activity was four times higher. Glutathione peroxidase in microvessels is active on both hydrogen peroxide and cumen hydroperoxide, and it is strongly inhibited by mercaptosuccinate. After several hours of preparation, the concentration of reduced glutathione in isolated microvessels was 0.7 mumol/mg of protein, which corresponds to a concentration of approximately 3.5 mM. Our results indicate that the blood-brain barrier contains large amounts of peroxide-detoxifying enzymes, which may act, in vivo, to protect its highly polyunsaturated membranes against oxidative alterations.  相似文献

Previous studies indicated that DL-buthionine sulfoximine (DL-BSO), an agent that inhibits the biosynthesis of GSH in liver and other peripheral organs, fails to suppress levels of GSH in the CNS. In the current study, preweanling mice responded to repeated injections of L-BSO with marked declines (79.6-86.5%) of GSH content in brain and spinal cord. In adult mice, the same treatment schedule produced only modest declines (17.8-29.2%) of GSH content in brain and a 55.9% decline in spinal cord. Pretreatment of preweanling mice with L-BSO represents a tool for studying the role of GSH in the CNS.  相似文献

Glutathione synthetase activity (EC 6.3.2.3) was analysed in ammonium sulfate precipitates of extracts l'rom photohetevotrophically grown cells of Nicotiana tabactm L. cv. Samsun by determination of glutathione as its monobromobimane derivative. Maximal enzyme activity was obtained at pH 8.0–9.0 in Tris-HCl and CHES as buffer systems. The enzyme showed an absolute requirement for Mg2+ and was slightly stimulated by K+. When Mg2+ was replaced by Mn2+ less synthetase activity was observed, and above 30 m M Mn2+ no activity was found. The enzyme was specific for glycine (KM = 0.308 m M ). No product formation was observed with ß -alanine and γ y-aminobutyrate using substrate conccntrations of 10 m M . The apparent KM values for γ -glutamylcysteine and γ -glutamyl- l -α-aminobutyrate were, respectively, 0.022 and 0.033 m M . By chloroplast Isolation ca 24% of the total glutathione synthetase activity of the cells could be shown to be localized in the chloroplasts, the rest being attributed to the cytoplasm of the tobacco cells.  相似文献
