In n previous report dealing with the pathology of bovine fascio-liasis the author described an unknown cell type in the epithelium of bile ducts. The histological and histochemical investigations published in this paper suggest that the cell may be considered a globule leukocyte. Globule leukocytes are rare in uninfected livers but are occurring in abundance in main bile ducts of cattle with spontaneous fascioliasis and also in small perilobular ducts in dicrocoeliasis. Liver fluke infection causes an increase in the population of subepithelial mast cells. Mast cell and globule leukocyte present similarities In their cytochemical properties. However, at low pH toluldinc blue shows a stronger but Alcian blue a weaker affinity for mast cells than for globule leukocytes.  相似文献

Morphological changes of pancreatic tissue in young pigs caused by surgical ligation of the main pancreatic duct are described. Nineteen animals from 6 to 7 weeks in age were operated on and necropsied 3 or 6 to 8 weeks later. Twelve pigs developed a pronounced chronic pancreatitis with complete exocrine insufficiency. Of the 7 animals failing to develop ectasia of pancreatic ducts, 2 died due to surgical complications. In addition, 3 pigs were sham-operated and served as controls. In macroscopical studies it was observed that in the pronounced pancreatitis cases the ligated duct was greatly dilated by a clear watery fluid. Only remnants of pale and firm grandular tissues were seen around the ectatic ducts. Microscopically, typical changes of chronic pancreatitis were noted. Complete disappearance of acini was followed by ductular cell proliferations. Glandular tissues were divided into lobuli by fibrotic tissues and fat cells. The wall of the main pancreatic duct was greatly thickened and fibrotic, presenting intensely proliferating ductular cells and round cell infiltrates. Furthermore, enlarged endocrine islets surrounded by connective tissue fibres were seen.  相似文献

Recognition of extracellular peptides by plasma membrane-localized receptor proteins is commonly used in signal transduction. In plants, very little is known about how extracellular peptides are processed and activated in order to allow recognition by receptors. Here, we show that induction of cell death in planta by a secreted plant protein GRIM REAPER (GRI) is dependent on the activity of the type II metacaspase METACASPASE-9. GRI is cleaved by METACASPASE-9 in vitro resulting in the release of an 11 amino acid peptide. This peptide bound in vivo to the extracellular domain of the plasma membrane-localized, atypical leucine-rich repeat receptor-like kinase POLLEN-SPECIFIC RECEPTOR-LIKE KINASE 5 (PRK5) and was sufficient to induce oxidative stress/ROS-dependent cell death. This shows a signaling pathway in plants from processing and activation of an extracellular protein to recognition by its receptor.  相似文献

Toll-like receptor 9 mediates CpG oligonucleotide-induced cellular invasion   总被引:1,自引:0,他引:1  
Toll-like receptor 9 (TLR9) belongs to the innate immune system and recognizes microbial and vertebrate DNA. We showed previously that treatment with the TLR9-agonistic ODN M362 (a CpG sequence containing oligonucleotide) induces matrix metalloproteinase-13-mediated invasion in TLR9-expressing human cancer cell lines. Here, we further characterized the role of the TLR9 pathway in this process. We show that CpG oligonucleotides induce invasion in macrophages from wild-type C57/B6 and MyD88 knockout mice and in human MDA-MB-231 breast cancer cells lacking MyD88 expression. This effect was significantly inhibited in macrophages from TLR9 knockout mice and in human MDA-MB-231 breast cancer cells stably expressing TLR9 small interfering RNA or dominant-negative tumor necrosis factor receptor-associated factor 6 (TRAF6). Sequence modifications to the CpG oligonucleotides that targeted the stem loop and other secondary structures were shown to influence the invasion-inducing effect in MDA-MB-231 cells. In contrast, methylation of the cytosine residues of the parent CpG oligonucleotide did not affect the TLR9-mediated invasion compared with the unmethylated parent CpG oligonucleotide. Finally, expression of TLR9 was studied in clinical breast cancer samples and normal breast epithelium with immunohistochemistry. TLR9 staining localized in epithelial cells in both cancer and normal samples. The mean TLR9 staining intensity was significantly increased in the breast cancer cells compared with normal breast epithelial cells. In conclusion, our results suggest that TLR9 expression is increased in breast cancer and CpG oligonucleotide-induced cellular invasion is mediated via TLR9 and TRAF6, independent of MyD88. Further, our findings suggest that the structure and/or stability of DNA may influence the induction of TLR9-mediated invasion in breast cancer.  相似文献

Serum levels of fructosamine were assayed in 57 samples from 23 Norwegian Red Cattle dairy cows in early lactation. The animals were assigned to a 22 factorial feeding experiment. The groups differed in energy and protein supply. All samples were collected before morning feeding. The relationship between fructosamine levels and the plasma concentrations of glucose, acetoacetate, free fatty acids, total cholesterol, lipoproteins, triglycerides, total proteins, progesterone and to weight and energy balance was studied. The overall mean fructosamine level was 1.41 ± 0.20 mmol/1 ranging from 0.92–1.92 mmol/1. Significant relationships were recorded between fructosamine and the concentrations of glucose, acetoacetate, free fatty acids, triglyceride, total cholesterol and energy and weight balance. Free fatty acids and cholesterol showed the best correlation with fructosamine (rs =–0.67 and 0.70, respectively, p < 0.001). The levels of fructosamine increased significantly between 2 and 4 weeks and between 4 and 8 weeks after calving. Animals with a weight gain during 3 weeks prior to fructosamine measurement had higher fructosamine levels than those showing a weight loss (<–7 kg/week) (p < 0.05). The results indicate that serum fructosamine levels in cows may be of interest as an indicator of metabolic status in the early stages of lactation.  相似文献

The normal ultrastructure of the intestinal epithelium of F. hepatica has been described in detail e. g. by Thorsell &amp; Björk- man (1965). However, experimentally produced pathological phenomena of the epithelium have not been investigated, though such studies might yield further information on the physiological function of the epithelial cell. Light and electron microscopy of the intestinal epithelium during in vitro cultivation of the liver fluke does therefore seem to be appropriate. This preliminary report, giving an account of the cultivation procedures, deals only with light microscopy of the cultivated liver flukes.  相似文献

The effect of molybdenum (Mo) and copper (Cu) supplementation on some blood parameters and trace elements of tissues of cocks was investigated. The animals were fed with commercial poultry feed and water ad libitum and the experimental groups received peroral supplementation of different amounts of Mo or Cu during 4 weeks. Lowered values of serum uric acid were established in animals receiving 400 µg supplementation of either Mo or Cu. In contrast, the cocks receiving 100 µg supplementation of Cu displayed elevated concentrations of uric acid in the serum. A very significant statistical difference was noted between the uric acid levels of the animals receiving either moderate (group IV) or excess (group V) supplementation of Cu. The cocks with an excess of Cu displayed lowered hemoglobin and hematocrit values but no signs of Cu-intoxication were found in macro- and microscopical studies. Atomic absorption spectrophotometry was employed in the determinations of the content of Mo, Cu, Zn, Mn, Fe and & in the liver and kidneys of the cocks. The authors suggest that the results of the trace element analyses presented are to be considered as preliminary values. Furthermore, the effect of Mo and Cu on uric acid metabolism should be additionally clarified by applying histochemical studies on xanthine oxidase in different tissues of cocks in order to make conclusions on the significance of Mo and Cu in the etiology of avian gout.  相似文献
