棉花高频体细胞胚胎发生及再生体系建立

棉花的体细胞胚胎发生率低,限制了转基因棉花的发展。为了扩大可再生棉花的基因型,发展新疆优质棉花的特点,以新疆的主要栽培品种新陆早33为材料,以下胚轴为外植体,使用葡萄糖,麦芽糖作为主要碳源,用Phytagel固化,对不同激素的浓度组合和培养条件进行探索,对棉花胚性愈伤的诱导到体细胞胚胎的发生阶段进行优化,体细胞胚胎成熟以后进行萌发培养可以获得正常的植株。通过试验证明,有利于棉花的愈伤组织生长的激素浓度为KT 0.1 mg/L,2,4-D 0.1 mg/L,下胚轴的中部诱导愈伤形态最好。体细胞胚胎发生阶段以KT,IBA组合,以低盐培养基进行子叶胚的成苗,建立了再生体系。     

A noninvasive online system for biomass monitoring in shaker flasks using backward scattered light

This paper presents a noninvasive optical sensor system for monitoring cell growth in shaker flasks commonly used in biological laboratories. The system uses an open-source microprocessor board to monitor concentration of Escherichia coli host cells. To allow measurement for a range of filling degrees and shaker speeds, the backscattering angle is chosen to minimize interference from surface reflections and the measurement window is synchronized to the position of the shaker flask. A nonlinear calibration model of scattered light can predict offline optical density with a mean relative error of 5.2%, an accuracy which is comparable to the classical offline method and sufficient for biotechnology applications.     

离子注入选育高产L-乳酸的米根霉突变株及其发酵特性研究

通过氮离子注入获得米根霉突变株RQ4012,其利用木糖的能力比出发菌株提高了1.6倍;通过多次传代,证明其具有良好的遗传稳定性。试验测定菌株RQ4012发酵木糖生产L-乳酸的最佳发酵条件:木糖10%,生理盐水浸泡孢子9 h,(NH4)2SO43 g/L,接种量4%,CaCO3添加量6%,装液量20%,温度37℃,转速200 r/min,在此条件下,乳酸产量达到79.51 g/L。对混合糖的发酵进行了探索,结果表明该菌能高效利用混合糖生产L-乳酸,在利用植物纤维素水解液生产L-乳酸上有良好的应用前景。     

Autoimmune-associated PTPN22 R620W Variation Reduces Phosphorylation of Lymphoid Phosphatase on an Inhibitory Tyrosine Residue

A missense C1858T single nucleotide polymorphism in the PTPN22 gene recently emerged as a major risk factor for human autoimmunity. PTPN22 encodes the lymphoid tyrosine phosphatase (LYP), which forms a complex with the kinase Csk and is a critical negative regulator of signaling through the T cell receptor. The C1858T single nucleotide polymorphism results in the LYP-R620W variation within the LYP-Csk interaction motif. LYP-W620 exhibits a greatly reduced interaction with Csk and is a gain-of-function inhibitor of signaling. Here we show that LYP constitutively interacts with its substrate Lck in a Csk-dependent manner. T cell receptor-induced phosphorylation of LYP by Lck on an inhibitory tyrosine residue releases tonic inhibition of signaling by LYP. The R620W variation disrupts the interaction between Lck and LYP, leading to reduced phosphorylation of LYP, which ultimately contributes to gain-of-function inhibition of T cell signaling.     

Carbonate Ion-Enriched Hot Spring Water Promotes Skin Wound Healing in Nude Rats

Hot spring or hot spa bathing (Onsen) is a traditional therapy for the treatment of certain ailments. There is a common belief that hot spring bathing has therapeutic effects for wound healing, yet the underlying molecular mechanisms remain unclear. To examine this hypothesis, we investigated the effects of Nagano hot spring water (rich in carbonate ion, 42°C) on the healing process of the skin using a nude rat skin wound model. We found that hot spring bathing led to an enhanced healing speed compared to both the unbathed and hot-water (42°C) control groups. Histologically, the hot spring water group showed increased vessel density and reduced inflammatory cells in the granulation tissue of the wound area. Real-time RT-PCR analysis along with zymography revealed that the wound area of the hot spring water group exhibited a higher expression of matrix metalloproteinases-2 and -9 compared to the two other control groups. Furthermore, we found that the enhanced wound healing process induced by the carbonate ion-enriched hot spring water was mediated by thermal insulation and moisture maintenance. Our results provide the evidence that carbonate ion-enriched hot spring water is beneficial for the treatment of skin wounds.     

Promiscuous esterase activities of the C–C hydrolases from <Emphasis Type="Italic">Dyella ginsengisoli</Emphasis>

A C–C hydrolase gene (bphD _LA-4 ) from strain Dyella ginsengisoli LA-4 was cloned and expressed in Escherichia coli BL21 (DE3). BphD_LA-4 together with another hydrolase MfphA_LA-4, which derived from the same strain, possessed esterase activities. p-Nitrophenyl butyrate was the best substrate for both enzymes. BphD_LA-4 had high catalytic efficiency to p-nitrophenyl benzoate, whereas MfphA_LA-4 had no activity. Homology modeling and docking studies demonstrated that the proper hydrogen bond interaction was important for the reactivity of specific substrate.     

原子力显微技术成像在生物医学中的应用

原子力显微技术利用探针尖端与标本之间相互作用的力场对标本进行三维成像。这种成像可在生理条件下进行 ,可进行动态观察和标本容易制备是有别于其它成像技术如电子显微镜成像等的特点。对于细胞和生物大分子 ,能够在生理条件下成像具有重要意义。它意味着人们在认识生命本质的方法学方面 ,又向前迈出了新的一步。本文简要综述对细胞和生物大分子的成像在生物医学方面的应用。     

植物病原真菌广谱拮抗菌的筛选鉴定及发酵条件初步研究

通过对峙培养法筛选到对8种植物病原真菌具有拮抗作用的芽孢杆菌菌株,通过测量10-30℃下的抑菌圈直径发现AFB037菌株在不同温度下抑菌效果最好。通过表型和16S rDNA序列分析将AFB037菌株鉴定为多粘类芽孢杆菌。AFB037菌株抗菌成分的产生需要真菌菌体成分的诱导,提示其有效成分可能为细胞壁降解酶类。
