Electric circuit techniques are employed to derive the kinetic equations for a class of reactions occurring in complex networks as encountered in cellular systems, namely, exchange at equilibrium. The equations connecting equilibrium velocities with kinetic parameters of individual steps of an enzymatic reaction are derived in three ways : by direct chemical considerations, by showing that a form of Kirchhoff's rules is valid for exchange at equilibrium, and by employing the device known in electricity as the delta-star transformation. 相似文献

The expansion of P. halepensis from plantations into natural sites of high conservation value is becoming a frequent occurrence across the Mediterranean
zone of Israel. We studied how colonization of Pinus halepensis in natural Mediterranean habitats is related to afforestation, cattle grazing and fire. The study was conducted in a Mediterranean
garrigue (Shrubland) located in Ramat Hanadiv Nature Park, southern Mt. Carmel region, Israel. The study area (ca. 350 ha)
was divided into cells (100 × 100 m) each of which was categorized with respect to distance from planted pines, grazing (grazed
since 1990/ungrazed), fire (burned in 1980/unburned), and vegetation structure (garrigue, dense garrigue, dense woodland).
The location of colonizing pines, typically three m or more in height, was determined using an aerial photograph. Density
of colonizing pines decreased linearly with the distance from planted pines within 300 m from planted pines with a long tail
that extended out to ca 1,000 m. Over 90% of the colonizing pines that were found were located within a distance of 300 m
(56% of the park area) from planted pines. Colonization was about two times greater under grazing than without grazing. The
effects of fire and of the interaction fire × grazing were found insignificant. A separate analysis reveled that colonization
was about 2 times larger in patches of sparse woody cover than in those of dense cover. In conclusion, pine colonization was
mainly determined by the proximity to seed sources. Additionally, pine colonization was enhanced by cattle grazing probably

Recombinase-mediated cassette exchange, or RMCE, is a clean approach of gene delivery into a desired chromosomal location, as it is able to insert only the required sequences, leaving behind the unwanted ones. RMCE can be mediated by a single site-specific DNA recombinase or by two recombinases with different target specificities (dual RMCE). Recently, using the Flp–Cre recombinase pair, dual RMCE proved to be efficient, provided the relative ratio of the enzymes during the reaction is optimal. In the present report, we analyzed how the efficiency of dual RMCE mediated by the Flp–Int (HK022) pair depends on the variable input of the recombinases—the amount of the recombinase expression vectors added at transfection—and on the order of the addition of these vectors: sequential or simultaneous. We found that both in the sequential and the simultaneous modes, the efficiency of dual RMCE was critically dependent on the absolute and the relative concentrations of the Flp and Int expression vectors. Under optimal conditions, the efficiency of ‘simultaneous’ dual RMCE reached ∼12% of the transfected cells. Our results underline the importance of fine-tuning the reaction conditions for achieving the highest levels of dual RMCE. 相似文献

Question: We studied the interactive effects of grazing and dwarf shrub cover on the structure of a highly diverse annual plant community. Location: Mediterranean, semi‐arid shrubland in the Northern Negev desert, Israel. Methods: Variation in the biomass and plant density of annual species in the shrub and open patches was monitored during four years, inside and outside exclosures protected from sheep grazing, in two contrasting topographic sites: north and south‐facing slopes that differed in their dominant dwarf shrubs species: Sarcopoterium spinosus and Corydothymus capitatus, respectively. Results: Above‐ground biomass, density and richness of annual species were lower under the canopy of both shrub species compared to the adjacent open patches in the absence of grazing. Grazing reduced the biomass of annuals in open patches of both topographic sites, but not in the shrub patches. On the north‐facing slope, grazing also reduced plant density and richness in the open patches, but increased plant density in the shrub patches. At the species level, various response patterns to the combined effects of grazing and patch type were exhibited by different annuals. Protection against the direct impacts of grazing by shrub cover as well as species‐specific interactions between shrubs and annuals were observed. A conceptual mechanistic model explaining these interactions is proposed. Conclusion: In semi‐arid Mediterranean shrublands grazing and dwarf shrub cover interact in shaping the structure of the annual plant community through (1) direct impacts of grazing restricted to the open patches, (2) species‐specific facilitation/ interference occurring in the shrub patches and (3) subsequent further processes occurring among the interconnected shrub and open patches mediated through variation in seed flows between patches. 相似文献

Summary The effect of deoxyadenosine (AdR) and 5-fluorodeoxyuridine (FUdR) on the incorporation of 5-bromodeoxyuridine (BUdR) into DNA of thymine-non requiring cells of Escherichia coli was studied. This incorporation renders the cells sensitive to irradiation at near-UV light. It was found that the combination of AdR and FUdR increases incorporation of BUdR and sensitivity to irradiation.An enrichment of auxotrophs by a factor of 105 was obtained from a mixed population grown in minimal medium containing BUdR, AdR and FUdR and irradiated subsequently.Part of an M.Sc.
