Stearoyl‐CoA desaturase 1 (SCD1) plays important roles in organ development, glucose tolerance, insulin sensitivity, and cancer. Here, we examined the role of SCD1 for the differentiation of human induced pluripotent stem (hiPS) cells to liver cells by using drug inhibition and biochemical experiments. hiPS cells cultured in a pro‐hepatic medium were exposed to an SCD1 inhibitor at various stages throughout differentiation. Liver‐specific markers, specifically α‐fetoprotein, albumin and urea in conditioned medium, and hepatocyte nuclear factor 4α (HNF4α) and cytochrome P450 7A1 (CYP7A1) gene expressions and triglyceride in cellular extracts were analyzed at various development stages. Measures of hepatocyte‐specific function and triglyceride accumulation in later stages were strongly inhibited a minimum of −29% (< 0.05) by SCD1 inhibitor in the early stage of hepatic differentiation and effectively reversed (>30%, P < 0.01) by the addition of oleate. The results were also reproducible with human primary mononuclear cells (hPMN). SCD1 inhibitor had no significant effect on liver‐specific markers when it was added in the hepatic maturation stage. However, it strikingly led to higher albumin (1.6‐fold, = 0.03) and urea (1.9‐fold, = 0.02) production, and HNF4α (1.9‐fold, = 0.02) and CYP7A1 (1.3‐fold, = 0.03) expression upon incubation during the lineage‐commitment stage. Hepatic differentiation from cultured hiPS cells is sensitive to SCD1 inhibition and this sensitivity is affected by the stage of cellular differentiation. Notably, findings also indicate that this notion can be extended to hPMN. The requirement for SCD1 activity in functional differentiation of hepatocytes may have relevance for human liver disease and metabolic dysregulation.  相似文献

The Iranian species in the Chrysis pulchella and C. varidens species groups are investigated. Six species are recognized, two of which are described for the first time: Chrysis gamberoonensis Farhad, Rosa and Talebi sp. nov. and Chrysis brunneamarginata Farhad, Rosa and Talebi sp. nov.. Chrysis gamberoonensis Farhad, Rosa and Talebi sp. nov. belongs to the C. pulchella group and is recognizable within this species group by its unique blue body coloration, scattered mesosoma punctures, polished pit row with elongated pits, and a small median tooth on the lateral edge of metasomal tergum 3. Chrysis brunneamarginata Farhad, Rosa and Talebi sp. nov. belongs to the C. varidens group and is recognizable by the usually colorless apical rim of metasomal tergum 3, and the unique shape of the anterior corners of the scutellum which are enlarged, thickened and directed backward covering the axillary trough. Dichotomous keys and distributional data for Iranian species included in these species groups are provided. Chrysis schwarzi Linsenmaier, 1968 is raised to species rank. Hepcidin is a peptide hormone that plays an important role in iron metabolism. We have produced a recombinant mouse hepcidin-1 by using baculovirus expression system. Its expression yield was 25 μg/ml when cell culture media were supplemented with a protease inhibitor cocktail. The recombinant mouse hepcidin-1 and synthetic human hepcidin-25 had similar effects on reducing ferroportin expression in J774A cell line and in peritoneal macrophages. However, synthetic human hepcidin-25 was more efficient than recombinant mouse hepcidin-1 in reducing iron concentration in blood circulation (p < 0.01).  相似文献

Background

Aberrant DNA methylation as the most important reason making epigenetic silencing of genes is a main mechanism of gene inactivation in patients with colorectal cancer. In this study, we decided to identify promoter methylation status of ten genes encoding WNT negative regulators, and measure the expression of DNMT1 enzyme in colorectal cancer samples.

Results

Aberrant methylation of APC gene was statistically significant associated with age over 50 (p = 0.017), DDK3 with male (p < 0.0001), SFRP4, WIF1, and WNT5a with increasing tumor stage (p = 0.004, p = 0.029, and p = 0.004), SFRP4 and WIF1 with tumor differentiation (p = 0.009 and p = 0.031) and SFRP2 and SFRP5 with histological type (p = 0.001 and p = 0.025). The increasing number of methylated genes correlated with the expression levels of the DNMT1 mRNA.

Conclusions

The rate of gene promoter methylation of WNT pathway regulators is high in colorectal cancer cells. Hyper-methylation is associated with increased expression of the DNMT1 enzyme.  相似文献

The aim of this study was to quantify the levels of toxic heavy metals and sodium in topsoil of farmlands around the Urmia Lake. A total of 96 topsoil samples were collected, pre-treated, and analyzed for metals using ICP-AES.

Median concentrations of As, Cd, Cu, Ni, Pb, Zn, and Na in sampling sites were 5, 0.26, 30, 40, 13, 84, and 251 mg/kg, respectively. The enrichment factor, geoaccumulation index, and contamination factor of the metals ranked them in the order of: Na > Pb > Cu > As > Cd > Zn > Ni, indicating minor contamination for them except Na and Pb with moderate contamination. Furthermore, the spatial analysis indicated that Cd, Cu, Pb, Ni, and Zn had similar distribution patterns in the north and northwest lands of the lake. Principal component analysis revealed anthropogenic sources for Cd, Cu, Ni, Pb, and Zn in the soil, whereas, sources of As and Na were most likely related to the dust emitting from the dried bed of Urmia Lake and from a cement industry. Potential ecological risk index in 7% of the sampling sites was at serious or considerable pollution level, and Cd and Pb were identified as the main pollutants.  相似文献

There are gaps in our understanding of plant responses under different insect phytophagy modes and their subsequent effects on the insect herbivores’ performance at late season. Here we compared different types of insect feeding by an aphid, Lipaphis erysimi, and a lepidopteran, Plutella xylostella, and how this affected defensive metabolites in leaves of 2 Brassica species when plants gain maturity. Thiocyanate concentrations after P. xylostella and L. erysimi feeding activities were the same. Total phenolics was higher after the phloem feeder feeding than the folivore activity. The plants compensatory responses (i.e., tolerance) to L. erysimi feeding was significantly higher than the responses to P. xylostella. This study showed that L. erysimi had higher carbon than P. xylostella whereas nitrogen in P. xylostella was 1.42 times that in L. erysimi. Population size of the phloem feeder was not affected by plant species or insect coexistence. However, there was no correlation between plant defensive metabolites and both insects’ population size and biomass. This suggests that plant root biomass and tolerance index after different insect herbivory modes are not necessarily unidirectional. Importantly, the interaction between the folivore and the phloem feeder insects is asymmetric and the phloem feeder might be a trickier problem for plants than the folivore. Moreover, as both plants’ common and special defenses decreased under interspecific interference, we suggest that specialist insect herbivores can be more challenged in ecosystems in which plants are not involved in interspecific interference.  相似文献

Interference competition is a common phenomenon that occurs among adult females of different species to gain the highest food resources at the same time. In this research, extrinsic competition between Aphidius matricariae and Praon volucre on different densities and stages of two important pests of greenhouse crops in the world, Aphis gossypii and Myzus persicae, were investigated. The results of this study showed that total percentage parasitism of second, third and fourth nymphal instars of A. gossypii and M. persicae by A. matricariae, and P. volucre were affected by extrinsic competition. A combination of A. matricariae and P. volucre on A. gossypii had a negative effect on performance of these parasitoids. Although extrinsic competition reduced the efficiency of A. matricariae and P. volucre on M. persicae, total parasitism of M. persicae by A. matricariae and P. volucre (combination of two parasitoids) increased compared to A. matricariae or P. volucre acting alone. Also the results indicated that the extrinsic competition between A. matricariae and P. volucre on both A. gossypii and M. persicae was apparently more intense when presented with 25 hosts compared to 50 and 100 hosts. The results of this research show important information to determine an appropriate combination of these two parasitoid wasps for biological control of A. gossypii and M. persicae in greenhouses.  相似文献

The free-living mite species Sancassania rodionovi (Zachvatkin) (Acari: Acaridae), is a serious pest of mushrooms in Iran. Studies were conducted to examine the development of this mite in relation to temperature on two mushroom species: Agaricus bisporus Lange (button mushroom) and Pleurotus ostreatus Kummer (oyster mushroom). The developmental time of this acarid mite was studied at eight constant temperatures, ranging from 5 to 40 degrees C, and developmental rates were modeled as a function of temperature. Sancassania rodionovi completed immature development in 17.35 +/- 0.58 and 20.17 +/- 0.88 d at 25 degrees C on button and oyster mushrooms, respectively. When the mite fed on button mushroom, the rate of development increased gradually from 10 to 35 degrees C. Using a linear model, the developmental zero was estimated to be 3.50 degrees C with a thermal constant of 357.14 degree-days. The Logan 10, Briere 1, and Thermodynamic models adequately described the data for this mite and yielded R2 values >0.95; these models provided estimates of optimum temperature for development of 33.244, 32.145, and 32.148 degrees C, respectively. Understanding the influence of temperature on development of S. rodionovi is discussed with respect to pest management in mushroom production.  相似文献

Traditionally estimating pesticide effects by measuring only lethal effect may underestimate the total negative effect on beneficial arthropods and sublethal effects should be assessed to estimate the total effect of their applications. In this study, sublethal effect of the acaricide abamectin (Vermectin® 1.8% EC, Giah, Iran) on the predatory mite Phytoseius plumifer (Canestrini &; Fanzago) (Acari: Phytoseiidae) fed on Tetranychus urticae Koch was assessed in laboratory conditions. The adult predators were exposed to the residues of the acaricide on fig leaves and the LC50 value was determined based on a concentration–response analysis. The results showed that sublethal concentrations (LC10, LC20 and LC30) of abamectin severely affected the fecundity and longevity of the treated females of P. plumifer. Furthermore, reproductive and life table parameters of the subsequent generation were affected. The results indicated that adverse effects of abamectin on population growth of P. plumifer were significant, so the results from this study can be used to develop approximate guidelines for the use of abamectin in order to minimize their impact on P. plumifer and related natural enemies.  相似文献
