PGRP-S (Tag7) is an innate immunity protein involved in the antimicrobial defense systems, both in insects and in mammals. We have previously shown that Tag7 specifically interacts with several proteins, including Hsp70 and the calcium binding protein S100A4 (Mts1), providing a number of novel cellular functions. Here we show that Tag7–Mts1 complex causes chemotactic migration of lymphocytes, with NK cells being a preferred target. Cells of either innate immunity (neutrophils and monocytes) or acquired immunity (CD4+ and CD8+ lymphocytes) can produce this complex, which confirms the close connection between components of the 2 branches of immune response. 相似文献
The centromere is the DNA locus that dictates kinetochore formation and is visibly apparent as heterochromatin that bridges sister kinetochores in metaphase. Sister centromeres are compacted and held together by cohesin, condensin, and topoisomerase-mediated entanglements until all sister chromosomes bi-orient along the spindle apparatus. The establishment of tension between sister chromatids is essential for quenching a checkpoint kinase signal generated from kinetochores lacking microtubule attachment or tension. How the centromere chromatin spring is organized and functions as a tensiometer is largely unexplored. We have discovered that centromere chromatin loops generate an extensional/poleward force sufficient to release nucleosomes proximal to the spindle axis. This study describes how the physical consequences of DNA looping directly underlie the biological mechanism for sister centromere separation and the spring-like properties of the centromere in mitosis. 相似文献
Internal parasites typically are associated with a range of negative effects on their hosts, including reduced energy, which can manifest in behavioral alterations. With this in mind, we examined effects of a naturally-occurring nematode parasite, Chondronema passali, on locomotor activity level in horned passalus beetles, Odontotaenius disjunctus from Georgia, USA. This parasite is not well-studied but can number in the thousands in severely parasitized hosts. Prior study in our lab revealed that parasitized beetles actually consume more wood than unparasitized ones do, leading us to ask here, if parasitized beetles are also more physically active. Beetles were collected from nearby forests and housed individually in our lab. We created a simple tabletop arena to observe beetle locomotor activity, which was gridded and included small stones and paper objects. We allowed individual beetles to traverse the arena for 5 min and recorded the number of grid squares crossed. Then, beetles were dissected to determine parasite presence and level of infection (on a categorical scale). A total of 140 beetles were examined across three collections. Statistical analyses of locomotor activity revealed parasite severity predicted locomotor activity, but paradoxically, lightly-infected beetles were twice as active as those without this nematode. Activity diminished with increasing worm burdens thereafter, but even the group with the most severe burdens did not move less than those with no worms. From these results we conclude that this parasite does not result in overall reduction in activity, but rather it appears to come with heightened locomotion. Alternatively, this result could stem from the fact that more active beetles are simply more likely to contract the parasite.
Objective: This study has aimed to study different culture systems that might stimulate an increase in cell proliferation of normal and osteoarthritis chondrocytes from articular cartilage in rat model. Material and Methods: Three culture systems using chondrocytes embedded in alginate beads were tested: chondrocytes cultured in Dulbecco's modified Eagle's medium (DMEM) as control, a co-culture system consisting of a monolayer of de-differentiated chondrocytes as a source of mitotic factors, and an enriched medium containing culture medium obtained from a monolayer of chondrocytes and DMEM. Normal and osteoarthritis chondrocytes were stained with 5-carboxyfluorescein diacetate succinimidyl ester and were cultured in each of the three systems. After 5 days of culture cell, proliferation was detected by flow cytometry. Chondrocyte phenotype was confirmed by collagen type II and MMP-3 expression. To determine possible molecules released into the medium by the cultured chondrocyte monolayer and which would probably be involved in cell proliferation, a study of mRNA and expression of transforming growth factor-β1 (TGF-β1), fibroblastic growth factor-2 (FGF-2), epidermal growth factor (EGF), platelet derived growth factor-A (PDGF-A) and insulin-like growth factor-1 (IGF-1) proteins was conducted. Results and Conclusions: Chondrocytes in the co-culture system or in enriched medium showed an increase in proliferation; only when osteoarthritis chondrocytes were cultured in enriched medium would they display a statistically significant increase in their proliferation rate and in their viability. When chondrocytes from the monolayer were analysed, differential mRNA expression of TGF-β1 and IGF-1 was found during all passages, which suggests that these two growth factors might be involved in chondrocyte proliferation. 相似文献
This article interrogates the impact of readers’ ethnicity (Latino and non-Latino) on meanings crafted from Chicano literature. This article addresses the effect of Chicano literature on ethnic identity and worldview formation. The data source is participant observation and interviews with eighteen students in a college-level Chicano literature seminar in the United States. Latino students found that the literature spoke to their experience as Latinos, inciting a sense of membership in a community that merits inclusion in a college course. This connection with the literature transmuted into ethnic validation for nearly all Latino students. Non-Latinos, hindered from relating to the course content on an ethnic level, discovered alternate entryways into the texts that allowed for identification on other particular levels. Non-Latinos ascertained how ethnic distinctions partially shape human experience and that this understanding is critical to cross-cultural appreciation and empathy. Non-Latinos additionally perceived that a common humanity tunnels beneath ethnic difference. 相似文献
In a three-hour bioassay, we tested the palatability and feeding preferences of Uresiphita maorialis (kōwhai moth) for Sophora tetraptera, Sophora microphylla and Sophora prostrata. Palatability tests showed no differences among the Sophora species. Feeding preferences, on the other hand, showed that S. tetraptera and S. microphylla leaves are preferred over S. prostrata leaves. Our results support our field observations in Wellington city parks and gardens showing that S. tetraptera and S. microphylla plants frequently have higher densities of larvae than S. prostrata. 相似文献
We studied effects of habitat structure on routine travel velocities,
intermittent locomotion, and vigilance by the degu (Octodondegus), a diurnal rodent of central Chile. We predicted thattravel speed, pauses during locomotion, and vigilance wouldbe greater in open (riskier) than in shrub (safer) habitats.Video recordings of marked individuals in the wild were used
to measure speed and other variables of spontaneous locomotionnot triggered by predatory attack or any other noticeable stimulusduring nonforaging periods. Time spent vigilant while foragingwas also measured. Because degus use bare-ground runways fordistant movements (e.g., between burrow openings and/or foodpatches), data on locomotion decisions were not confounded by
effects of obstructive vegetation cover and/or resource abundance.When moving across the habitat between different feeding places,degus showed an intermittent pattern of locomotion, interruptingrunning events with short pauses. As predicted, travel speedand the duration of pauses between locomotion bursts were significantlygreater in open habitats. Further, the duration of locomotionbursts between feeding sites or between feeding sites and burrowswas significantly longer in open habitats. Our assumption that
pauses and velocities are independent decisions was supportedby the lack of correlation between pauses and speeds duringlocomotion events. During foraging, degus devoted more timeto vigilance in open than in shrub habitats. The static positionadopted by degus during pauses, the speeds attained during
movements, and the concordance between pausing behavior andvigilance across habitats suggest that pausing has an antipredatoryrole and is not limited to orientation and/or physiologicalrecovery. Our results support the view that degus perceivehigher predation risk in open areas and that flexible movement
