Background

Artemisinin analogue SM934 was previously reported to possess immunosuppressive properties. The aim of this study was to determine the effects and the underlying mechanisms of SM934 in murine experimental autoimmune encephalomyelitis (EAE).

Methods

Female C57BL/6 mice immunized with MOG35–55 were treated with or without SM934, then the clinical scores and other relevant parameters were assessed. Th1, Th17 and regulatory T (Treg) cell profiles were determined through ELISA, qRT-PCR, flow cytometry and BrdU incorporation assay. The effects of SM934 on Th1, Th17 and Treg cells differentiation were explored through intracellular staining and flow cytometry examination.

Results

In vivo, administration of SM934 significantly inhibited the development of EAE and suppressed the elevation of serum IL-17. Ex vivo, upon antigen-recall stimulation, IL-2, IFN-γ, IL-17 and IL-6 production were decreased, whereas IL-10 and TGF-β production were increased from the splenocytes isolated from SM934-treated mice. Consistently, both flow cytometry and qRT-PCR results showed that SM934 treatment significantly increased the Treg, while strongly suppressed the Th17 and Th1, responses in the peripheral. Furthermore, in the spinal lesion, SM934 treatment dramatically decreased the infiltration of CD4+ T cells, within which the Treg cells percentage was enlarged, whereas the Th17, but not Th1 percentage, was significantly decreased comparing with the vehicle-treated groups. Finally, both BrdU incorporation and in vitro Treg differentiation assays revealed that SM934 treatment could directly promote the expansion of Treg cells in vivo and in vitro.

Conclusion

Taken together, this study demonstrated that SM934 treatment could ameliorate the murine EAE disease, which might be mediated by inducing Treg differentiation and expansion.  相似文献

Breast cancer (BC) is the most prevalent malignant cancer in the world, is the leading cause of cancer-related death female. Recently, there is accumulating evidence that long noncoding RNAs (lncRNAs) might as an important role in the progression of BC. (epithelial-mesenchymal transition (EMT) is considered to play a vital role in tumor cells migration and invasion. Nevertheless, the entire biological mechanisms and functions of lncRNAs in tumor migration, invasion, and EMT remain uncertain. In the present research, we observed that the expression of lncRNA AC073284.4 was downregulated in BC paclitaxel-resistant (PR) cells (MCF-7/PR) and tissues. Bioinformatics analysis predicted that miR-18b-5p was a direct target of AC073284.4, which has been validated by dual-luciferase reporter gene assay. We further proved that AC073284.4 could directly bind to miR-18b-5p and relieve the suppression for dedicator of cytokinesis protein 4 (DOCK4). Furthermore, the underlying functional experiments demonstrated that AC073284.4 might sponge miR-18b-5p to attenuate the invasion, metastasis, and EMT of BC cell through upregulating DOCK4 expression. In summary, AC073284.4 might serve as a competing endogenous RNA (ceRNA) in BC progression via modulating miR-18b-5p/DOCK4 axis, which weakens EMT and migration of BC. These results suggesting that AC073284.4 might function as a potential novel diagnostic biomarker in the progression of BC.  相似文献

皮肤类器官作为一种新型的类器官模型,不仅能高度模拟皮肤组织的生理结构和功能,更好地在不同体外环境下还原较真实的皮肤生态,还可以应用于皮肤发育研究、皮肤疾病病理研究及药物筛选等领域。在干细胞研究中,皮肤类器官模型可以在特殊的生境下对具有特定功能的皮肤细胞及其附属物进行重建和改造,以弥补现有体外皮肤模型在结构、功能等方面的不足。基于此,皮肤类器官将会在皮肤再生、组织修复、药物筛选及医学美容等方面扮演越来越重要的角色。本文详述了皮肤类器官构建中所参与的细胞来源及近年来的应用,并对未来皮肤类器官的发展与优化做出了展望。  相似文献

生长因子受体结合蛋白10(Growth factor receptor-bound protein 10, Grb10)是一个存在于小鼠11号染色体和人7号染色体的母本表达的印记基因。文章利用原位杂交技术和定量RT-PCR方法对不同发育阶段的小鼠胚胎Grb10 基因进行时空表达谱的分析, 以确定该基因在胚胎发育中其表达与组织发育的关系。定量RT-PCR数据结果表明, Grb10在E8.5-E13.5 (Embryonic days 8.5-13.5), 表达水平逐渐增高, 在E13.5达到高峰, 后期表达量回落。在胚胎发育的中后期脑、心、肺组织的表达水平呈递减趋势。肝脏组织中Grb10表达水平较为恒定, 在E18.5出现高峰。原位杂交数据验证了定量RT-PCR的结果, 并且说明了Grb10在其他如骨、肾和肌肉等组织器官的高表达水平。研究结果表明Grb10基因是小鼠胚期发育的一个重要的基因。  相似文献

中缝核5-羟色胺能神经元通过其广泛的神经投射影响大脑多方面的功能,包括抑郁和焦虑、睡眠-觉醒周期、奖赏、决策中的耐心以及性别取向等.背侧中缝核和中央中缝核的5-羟色胺能神经元对嗅球有密集的神经投射,从而调控嗅觉信息的初步表征和编码.近年来,随着电生理、光学成像及光遗传技术的应用,关于中缝核5-羟色胺能神经元对嗅球的调制作用研究不断出现,大量离体和在体实验证据表明中缝核5-羟色胺能神经元对嗅球及嗅觉相关行为有广泛的调制.本文从嗅球不同神经元类型角度,就中缝核5-羟色胺能神经投射对嗅球的调控作用及其神经机制研究进展进行了总结.  相似文献

以毛茛科乌头属铁棒锤(Aconitum pendulum N.Busch)2个品系‘蓝花铁棒锤’(‘WSYB1’)和‘黄花铁棒锤’(‘WSYY1’)为材料,对其进行转录组测序(RNA-seq),采用生物信息学方法鉴定其中可能存在的花柱S基因(self-incompatibility gene)和花粉S基因,并对它们的序列特征进行分析。结果显示,转录组中共鉴定出2个在雌蕊中特异或高表达的花柱S基因(ApSRNase)和2个在雄蕊中特异表达的花粉S基因(ApSLF)。与耧斗菜(Aquilegia coerulea James)相似,铁棒锤中也存在S-RNase(S locus ribonucleases)和SLF(S locus F-box)控制的S-RNase类的自交不亲和系统,而不存在sS(stigma S-determinant)和pS(pollen S-determinant)控制的罂粟科类型的自交不亲和系统。  相似文献

背散射电子成像 (BackscatteredElectronImaging,简称BSE),是依托扫描电镜的一种电子成像技术,它的成像原理和特点非常适合用来研究那些表皮尚存的各类笔石标本,是二次电子成像 (SEM)无法替代的。文中简要介绍了这一技术的工作原理,并利用它对我国早奥陶世和中奥陶世部分笔石的始端发育和分枝方式等进行了研究。当前BSE图象显示了许多以往其他途径无法观察到的笔石微细结构,特别是笔石复杂的始端发育特征,结果验证了Psigraptusjacksoni的二分岔式和Rhabdinoporaflabelliformisparabola的四分岔式原始分枝的观点,显示它们都具有最原始的等称笔石式(isograptid type)始端发育型式。此外,BSE图象还显示,底栖固着的树形笔石Airograp tusfurciferus的胞管排列和分枝方式与奥陶纪最早期的漂浮笔石非常相似,表明二者可能存在祖 裔演化关系,或者至少是原始性状上的相似性。  相似文献
