Among the major constituents of Leea rubra (Family Vitaceae) leaves, phenolic and flavonoind compounds are most important for therapeutic purposes and the plant parts have been used in traditional medicine to treat several diseases for long. Thus, in order to scientifically confirm the traditional uses of the L. rubra leaves, the present study was designed to investigate the efficacy of the isolated flavones against AAPH induced oxidative damage to pUC19 DNA by gel electrophoresis and antineoplastic activity was evaluated on Ehrlich ascites carcinoma (EAC) bearing Swiss albino mice by evaluating percentage inhibition of cell growth, morphological changes of EAC cells and hematological parameters of the mice. The isolation was carried out by column chromatography and structure was revealed by 1H-NMR and 13C NMR. The result shows that, the isolated compound was identified as myricetin 4'-methoxy-3-O-α-l-rhamnopyranoside based on previously reported data. The isolated flavone effectively inhibited AAPH-induced oxidative damage to DNA; because it could inhibit the formation of circular and linear forms of the DNA. In anti-proliferative assay, 76% growth inhibition of EAC cells was observed as compare to the control mice (p<0.05) at a dose 100 mg/kg body weight. Thus the isolated flavone showed great importance as a possible therapeutic agent in preventing oxidative damage to DNA and the chronic diseases caused by such DNA damage, and can also become important in cancer chemotherapy.  相似文献

Staphylotrichum boninense, a new hyphomycete classified in the Chaetomiaceae (Ascomycota), was isolated from soils in the Bonin Islands, Japan. It is characterized morphologically by the production of yellow-orange colonies and subglobose holoblastic conidia. Morphologically the species is similar to S. coccosporum, but it is significantly different from S. coccosporum in phylogeny and also differs with respect to its secondary metabolite profile.  相似文献

The distribution of contractile forces generated in cytoskeletal stress fibers (SFs) contributes to cellular dynamic functions such as migration and mechanotransduction. Here we describe a novel (to our knowledge) method for measuring local tensions in SFs based on the following procedure: 1), known forces of different magnitudes are applied to an SF in the direction perpendicular to its longitudinal axis; 2), force balance equations are used to calculate the resulting tensions in the SF from changes in the SF angle; and 3), the relationship between tension and applied force thus established is extrapolated to an applied force of zero to determine the preexisting tension in the SF. In this study, we measured tensions in SFs by attaching magnetic particles to them and applying known forces with an electromagnetic needle. Fluorescence microscopy was used to capture images of SFs fluorescently labeled with myosin II antibodies, and analysis of these images allowed the tension in the SFs to be measured. The average tension measured in this study was comparable to previous reports, which indicates that this method may become a powerful tool for elucidating the mechanisms by which cytoskeletal tensions affect cellular functions.  相似文献

A fungal strain, FOM-8108, that was isolated from sea sand was found to produce gentisylquinone and chlorogentisylquinone, inhibitors of neutral sphingomyelinase. The fungus grew well under normal conditions, in the darkness, or on various agar media, but typical morphological characteristics were not observed to determine its taxonomy. Therefore, culture conditions were studied extensively, resulting in formation of a number of pycnidia by the fungus on the media containing seawater or on natural substrates such as hydrangea leaves, gardenia leaves, and rice straw under natural light or near-ultraviolet radiation exposure. Eventually, from the morphological characteristics of pycnidia, conidiogenous cells, and conidia, strain FOM-8108 was considered to belong to the genus Phoma. Culture studies showed seawater in the fermentation medium was necessary for the strain to produce gentisylquinones. Particularly, a full production of chlorogentisylquinone, which has a chloride ion in the structure, was performed in the medium with higher concentration (75%–100%) of seawater. Lecanicillium primulinum, a new hyphomycete classified in the Cordycipitaceae (Hypocreales, Ascomycota), was isolated from soils collected from the Okinawa's main island and the Bonin Islands, Japan. Using a combination of micro-morphological characteristics and sequences of the ribosomal RNA genes and ITS regions, the isolates were identified as a species of Lecanicillium which was previously undescribed. Phylogenetically the species is close to L. acerosum and Lecanicillium sp. 1 and morphologically it is similar to L. longisporum, L. psalliotae and Lecanicillium sp. 1, but its microconidia differ and it is clearly separated phylogenetically.  相似文献

Eighteen Simplicillium isolates were discovered from soil samples collected on the Honshu, Bonin, and Izu islands in areas under the jurisdiction of Tokyo, Japan. Using a combination of micro-morphological characteristics and sequences of the ribosomal RNA gene ITS region, the isolates were classified as six Simplicillium species, and five of them were previously undescribed. The five species (Simplicillium aogashimaense, Simplicillium cylindrosporum, Simplicillium obclavatum, Simplicillium subtropicum and Simplicillium sympodiophorum) were discovered from the Chichi-jima, Hachijo, and Aogashima islands, and Simplicillium minatense was discovered from Honshu. The five new species and three known species are distinguished by conidial morphology.  相似文献

Fungal strain FKA-25, isolated from forest soil collected on Yakushima Island, Kagoshima Prefecture, Japan, was assigned to genus Pseudobotrytis based on its morphological characteristics. Conidiophores were erect, slightly swollen at the end of the tip, and gave rise to umbellate conidiogenous cells that were in an expanded denticulate portion at the end and formed ellipsoidal to clavate conidia in sympodial succession. Identification as species P. terrestris was made on the basis of the character of 1-septate conidium. Although no secondary metabolites have been reported from the genus Pseudobotrytis, four secondary metabolite compounds (designated A to D) were isolated from the culture broth of strain FKA-25. Compounds B to D have been reported previously as FK-17-p2a, lunatinin, and 3,4-dihydro-3,4,8-trihydroxy-1(2H)-naphthalenone, respectively. Compound A was designated sespendole and possessed a novel indole-sesquiterpene skeleton.  相似文献

Embryonic and morphological development of larvae and juveniles of the amberjack,Seriola dumerili Risso, are described using specimens raised at Yaeyama Station (Ishigaki Island, Okinawa Pref.), Japan Sea Farming Association. The specimens obtained from brood fish (3 females, 3 males) were treated with gonadotropin and spawned on 6th of April 1987. The eggs of amberjack are pelagic, spherical in shape and 1.01–1.17 mm in diameter. The yolk is roughly segmented and has a single oil globule 0.22–0.24 mm in diameter. The perivitelline space is narrow. During development, a few melanophores and no xanthophores were observed on yolk. Hatching took place 35 hrs. 15 min. after spawning out at temperatures 23.1–23.7°C. The newly hatched larvae were 2.84–3.04mm in TL with 27 (13+14) myomeres and an oil globule anteriorly situated beyond the head. 3 days after hatching 4.00 mm TL, the mouth opened. 10 days after hatching 4.26 mm TL, small denticles appeared on the margin of the upper jaw and there were 1 anterior and 2 posterior preopecular spines. At 5.96mm TL, notochord was slightly flexed. Caudal, dorsal and anal fins with rudiments of rays appeared at 8.00 mm TL. The specific numbers of all fin rays and spines were obtained in a juvenile 9.60 mm TL. In a juvenile 34.25 mm TL, 54 days after hatching, the characteristic brown band of amberjack had appeared on head. Some notable changes in relative growth were observed at 5 mm and 15 mm in TL.  相似文献

Novel bottromycin derivatives were synthesized from bottromycin A2 via a hydrazide derivative as a common intermediate. Seventeen derivatives were subjected to in vitro evaluation against drug-resistant gram-positive bacteria. Some compounds showed potent anti-MRSA and anti-VRE activity, as did bottromycin A2. Notably, a propyl ketone derivative exhibited good antibacterial activity with excellent metabolite stability.  相似文献

Stimulation of leucomycin production with Streptomyces kitasatoensis by magnesium phosphate (MgP), and a concomitant decrease of ammonium ion concentration in the culture supernatant have been reported from this laboratory. Cultural characteristics of the MgP-supplemented culture were further examined in relation to the stimulation of leucomycin production. When the leucomycin titer increased 4- to 5-fold in the presence of MgP, mycelial growth level and assimilation rate of a major carbon source (glycerol) increased about 2-fold. MgP-grown mycelia were not appreciably different in shape or in carbon and nitrogen contents from the control mycelia. Magnesium and phosphate ions were released from MgP into the medium, while the addition of magnesium or phosphate salts to the control medium was rather inhibitory for leucomycin production. The pH values of MgP-supplemented cultures were lower than those of the control culture. When modifications of the basal media and culture conditions were done so as to permit the pH-change almost identical with that of the control media, the leucomycin titer was still higher in the presence of MgP than in its absence. When uric acid, an insoluble compound, was used as the sole nitrogen source, the leucomycin production increased to a high level even in the absence of MgP, which was comparable to that seen when MgP was added to the ammonium lactate-containing medium. The ammonium ion concentration in the culture supernatant of the uric acid-containing medium was maintained at a considerably low level. While in the ammonium lactate-containing medium, leucomycin production was low and ammonium ion concentration remained at a high level. Thus, it was demonstrated that the stimulation of leucomycin production by MgP is related to the lowering of ammonium ion level, but not to the other factors examined such as growth level, carbon or phosphate regulation and pH-effect.  相似文献
