PCTAIRE-1 (cyclin-dependent kinase [CDK] 16) is a highly conserved serine/threonine kinase that belongs to the CDK family of protein kinases. Little is known regarding PCTAIRE-1 regulation and function and no robust assay exists to assess PCTAIRE-1 activity mainly due to a lack of information regarding its preferred consensus motif and the lack of bona fide substrates. We used positional scanning peptide library technology and identified the substrate-specificity requirements of PCTAIRE-1 and subsequently elaborated a peptide substrate termed PCTAIRE-tide. Recombinant PCTAIRE-1 displayed vastly improved enzyme kinetics on PCTAIRE-tide compared to a widely used generic CDK substrate peptide. PCTAIRE-tide also greatly improved detection of endogenous PCTAIRE-1 activity. Similar to other CDKs, PCTAIRE-1 requires a proline residue immediately C-terminal to the phosphoacceptor site (+1) for optimal activity. PCTAIRE-1 has a unique preference for a basic residue at +4, but not at +3 position (a key characteristic for CDKs). We also demonstrate that PCTAIRE-1 binds to a novel cyclin family member, cyclin Y, which increased PCTAIRE-1 activity towards PCTAIRE-tide >100-fold. We hypothesised that cyclin Y binds and activates PCTAIRE-1 in a way similar to which cyclin A2 binds and activates CDK2. Point mutants of cyclin Y predicted to disrupt PCTAIRE-1-cyclin Y binding severely prevented complex formation and activation of PCTAIRE-1. We have identified PCTAIRE-tide as a powerful tool to study the regulation of PCTAIRE-1. Our understanding of the molecular interaction between PCTAIRE-1 and cyclin Y further facilitates future investigation of the functions of PCTAIRE-1 kinase.  相似文献

A survey was carried out to detect the presence of aflatoxin B1 in 60 duplicated samples (120 samples) of peanuts butter purchased from the local markets and other traditionally prepared and distributed by the street sellers in Khartoum state, Sudan. AflaTest-P affinity column was used to extract the toxin from the samples, and the concentration was measured by calibrated Vicam fluorometer. Aflatoxin B1 was detected at variable levels in 100% of the screened samples. Traditionally prepared samples showed the highest incidence of aflatoxin B1 which is above the internationally regulated tolerance levels (5–20 ppb). The means and the ranges of the aflatoxin B1 recovered were as follows: 63.9 ppb (29–128 ppb), 54.5 ppb (21–131 ppb) and 101 ppb (17–170 ppb) for samples collected from Khartoum, Khartoum North and Omdurman areas, respectively. Samples from retail stores presented relatively low aflatoxin B1 incidences 14.5 ppb (1–57 ppb), but only 30% of the samples revealed aflatoxin level below 10 ppb. Laboratory segregated and carefully prepared butter from good grade nuts showed the lowest levels of this toxin (3.3 ppb; 2–6 ppb). The results showed that peanuts butter prepared by the street sellers and distributed by the retail stores are evidently hazardous to human health. There is therefore urgent need for strong form of quality control measures and public awareness. The use of excellent grade peanuts and care during processing and storage are priority.  相似文献

Groups af White Leghorn chicks obtained from dams deprived on selenium (Se), were fed from hatching a low-Se-vitamin E basal diet alone, or supplemented with 0.02, 0.04, 0.06 or 0.08 mg Se/kg diet, as sodium selenite (Na2SeO3 · 5H2O), wheat, barley or fish meal. Prevention of the Se-vitamin E deficiency responsive disease exudative diathesis (ED) as it was clinical observed, induction of the plasma Se dependent enzyme glutathione peroxidase (GSH-Px) activity, and Se concentration in the cardiac muscle were observed to be dietary Se level and source dependent. Slope ratio assay was applied to estimate the biological availability of Se in the natural sources relative to Se in sodium selenite. For the prevention of ED, the bioavailability of Se in wheat, barley and fish meal was 99, 85 and 80 %, respectively. The increase in the plasma GSH-Px activity revealed a bioavailability for Se in wheat, barley and fish meal of 79, 71 and 66 %, respectively. Using retention of Se in the cardiac muscle as the bioassay, a bioavailability of 108, 87 and 100 % was calculated for wheat, barley and fish meal Se, respectively.  相似文献

Whole blood, red blood cells (RBC), and plasma vitamin E (VE) levels in chicks fed dietary VE (dl-α-tocopheryl acetate, dl-αTa) supplementation in steps of 0.0, 5.0, 10.0, 15.0, 20.0 and 30.0 mg/Kg were determined to examine their usefulness as an index of VE status. The increase in VE level was significant and linear in whole blood (r = 0.90), RBC (r = 0.89) and plasma (r = 0.93) in response to dietary VE intake. There was a close correlation between VE in plasma vs whole blood (r = 0.90), plasma vs RBC (r = 0.91) and whole blood vs RBC (r = 0.95). The plasma VE content was 1.2–1.8 times greater than that of whole blood, and 6.6–12.5 times greater than that of RBC. The plasma total lipids content was not affected by the dietary VE intake, whereas the level of VE in the plasma total lipids was significantly increased with increasing supplementation. Alpha tocopherol was the major isomer (ca 92 %) of VE in whole blood, RBC and plasma at hatching. The small proportions of β-tocopherol (ca 2 %), γ-tocopherol (ca 5 %) and α-tocotrienol (ca 1 %) observed at 1 day of age had decreased or totally disappeared by 7 days of age after feeding the VE-free basal diet. The data showed that in the chick, the whole blood and RBC levels of VE were as sensitive and reliable indexes of dietary VE status as was that of the plasma.  相似文献
