The tribe Naucleeae has recently been recircumscribed on the basis of both morphological and molecular [ rbcL , trnT-F , internal transcribed spacer (ITS)] evidence, and has been found to be the sister group of the tribe Hymenodictyeae Razafim. & B. Bremer. In order to find pollen morphological support for this new classification, the pollen and orbicules of 65 species, representing 23 Naucleeae and the two Hymenodictyeae genera, were investigated by scanning electron and light microscopy. Naucleeae pollen is very small (< 20 µm) to small (20–30 µm) and its shape in equatorial view is suboblate to spheroidal or, more rarely, subprolate. Three compound apertures are present, each comprising a long and narrow ectocolpus, a circular to slightly lolongate mesoporus, and an often H-shaped endoaperture. The sexine ornamentation is perforate, rugulate, or (micro)reticulate, and supratectal elements are always absent. Apart from the variation in sexine ornamentation, the tribe is rather stenopalynous. The pollen of Hymenodictyeae is very similar to that of Naucleeae. The H-shaped endoapertures often observed probably form a synapomorphy for the clade comprising Naucleeae and Hymenodictyeae. Our pollen morphological observations are not in conflict with the widened delimitation of Naucleeae. Unambiguous pollen support for the recent subtribal or generic concepts of Naucleeae could not be found because of a lack of variation of pollen characters within the tribe. Orbicules are invariably present in the ten Naucleeae taxa investigated. Pollen and orbicule morphology of 35 Dioscorea L. species is described based on observations with light microscopy, and scanning and transmission electron microscopy. Pollen and orbicule characters are critically evaluated and discussed in the context of existing hypotheses of systematic relationships within the genus. Pollen is mostly bisulcate (sometimes monosulcate) with a perforate, microreticulate or striate sexine. Our results indicate that pollen data may be significant at sectional rank. The close relationship between sections Asterotricha and Enantiophyllum proposed by Burkill and Ayensu is supported by pollen morphology as all species investigated share bisulcate, perforate pollen with small perforations and a high perforation density. Macromorphological differences between the two compound-leaved sections Botryosicyos and Lasiophyton are also supported by pollen morphology; pollens of these two sections have very different perforation patterns. Orbicules in Dioscorea are mostly spherical and possess a smooth or spinulose surface. The latter is often correlated with a striate sexine.  相似文献

Pollen and orbicule morphology of 26 species in the tribe Hillieae is described based on observations by light, scanning and transmission electron microscopy. Pollen and orbicule characters are critically evaluated and discussed in the context of existing hypotheses of systematic relationships within the tribe. Pollen is 3-zonocolporate with a perforate, microreticulate, reticulate or eutectate sexine. In the two species of Blepharidium , however, the pollen has one, four or five apertures. These pollen morphological data were incorporated into an existing macromorphological matrix of the group and cladistically analysed. The resulting phylogenies indicate that it may be appropriate to reduce Cosmibuena to a subgenus of the genus Hillia , while Blepharidium should be removed from Hillieae because of the deviating pollen type that is unique to the Rubiaceae. A detailed morphological and anatomical study of the tropical African genus Virectaria is presented. The observations are used to characterize the genus, to propose a key to all eight species, and to unravel the relationships between the species using cladistics. A taxonomic survey of the genus is also given. Verdcourt's infraspecific taxa based on calyx morphology in V major are adopted, but it was found that morphological distinction is correlated with ecological, habit and distributional differences. Hence, it is better to recognize Verdcourt's varieties spathulata and major as subspecies of V. major. The problematic systematic position of the genus is discussed in detail. New evidence is given for the exclusion of the genus from the Hedyotideae and Ophiorrhizeae (subfamily Rubioideae). The recently proposed position in the Sabiceeae (subfamily Ixoroideae) is not convincing either, since none of the genera included in the Sabiceeae matches Virectaria with respect to pollen, fruit, flower and growth habit. Exclusion from the Rubioideae and a position near the Sabiceeae is supported by lack of raphides, seed anatomy, placentation, stipule morphology and molecular evidence. Molecular data from a larger number of taxa are needed to confirm the position of the genus.  相似文献

This paper adds new data on the pollen and orbicule morphology of 61 Dioscorea L. (Dioscoreaceae) species to the survey of Schols et al . (2001). The results indicate that pollen characters may be significant in infrageneric systematics in Dioscorea . Pollen and orbicule characters are described based on observations with light microscopy, and scanning and transmission electron microscopy, and are critically evaluated and discussed in the context of existing hypotheses of systematic relationships within the genus. Pollen is mostly disulculate (sometimes monosulcate) with a perforate, microreticulate, striate, gemmate, rugulate, or cerebroid perforate sexine. The basal section Stenophora is one of the few sections with monosulcate pollen. Brachyandra , Cardiocapsa , and Seriflorae , three Malagasy sections, are characterized by striate pollen. Pollen morphology strongly supports section Enantiophyllum as a monophyletic group. The correlation between pollen size and tuber type, as suggested previously by P. Su (1987), is confirmed by our data. Two new Pauridiantha species (Pauridiantheae, Rubiaceae) from Gabon are described and illustrated. Pauridiantha pleiantha is characterized by a combination of entirely glabrous stems, stipules with a narrow base, and large inflorescences. Pauridiantha smetsiana is characterized by entirely glabrous stems, narrow, early deciduous stipules, leaves red-tinged when dry, and small inflorescences. The two new species, especially P. smetsiana , close the morphological gap between Commitheca and Pauridiantha in combining typical Commitheca characters, such as glabrous stems and a narrow stipular base, with Pauridiantha characters, such as eucamptodromous venation. A detailed morphological comparison (including ovary structure, seed morphology, exotesta anatomy, and pollen morphology) between the two genera is given; in conclusion, Commitheca is relegated to the synonymy of Pauridiantha . The extent of oxidative stress during ripening of saskatoon(AmelanchieralnifoliaNutt.) fruit was examined. Lipid peroxidation duringfruit development from the mature green to the fully ripe (purple)stage was evidenced by the accumulation of ethane and 2-thiobarbituricacid reactive substances. Fruit polar lipid and free fatty acidconcentrations also declined during ripening. Moreover, thedouble bond index of fatty acids in the polar lipid fractionfell during ripening, reflecting a progressive increase in thesaturation of membrane lipids. This increase in saturation waspartly due to a 65% decline in the concentration of linolenicacid. Activities of superoxide dismutase (SOD) and catalase(CAT) fell about 4-fold and 18-fold, respectively, during development,indicating higher potential for the accumulation of cytotoxicH2O2. Peroxidase activity remained relatively low and constantfrom the mature green to the dark red stage of development,then increased towards the end of ripening as fruits turnedpurple. Lipoxygenase (LOX) activity increased 2.5-fold fromthe mature green to the fully ripe stage. Tissue prints showedLOX to be present throughout fruit development and Western analysisrevealed that the increase in activity during ripening was dueto increased synthesis of the enzyme. Collectively, these resultsprovide evidence that ripening of this climacteric fruit isaccompanied by a substantial increase in free-radical-mediatedperoxidation of membrane lipids, probably as a direct consequenceof a progressive decline in the enzymatic systems responsiblefor catabolism of active oxygen species. The role of glutathione-mediatedfree-radical scavenging was also examined as a potential systemfor coping with this increased oxidative stress. Concentrationsof reduced and oxidized glutathione (GSSG) increased 2-foldand GSSG increased as a percentage of total glutathione, reflectingthe increase in oxidative status of fruits during ripening.Tissue prints of glutathione reductase (GRase) and transferase(GTase) showed these enzymes to be distributed throughout thepericarp at all stages of fruit development. GRase and GTaseactivities rose sharply during the later stages of fruit ripening,correlating well with substantial increases in the levels ofboth enzymes. Hence, the glutathione-mediated free-radical scavengingsystem was up-regulated towards the end of ripening, perhapsin response to the increasing oxidative stress resulting fromthe accumulation of lipid hydroperoxides from increased LOXactivity, in conjunction with a decline in SOD/CAT activities.Copyright1998 Annals of Botany Company Amelanchier alnifoliaNutt.; saskatoon fruit; ripening; oxidative stress.  相似文献
