A total of 71 lactating and nonlactating buffalo-cows of the Murrah breed and F(1)-F(3) crossbreds of Murrah x Bulgarian buffalo were used for a year as donors of embryos after a preliminary treatment for superovulation induction with pregnant mare serum gonadotrophin (PMSG) or follicle stimulating hormone (FSH) in combination with prostaglandin F-2 alpha analog (PGF-2 alpha) according to general application procedures in cows. From 36 to 72 h following prostaglandin injection, the buffalo-cows were checked with the help of a teaser bull for detection of estrus. The animals in estrus were inseminated twice either naturally or artificially with frozen semen. Nonsurgical flushing of the uterine horns was done in 45 of the buffalo-cows between 108 and 162 h after the onset of estrus. After slaughter the uterine horns and oviducts of the other 26 animals were flushed separately between 74 and 108 h after the beginning of estrus. Seven late morulae and eight hatched blastocysts were recovered between 114 and 116 h from the onset of estrus as a result of nonsurgical flushing. All of the 40 embryos recovered after 117 h were in the hatched blastocyst stage. As a result of flushing the oviducts and the uterine horns of slaughtered donors between 74 and 100 h, eggs were obtained only from the oviducts, while flushing conducted between 102 and 108 yielded eggs from both the oviducts and the uterine horns.  相似文献

We report a novel class of transmission electron microscope (TEM), the difference-contrast electron microscope (DTEM), which displays nanostructures of thin specimen objects in a topographical manner. Topography obtained by the difference-contrast develops shadowgraphs in pseudo three-dimension, namely volume-like representation of projected objects as if things are illuminated by light from one direction. The specific optical device tomanipulate electron waves for DTEM is the hemicircular phase-plate, which appears to be quite distinguishable from the Zernike phase plate utilized in Zernike phase-contrast TEM, while both have to be placed onto the back-focal plane of the objective lens. The topographic images obtained with DTEM for ultrathin sections of kidney cells were compared with those obtained with conventional TEM. DTEM confirmed the experimental advantage of high contrast topography by visualizing ultrastructural details inside the cells.  相似文献

Carboxysomes in rapidly frozen ice-embedded whole cells of the cyanobacterium Synechococcus sp. strain PCC 7942 were visualized by the recently developed Hilbert differential contrast transmission electron microscope. Structural details of carboxysomes were especially clearly visualized in the ruptured cells. The novel electron microscopy exhibited the paracrystalline arrays of molecules of the enzyme ribulose-1,5-bisphosphate carboxylase/oxygenase in the carboxysomes in much better contrast than conventional transmission electron microscopy with ultrathin sections of cells. The carboxysome was surrounded by a 5- to 6-nm-thick monolayer shell which consisted of orderly arrays of globular particles.  相似文献

Phase contrast transmission electron microscopy (TEM) based on thin-film phase plates has been developed and applied to biological systems. Currently, development is focused on two techniques that employ two different types of phase plates. The first technique uses a Zernike phase plate, which is made of a uniform amorphous carbon film that completely covers the aperture of an objective lens and can retard the phase of electron waves by pi/2, except at the centre where a tiny hole is drilled. The other technique uses a Hilbert phase plate, which is made of an amorphous carbon film that is twice as thick as the Zernike phase plate, covers only half of the aperture and retards the electron wave phase by pi. By combining the power of efficient phase contrast detection with the accurate preservation achieved by a cryotechnique such as vitrification, macromolecular complexes and supermolecular structures inside intact bacterial or eukaryotic cells may be visualized without staining. Phase contrast cryo-TEM has the potential to bridge the gap between cellular and molecular biology in terms of high-resolution visualization. Examples using proteins, viruses, cyanobacteria and somatic cells are provided.  相似文献

In order to acquire phase-contrast images with adequate contrast, conventional TEM requires large amount of defocus. Increasing the defocus improves the low-frequency components but attenuates the high-frequency ones. On the other hand, Zernike phase-contrast TEM (ZPC-TEM) can recover low-frequency components without losing the high-frequency ones under in-focus conditions. ZPC-TEM however, has another problem, especially in imaging of complex biological specimens such as cells and tissues; strong halos appear around specimen structures, and these halos hinder the interpretation of images. Due to this problem, the application of ZPC-TEM has been restricted to imaging of smaller particles. In order to improve the halo appearance, we fabricated a new quarter-wave thin film phase-plate with a smaller central hole and tested it on vitreous biological specimens. ZPC-TEM with the new plate could successfully visualize, in in-focus images, the intracellular fine features of cultured cells and brain tissues. This result indicates that reduction of the central hole diameter makes ZPC-TEM applicable on size scales ranging from protein particles to tissue sections. The application of ZPC-TEM to vitreous biological specimens will be a powerful method to advance the new field of imaging science for ultrastructures in close-to-physiological state.  相似文献

The Volta phase plate is a recently developed electron cryo-microscopy (cryo-EM) device that enables contrast enhancement of biological samples. Here we have evaluated the potential of combining phase-plate imaging and single particle analysis to determine the structure of a small protein–DNA complex. To test the method, we made use of a 200 kDa Nucleosome Core Particle (NCP) reconstituted with 601 DNA for which a high-resolution X-ray crystal structure is known. We find that the phase plate provides a significant contrast enhancement that permits individual NCPs and DNA to be clearly identified in amorphous ice. The refined structure from 26,060 particles has an overall resolution of 3.9 Å and the density map exhibits structural features consistent with the estimated resolution, including clear density for amino acid side chains and DNA features such as the phosphate backbone. Our results demonstrate that phase-plate cryo-EM promises to become an important method to determine novel near-atomic resolution structures of small and challenging samples, such as nucleosomes in complex with nucleosome-binding factors.  相似文献

The ability to produce several antibacterial agents greatly increases the chance of producer’s survival. In this study, red-pigmented Vibrio sp. DSM 14379 and Bacillus sp., both isolated from the same sampling volume from estuarine waters of the Northern Adriatic Sea, were grown in a co-culture. The antibacterial activity of the red pigment extract was tested on Bacillus sp. in microtiter plates. The MIC50 for Bacillus sp. was estimated to be around 10−5 mg/L. The extract prepared form the nonpigmented mutant of Vibrio sp. had no antibacterial effect. The pigment production of Vibrio sp. was studied under different physicochemical conditions. There was no pigment production at high or low temperatures, high or low salt concentrations in peptone yeast extract (PYE) medium, low glucose concentration in mineral growth medium or high glucose concentration in PYE medium. This indicates that the red pigment production is a luxurious good that Vibrio sp. makes only under favorable conditions. The Malthusian fitness of Bacillus sp. in a co-culture with Vibrio sp. under optimal environmental conditions dropped from 4.0 to −7.6, which corresponds to three orders of magnitude decrease in the number of CFU relative to the monoculture. The nonpigmented mutant of Vibrio sp. in a co-culture with Bacillus sp. had a significant antibacterial activity. This result shows that studying antibacterial properties in isolation (i.e. pigment extract only) may not reveal full antibacterial potential of the bacterial strain. The red pigment is a redundant antibacterial agent of Vibrio sp.  相似文献

Energy redistribution between growth and maintenance in salt-stressed cells is especially important for bacteria living in estuarine environments. In this study, Gram-negative bacterium Vibrio sp. DSM14379, isolated from the estuarine waters of the northern Adriatic Sea, was grown aerobically in a peptone–yeast extract medium with different salt concentrations (ranging from 0.3% to 10% (w/v) NaCl). Carbon flux through the central metabolic pathways was determined at low and high salt concentrations. At low salt concentrations, total endogenous respiration, dehydrogenase activity, and net intracellular adenosine triphosphate (ATP) concentration significantly increased, the phosphofructokinase and pyruvate kinase activity decreased, whereas glucose-6-phosphate dehydrogenase activity remained unchanged. The carrying capacity of bacterial culture decreased dramatically, indicating a severe metabolic imbalance at low salt concentrations. At high salt concentrations, carrying capacity decreased gradually. There was a large increase in glucose-6-phosphate dehydrogenase activity, which correlated with a 10-fold increase in concentration of osmoprotectant l-proline. There was no significant change of net intracellular ATP concentration, phosphofructokinase, or pyruvate kinase activity. The results indicate that Vibrio sp. DSM14379 central metabolic pathways respond to low and high salt concentrations asymmetrically; cells are better adapted to high salt concentrations. In addition, cells in the stationary phase can tolerate induced salt stress without a significant change in dehydrogenase activity or endogenous respiration for at least 1 h, but need to alter their macromolecular composition and carbon flux distribution for long-term survival.  相似文献
