The rat excretes around 2 nmol epidermal growth factor (EGF) in the urine per 24 h. The urinary EGF might be derived from plasma and/or might be synthesized in the kidneys. We have used the rat to study the renal uptake and excretion of homologous EGF from plasma. I.v. injected 125I-EGF was removed from the circulation within a few minutes. 5 min after the injection, the kidneys contained 12% of the 125I-EGF. The kidneys seemed to degrade most of the 125I-EGF which they accumulated from blood, as only 4% of the injected label was excreted as intact 125I-EGF in the urine. The amount of endogenous EGF in plasma was under the detection limit of our enzyme-linked immunosorbent assay (0.03 nmol/l) and it remained so after bilateral nephrectomy. Even if plasma EGF was 0.03 nmol/l excretion of EGF from plasma could account for less than 5% of the urinary EGF. This study shows that the kidneys are able to accumulate EGF from plasma and excrete a part of it as intact EGF in the urine. However, excretion of immunoreactive EGF from plasma can only account for a minor part of the urinary EGF.  相似文献

The sugar specificity properties of the lactose permease were investigated. Free galactose was shown to competitively inhibit the lactose permease yielding a Ki value of 7.4 mM. This value was severalfold higher than the observed Km for lactose (1.3 mM). A variety of other monosaccharides also showed significant inhibition of lactose transport. With regard to -OH groups along the galactose ring it appears that the relative importance is OH-3 greater than OH-4 greater than OH-6 greater than OH-2 greater than OH-1. In general, galactosides with alpha-linkages exhibited significantly higher affinities compared with their beta-linked counterparts. An optimal size for the aglycone portion of the galactoside was reached with aglycones containing hexose residues or a benzene ring. The preferred size of the aglycone appears to be hexose, benzene ring greater than methyl group greater than no aglycone much greater than disaccharide greater than trisaccharide. However, neither the specific structure of the aglycone nor its relative hydrophobicity appeared to be important factors in permease recognition. For example, the hydrophobic beta-nitrophenyl-galactosides had lower affinities compared with lactose (a beta-galactoside), whereas the alpha-nitrophenylgalactosides generally had higher affinities compared with melibiose (an alpha-galactoside). In addition, no consistent preference was seen when considering the location of the nitro group on the benzene ring. From this work, a model is presented which depicts the binding of galactosides to the lactose permease.  相似文献

Type IB topoisomerases cleave and rejoin DNA through a DNA-(3'-phosphotyrosyl)-enzyme intermediate. A constellation of conserved amino acids (Arg-130, Lys-167, Arg-223, and His-265 in vaccinia topoisomerase) catalyzes the attack of the tyrosine nucleophile (Tyr-274) at the scissile phosphodiester. Previous studies implicated Arg-223 and His-265 in transition state stabilization and Lys-167 in proton donation to the 5'-O of the leaving DNA strand. Here we find that Arg-130 also plays a major role in leaving group expulsion. The rate of DNA cleavage by vaccinia topoisomerase mutant R130K, which was slower than wild-type topoisomerase by a factor of 10(-4.3), was stimulated 2600-fold by a 5'-bridging phosphorothiolate at the cleavage site. The catalytic defect of the R130A mutant was also rescued by the 5'-S modification (190-fold stimulation), albeit to a lesser degree than R130K. We surmise that Arg-130 plays dual roles in transition state stabilization and general acid catalysis. Whereas the R130A mutation abolishes both functions, R130K permits the transition state stabilization function (via contact of lysine with the scissile phosphate) but not the proton transfer function. Our results show that the process of general acid catalysis is complex and suggest that Lys-167 and Arg-130 comprise a proton relay from the topoisomerase to the 5'-O of the leaving DNA strand.  相似文献

A questionnaire survey regarding endoparasite control practices in Danish horse herds was carried out in 1995. The participating veterinarians and herd owners were sampled using convenience and purposive sampling. In the analysis of risk factors for development of a high endoparasitic burden (>200 eggs per gram faeces) 903 horses were sampled and the analysis of the efficiency of a single anthelmintic treatment was based on 605 horses. The following factors had a significant effect on the endoparasitic burden: herd type, age of the horses, use of pasture rotation, anthelmintic treatment of horses visiting the herd, use of an adviser in the planning of endoparasite control and advice regarding pasture rotation. An interaction between pasture rotation and advice regarding pasture rotation was found, but due to high collinearity this was not reported. The factors influencing significantly on the reduction of the faecal egg count after a single anthelmintic treatment were the type of herd, the age of the horse, the drug used, and the anthelmintic-resistance-status of the herd. A negative effect of permanent pastures was observed. If pasture hygiene was performed on the advice of the veterinarian, the effect of a single anthelmintic treatment was less compared to a single anthelmintic treatment without any advice. An interaction between the treatment group and the resistance-status of the herd was found. Additional factors, normally accounted for, when endoparasites and anthelmintic resistance is discussed, were investigated, but not found significant in this study.  相似文献

Cells of amputated, denervated larval Ambystoma forelimbs dedifferentiate and enter the cell cycle but do not subsequently proliferate sufficiently to form a blastema. The denervated limb stump resorbs slowly until reinnervation stimulates regeneration. We used this system to investigate the fate of cells in denervated limbs which undergo early but limited cycling in response to amputation. In Experiment 1, cells were labeled with [3H]thymidine (3H-T) on Day 4 postamputation (PA)/Day 3 postdenervation (PD). Labeled cells were still present on Day 7 PA, but were less frequently observed on Day 13 PA when the limbs were reinnervated and beginning to regenerate. In Experiment 2 we denervated 1 day preamputation to obtain earlier reinnervation and prevent loss of Day 4 PA labeled cells. Cells labeled with 3H-T on Day 4 PA/Day 5 PD were present throughout the denervation period and most were still present on Day 13 PA. Little or no mitotic activity was found among the labeled cells after the initial round of cycling. The apparent cell cycle block was released upon reinnervation on Days 12 and 13 PA when cycling resumed. Labeled mitotic figures were present on Day 13 PA, and the mitotic index of the labeled population increased as a result of reinnervation. These results demonstrate that blocked cells are rescued by nerves, re-enter the cell cycle, and thus contribute to the reinnervation blastema.  相似文献
