Angiogenesis is a vital step in tissue regeneration. Hence, the current study aimed to prepare oxidized dextran (Odex)/collagen (Col)-hydrogels with laminin (LMN), as an angiogenic extracellular matrix (ECM) component, for promoting human umbilical vein endothelial cell (HUVEC) proliferation and function. Odex/Col scaffolds were constructed at various concentrations and temperatures. Using oscillatory rheometry, scanning electron microscopy (SEM), and cell viability testing, the scaffolds were characterized, and then HUVEC proliferation and function was compared with or without LMN. The gelation time could be modified by altering the Odex/Col mass ratio as well as the temperature. SEM showed that Odex/Col hydrogels had a more regular three-dimensional (3D) porous structure than the Col hydrogels. Moreover, HUVECs grew faster in the Col scaffold (12 mg/mL), whereas the Odex (30 mg/mL)/Col (6 mg/mL) scaffold exhibited the lowest apoptosis index. Furthermore, the expression level of vascular endothelial growth factor (VEGF) mRNA in the group without LMN was higher than that with LMN, and the Odex (30 mg/mL)/Col (6 mg/mL) scaffold without LMN had the highest VEGF protein secretion, allowing the cells to survive and function effectively. Odex/Col scaffolds, with or without LMN, are proposed as a tissue engineering construct to improve HUVEC survival and function for angiogenesis.  相似文献

Differential rates of NH proton-exchange reactions have been determined for the guanidinium nitrogens of l-arginine, 1?, in the pH range of 0.5 to 11.5 by natural-abundance nitrogen-15 NMR spectroscopy. Base-catalyzed NH proton exchange of the NH group is found to be two times faster than for the guanidino NH2 groups. Lack of temperature sensation of myoelectric prosthetic hand limits the daily activities of amputees.To this end,a non-invasive temperature sensation method is proposed to train amputees to sense temperature with psychophysical sensory substitution.In this study,22 healthy participants took part besides 5 amputee participants.The duration time of the study was 31 days with five test steps according to the Leitner technique.An adjustable temperature mug and a Peltier were used to change the temperature of the water/phantom digits to induce temperature to participants.Also,to isolate the surround-ings and show colors,a Virtual Reality(VR)glass was employed.The statistical results conducted are based on the response of participants with questionnaire method.Using Chi-square tests,it is concluded that participants answer the experiment significantly correctly using the Leitner technique(P value<0.05).Also,by applying the"Repeated Measures ANOVA",it is noticed that the time of numbness felt by participants had significant(P value<0.001)difference.Participants could remember lowest and highest temperatures significantly better than other temperatures(P value<0.001);furthermore,the well-trained amputee participant practically using the prosthesis with 72.58%could identify object's temperature with only once time experimenting the color temperature.  相似文献

Venous and arterial thrombosis are conditions that have a considerable burden if left untreated. The hypoxia-induced by the occluded vessel can disrupt the circulation of any organ, the cornerstone of treating thrombosis is rapid diagnosis and appropriate treatment. Diagnosis of thrombosis may be made by using laboratory tests or imaging techniques in individuals who have clinical manifestations of a thrombotic event. The use of serum micro ribonucleic acids (RNAs) has recently been applied to the diagnosis of thrombosis. These small RNA molecules are emerging as new diagnostic markers but have had very limited applications in vascular disease. Most of the articles provided various microRNAs with different levels of accuracy. However, there remains a lack of an appropriate panel of the most specific microRNA in the literature. The purpose of the present review was to summarize the existing data on the use of microRNAs as a diagnostic biomarker for venous thrombosis.  相似文献

Retinopathy of prematurity (ROP) is a multifactorial disease, that cause visual impairment in premature children. The exact pathogenesis and etiology of ROP is unknown and genetic susceptibility is considered as risk factor. Vascular endothelial growth factor (VEGF) plays a major role in retinal neovascularization and subsequently retinal detachment. VEGF polymorphism is associated with proliferative ROP in some studies. We examined the possible association of the VEGF gene polymorphisms with ROP in preterm infants in south of Iran. A total of 111 preterm infants were examined by ophthalmologist and after that were genotyped. Genotyping of the VEGF +405 (rs2010963) and VEGF +936 (rs3025039) was done by the polymerase chain reaction and restriction fragment length polymorphism methods. The frequency of VEGF alleles, genotypes and haplotype distribution were compared between groups. The patients were divided in three groups: 66 to the normal group (normal fundoscopy), and 45 to the ROP group; 30 infants were not treated with Lasertherapy (Regressive group) and 15 treated with Lasertherapy. The frequency of VEGF +405 and VEGF +936 G/C genotypes as well as allele frequencies was not different between groups. No significant difference was found between ROP with treatment and ROP without lasertherapy. Our report indicate that there is no association between the carrier states of gene polymorphisms VEGF +405, VEGF +936 and progression or spontaneous regression of ROP in preterm infants in Iranian population. However, it should be considered that angiogenesis is a complex process and genetic factors in addition to environmental factors are contributed in this pathway.  相似文献

The generalised anomeric effect (GAE) and gauche effect (GE) associated with donor–acceptor delocalisations, dipole–dipole interactions and total steric exchange energies (TSEE) on the conformational properties of 2-methoxy- (1), 2-methylthio- (2), 2-methylseleno- (3), 2-fluoro- (4), 2-chloro- (5) and 2-bromocyclohexanone (6) have been studied by means of ab initio and hybrid density functional theory methods and natural bond orbital (NBO) analysis. All methods used showed that the axial conformation stability increased from 2-methoxy- (1) to 2-methylselenocyclohexanone (3) and also from 2-fluoro- (4) to 2-bromocyclohexanone (6), which is in agreement with reported NMR data. The results obtained by complete basis set 4 (CBS-4), B3LYP/6-311+G** and HF/6-311+G** levels for compounds 1, 5 and 6 are very similar, but the CBS-4 results for compound 4 are not in agreement with the reported experimental data (vapour phase). The NBO analysis showed that the GAE increases from compounds 1 to 3 and also from compounds 4 to 6. The low axial conformer populations of compounds 1 and 4 can be reasonably explained by their small GAE. GE does not have significant impact on the conformational behaviours of compounds 16 and GAE succeeds in accounting qualitatively for the increase in the axial preferences in both series of compounds. The results showed that the calculated Δ(TSEEeq–ax) values decrease from compounds 4 to 6 which contradicts the suggested arguments in the literature about these compounds. On the other hand, the calculated differences between the dipole moment values of the axial and equatorial conformations, Δ(μeq ? μax), increase from compounds 1 to 2, but decrease from compounds 2 to 3 and also decrease from compounds 4 to 6. The calculated GAE values are more significant for the explanation of the conformational preferences of compounds 16 than the dipole–dipole repulsion effects. The correlations between the GAE, GE, dipole–dipole interactions, Wiberg Bond Index, TSEE, donor and acceptor orbital energies and occupancies, structural parameters and conformational behaviour of compounds 16 have been investigated.  相似文献

Subsoil contains more than half of soil organic carbon (SOC) globally and is conventionally assumed to be relatively unresponsive to warming compared to the topsoil. Here, we show substantial changes in carbon allocation and dynamics of the subsoil but not topsoil in the Qinghai‐Tibetan alpine grasslands over 5 years of warming. Specifically, warming enhanced the accumulation of newly synthesized (14C‐enriched) carbon in the subsoil slow‐cycling pool (silt‐clay fraction) but promoted the decomposition of plant‐derived lignin in the fast‐cycling pool (macroaggregates). These changes mirrored an accumulation of lipids and sugars at the expense of lignin in the warmed bulk subsoil, likely associated with shortened soil freezing period and a deepening root system. As warming is accompanied by deepening roots in a wide range of ecosystems, root‐driven accrual of slow‐cycling pool may represent an important and overlooked mechanism for a potential long‐term carbon sink at depth. Moreover, given the contrasting sensitivity of SOC dynamics at varied depths, warming studies focusing only on surface soils may vastly misrepresent shifts in ecosystem carbon storage under climate change.  相似文献
