Actinomycete strain SA198, isolated from a Saharan soil sample of Algeria, exhibited antimicrobial activity against Gram-positive and Gram-negative bacteria, and phytopathogenic and toxinogenic fungi. The morphological and chemotaxonomic characteristics of the strain were consistent with those of the genus Saccharothrix. Analysis of the 16S rRNA gene sequence of strain SA198 showed a similarity level ranging between 97.2 and 98.8% within Saccharothrix species, S. australiensis being the most closely related. Two new active products were isolated by reverse HPLC using a C18 column. The ultraviolet–visible (UV–VIS), infrared (IR), mass, and 1H and 14C nuclear magnetic resonance (NMR) spectra showed that these products were new bioactive compounds. The minimum inhibitory concentrations of these antibiotics showed a strong activity against fungi and moderate activities against Gram-positive and Gram-negative bacteria.  相似文献

The diversity of a population of 52 halophilic actinomycetes was evaluated by a polyphasic approach, which showed the presence of members of the Actinopolyspora, Nocardiopsis, Saccharomonospora, Streptomonospora, and Saccharopolyspora genera. One strain was considered to be a new member of the last genus, and several other strains seemed to be new species. Furthermore, 50% of strains were active against a broad range of indicators and contained genes encoding polyketide synthetases and nonribosomal peptide synthetases.  相似文献

The amino acid and humic acid requirements of Saccharothrix algeriensis NRRL B-24137 for growth and production of the dithiolopyrrolone antibiotics were studied in a semi-synthetic medium (SSM). Nature and concentration of amino acids and humic acid strongly influenced the growth and dithiolopyrrolone specific production.

The highest value of thiolutin (acetyl-pyrrothine) specific production was obtained in the presence of 1 g/l humic acid (336 mg/g DCW), and in the presence of 5 mM l-cystine (309 mg/g DCW) as compared to 19 mg/g DCW obtained with the control. Furthermore, thiolutin production was increased about six-fold, four-fold and three-fold in the presence of l-proline, l-glutamic acid and dl-histidine, respectively. In contrast, the production of thiolutin was reduced by addition of other amino acids such as l-glutamine, dl-ethionine, l-methionine and l-arginine. The highest value of isobutyryl-pyrrothine production was obtained in the presence of 2,6-diaminopimelic acid and l-lysine (7.8 and 1.0 mg/g DCW, respectively). However, the highest value of butanoyl-pyrrothine production was obtained in the presence of humic acid (6.6 mg/g DCW), followed by l-cysteine and l-proline (3.6 and 3.2 mg/g DCW, respectively). In addition, the maximum specific production of senecioyl-pyrrothine (29 mg/g DCW) and tigloyl-pyrrothine (21 mg/g DCW) was obtained in the presence of humic acid. We found that, except for isobutyryl-pyrrothine, production of all dithiolopyrrolones was favoured by addition of l-proline. The maximum specific production was obtained with l-proline at concentrations of 2.50 mM for thiolutin (133 mg/g DCW), 1.25 mM for senecioyl-pyrrothine, tigloyl-pyrrothine and butanoyl-pyrrothine production (29, 23 and 3.9 mg/g DCW, respectively). Production of all dithiolopyrrolones strongly decreased as the l-methionine or dl-ethionine concentration was increased in the culture medium.  相似文献

Three halophilic mycelium-forming actinobacteria, strains H195T, H150 and H151, were isolated from a Saharan soil sample collected from Béni-isguen in the Mzab region (Ghardaïa, South of Algeria) and subjected to a polyphasic taxonomic characterisation. These strains were observed to show an aerial mycelium differentiated into coccoid spore chains and fragmented substrate mycelium. Comparative analysis of the 16S rRNA gene sequences revealed that the highest sequence similarities were to Saccharopolyspora qijiaojingensis YIM 91168T (92.02 % to H195T). Phylogenetic analyses showed that the strains H195T, H150 and H151 represent a distinct phylogenetic lineage. The cell-wall hydrolysate was found to contain meso-diaminopimelic acid, and the diagnostic whole-cell sugars were identified as arabinose and galactose. The major cellular fatty acids were identified as iso-C15:0, iso-C16:0, iso-C17:0 and anteiso-C17:0. The diagnostic phospholipid detected was phosphatidylcholine and MK-9 (H4) was found to be the predominant menaquinone. The genomic DNA G+C content of strain H195T was 68.2 mol%. On the basis of its phenotypic features and phylogenetic position, we propose that strain H195T represents a novel genus and species, Mzabimyces algeriensis gen. nov., sp. nov., within a new family, Mzabimycetaceae fam. nov. The type strain of M. algeriensis is strain H195T (=DSM 46680T = MTCC 12101T).  相似文献

A halophilic actinomycete strain, designated H27T, was isolated from a soil sample collected from a hypersaline habitat in Djelfa Province (North-Central Algeria), and then investigated using a polyphasic taxonomic approach. The strain was observed to produce poor aerial mycelium, which formed short chains of oval to cylindrical-shaped spores at maturity, and non fragmented substrate mycelium. The optimum NaCl concentration for growth was found to be 10–15 % (w/v) and the optimum growth temperature and pH were found to be 28–37 °C and 6–7, respectively. The diagnostic diamino acid in the cell-wall peptidoglycan was identified as meso-diaminopimelic acid. The predominant menaquinones of strain H27T were identified as MK-11 (H4) and MK-10 (H6). The major fatty acids were found to be iso-C16:0, anteiso-C17:0, 10 methyl C17:0 and 10 methyl C16:0. The diagnostic phospholipids detected were phosphatidylethanolamine, diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine and phosphatidylinositol. The chemotaxonomic properties of strain H27T are consistent with those shared by members of the genus Streptomonospora. 16S rRNA gene sequence analysis indicated that strain H27T is most closely related to Streptomonospora alba DSM 44588T (98.8 %) and Streptomonospora flavalba DSM 45155T (98.7 %) whereas the DNA–DNA relatedness values between strain H27T and the two type strains were 17.1 and 57.9 %, respectively. Based on the combined genotypic and phenotypic evidence, it is proposed that strain H27T should be classified as representative of a novel species, for which the name Streptomonospora algeriensis sp. nov. is proposed. Rhizoctonia solani is one of the most problematic soil-borne pathogenic fungi for several crop cultures worldwide. This study highlights the effectiveness of high-antagonistic Streptomyces rochei strain PTL2, isolated from root tissues of Panicum turgidum, in controlling the R. solani damping-off and growth promotion of tomato (cv. Marmande) seedlings. The isolate PTL2 was characterised for in vitro biocontrol and plant growth-promoting traits. It exhibited remarkable positive results in all trials, including production of hydrogen cyanide, siderophores, 1-aminocyclopropane-1-carboxylate deaminase and phytohormones, chitinolytic activity and inorganic phosphate solubilisation. PTL2 spores were formulated as wettable talcum powder, sodium alginate pellets and sodium alginate-clay pellets. Their abilities in the biocontrol of R. solani and plant growth promotion were investigated in autoclaved and non-autoclaved soils. Talcum powder and sodium alginate pellets significantly reduced the damping-off severity index compared to a positive control. The talcum powder exhibited the highest protective activity, reducing the disease incidence from 89.3% to 14.1%, whereas chemical seed treatment with Thiram® provided a disease incidence of 16.7%. Furthermore, the talc-based powder formulation resulted in greatest increases in the root length, shoot length and dry weight of seedlings. The interesting biocontrol potential and growth enhancement of tomato seedlings open up promising perspectives for the possible application of talcum powder formulation based on PTL2 spores in crop improvement.  相似文献

The taxonomic position of a new Saccharothrix strain, designated MB46T, isolated from a Saharan soil sample collected in Mzab region (Ghardaïa province, South Algeria) was established following a polyphasic approach. The novel microorganism has morphological and chemical characteristics typical of the members of the genus Saccharothrix and formed a phyletic line at the periphery of the Saccharothrix espanaensis subcluster in the 16S rRNA gene dendrograms. Results of the 16S rRNA gene sequence comparisons revealed that strain MB46T shares high degrees of similarity with S. espanaensis DSM 44229T (99.2%), Saccharothrix variisporea DSM 43911T (98.7%) and Saccharothrix texasensis NRRL B-16134T (98.6%). However, the new strain exhibited only 12.5–17.5% DNA relatedness to the neighbouring Saccharothrix spp. On the basis of phenotypic characteristics, 16S rRNA gene sequence comparisons and DNA-DNA hybridizations, strain MB46T is concluded to represent a novel species of the genus Saccharothrix, for which the name Saccharothrix ghardaiensis sp. nov. (type strain MB46T = DSM 46886T = CECT 9046T) is proposed.  相似文献
