Hairless (H) is the major antagonist within the Notch signalling pathway of Drosophila melanogaster. By binding to Suppressor of Hairless [Su(H)] and two co-repressors, H induces silencing of Notch target genes in the absence of Notch signals. We have applied genomic engineering to create several new H alleles. To this end the endogenous H locus was replaced with an attP site by homologous recombination, serving as a landing platform for subsequent site directed integration of different H constructs. This way we generated a complete H knock out allele H
attP, reintroduced a wild type H genomic and a cDNA-construct (H
gwt, H
cwt) as well as two constructs encoding H proteins defective of Su(H) binding (H
LD, H
iD). Phenotypes regarding viability, bristle and wing development were recorded, and the expression of Notch target genes wingless and cut was analysed in mutant wing discs or in mutant cell clones. Moreover, genetic interactions with Notch (N
5419) and Delta (Dl
B2) mutants were addressed. Overall, phenotypes were largely as expected: both H
LD and H
iD were similar to the H
attP null allele, indicating that most of H activity requires the binding of Su(H). Both rescue constructs H
gwt and H
cwt were homozygous viable without phenotype. Unexpectedly, the hemizygous condition uncovered that they were not identical to the wild type allele: notably H
cwt showed a markedly reduced activity, suggesting the presence of as yet unidentified regulatory or stabilizing elements in untranslated regions of the H gene. Interestingly, H

Juvenile material with the main focus on the upper jaw of the fossil predator Hyaenodon was evaluated to study the tooth eruption sequence and to examine the ontogeny of its dentition in detail. The comparison in size of milk to permanent teeth indicates a growth rate of 12–16 % in Hyaenodon. The thin section of a deciduous canine of a North American taxon shows four dental rings. Based on the knowledge of recent carnivores, this implies an age of 3–4 years in the last stage of tooth eruption and thus a long juvenile phase. The mandibles ascertained the most recent established tooth eruption sequence for North American and European species. For the first time ever, juvenile material from Asia is documented and interpreted. This study likewise shows a difference in the sequence of the upper jaw: the first upper premolar erupts before the first upper molar in North American species, whereas the European taxa show an earlier eruption of the first upper molar. Inhibition by ouabain of rheogenic Na+ transport across the basolateral membranes of frog skin is found to be manifest within 3–4 min. This rate of pump inhibition is not different from the rate of diffusion through extracellular tissue layers between the serosal bath and the actual site of action, i.e., the epithelial cell layers. We have defined the predominant site of p-nitrophenyl acetate reaction with hemoglobin. The site is involved with, at least, two modes of action: the imidazole catalysis of His beta 2 and the irreversible covalent acetylation of Lys beta 82. The effect of competitive inhibition of the reaction by 2,3-diphosphoglyceric acid, the dependence of the reaction rate on the protein conformation, hemoglobin mutants, and the diethylpyrocarbonate are consistent with the assignment of the active site.
