Sunflower honey (SH) is bright yellow, fragrant, pollen-flavoured, slightly herbaceous and has a unique taste. The present research aims to examine the enzyme inhibitory, antioxidant, anti-inflammatory, antimicrobial and anti-quorum sensing activities and phenolic compositions of 30 sunflower honeys (SHs) produced from several regions of Turkey with chemometric study. SAH from Samsun exhibited the best antioxidant activity in β-carotene linoleic acid (IC50: 7.33±0.17 mg/mL) and CUPRAC (A0.50: 4.94±0.13 mg/mL) assays, anti-urease activity (60.63±0.87 %) and anti-inflammatory activity against COX-1 (73.94±1.08 %) and COX-2 (44.96±0.85 %). SHs exhibited mild antimicrobial activity against the test microorganisms while they showed high quorum sensing inhibition zones measured in the range of 42–52 mm against the CV026 strain. The phenolic composition was determined by high performance liquid chromatography with diode array detection (HPLC-DAD) system and levulinic, gallic, p-hydroxybenzoic, vanillic and p-coumaric acids were identified in all studied SHs. The classification of SHs was performed the using PCA and HCA. This study revealed that phenolic compounds and biological properties are effective in classification of SHs according to their geographical origin. The results suggest that studied SHs could be valued as potential agents with versatile bioactivities in oxidative stress-related disease, microbial infections, inflammation, melanoma, and peptic ulcer.  相似文献

Renal cell carcinomas arise from the nephron but are heterogeneous in disease biology, clinical behavior, prognosis, and response to systemic therapy. Development of patient-specific in vitro models that efficiently and faithfully reproduce the in vivo phenotype may provide a means to develop personalized therapies for this diverse carcinoma. Studies to maintain and model tumor phenotypes in vitro were conducted with emerging three-dimensional culture techniques and natural scaffolding materials. Human renal cell carcinomas were individually characterized by histology, immunohistochemistry, and quantitative PCR to establish the characteristics of each tumor. Isolated cells were cultured on renal extracellular matrix and compared to a novel polysaccharide scaffold to assess cell-scaffold interactions, development of organoids, and maintenance of gene expression signatures over time in culture. Renal cell carcinomas cultured on renal extracellular matrix repopulated tubules or vessel lumens in renal pyramids and medullary rays, but cells were not observed in glomeruli or outer cortical regions of the scaffold. In the polysaccharide scaffold, renal cell carcinomas formed aggregates that were loosely attached to the scaffold or free-floating within the matrix. Molecular analysis of cell-scaffold constructs including immunohistochemistry and quantitative PCR demonstrated that individual tumor phenotypes could be sustained for up to 21 days in culture on both scaffolds, and in comparison to outcomes in two-dimensional monolayer cultures. The use of three-dimensional scaffolds to engineer a personalized in vitro renal cell carcinoma model provides opportunities to advance understanding of this disease.  相似文献

Cisplatin, one of the most active cytotoxic agents against cancer, has several toxicities. Hepatotoxicity is one of them occurred during high doses treatment. The aim of this study was to determine the effects of erdosteine against cisplatin-induced liver injury through tissue oxidant/antioxidant parameters and light microscopic evaluation. The rats were randomly divided into three groups: control (n=5), cisplatin (10 mg/kg, n=6) and cisplatin+erdosteine (50 mg/kg/day oral erdosteine, n=8) groups. The rats were sacrificed at the 5th day of cisplatin treatment. The liver tissues were examined with light microscopy and oxidant/antioxidant biochemical parameters. The malondialdehyde (MDA) and nitric oxide (NO) levels were increased in the cisplatin group in comparison with the control and cisplatin+erdosteine groups (p<0.05). There was no significant difference in MDA and NO levels between control and cisplatin+erdosteine groups. The activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) were higher in cisplatin+erdosteine group than cisplatin group (p<0.05). However, the CAT and GSH-Px activities were significantly lower in cisplatin group than in control group (p<0.05). The light microscopic examination revealed that cytoplasmic changes especially around cells of central vein were observed in cisplatin group. Hepatocellular vacuolization was seen in these cells. In the cisplatin plus erdosteine group, a decrease in cytoplasmic changes with the hepatocytes and sinusoidal dilatations around cells of central vein were noticed in as compared to cisplatin group. In the light of microscopic and biochemical results, it was concluded that cisplatin-induced liver damage in high dose and erdosteine prevented this toxic side effect by the way of its antioxidant and radical scavenging effects. A novel gene sequence, with two exons and one intron, encoding a metallothionein (MT) has been identified in durum wheat Triticum durum cv. Balcali85 genomic DNA. Multiple alignment analyses on the cDNA and the translated protein sequences showed that T. durum MT (dMT) can be classified as a type 1 MT. dMT has three Cys-X-Cys motifs in each of the N- and C-terminal domains and a 42-residue-long hinge region devoid of cysteines. dMT was overexpressed in Escherichia coli as a fusion protein (GSTdMT), and bacteria expressing the fusion protein showed increased tolerance to cadmium in the growth medium compared with controls. Purified GSTdMT was characterized by SDS- and native-PAGE, size exclusion chromatography, and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. It was shown that the recombinant protein binds 4 +/- 1 mol of cadmium/mol of protein and has a high tendency to form stable oligomeric structures. The structure of GSTdMT and dMT was investigated by synchrotron x-ray solution scattering and computational methods. X-ray scattering measurements indicated a strong tendency for GSTdMT to form dimers and trimers in solution and yielded structural models that were compatible with a stable dimeric form in which dMT had an extended conformation. Results of homology modeling and ab initio solution scattering approaches produced an elongated dMT structure with a long central hinge region. The predicted model and those obtained from x-ray scattering are in agreement and suggest that dMT may be involved in functions other than metal detoxification.  相似文献

Chrysin, apigenin, flavonoids, flavanones, naringenin, ethyl oleate, 3-4-dimethoxy-cinnamic acid and 9-octadecenoic acid were the predominant components of propolis samples collected from different regions of Turkey. The extracts of P3 from Denizli-Ba?karci, P5 from Denizli and P7 from Tekirda? had effective antibacterial activities on Gram-negatives. Chrysin, which has antibacterial activity, was found to be high concentration. The extracts of P3, P2B from Aydin and P6 from Konya had much more effective antibacterial activities on Gram-positives. The total antioxidant activity increased with the increasing amount of extracts added to linoleic acid emulsion. All doses of propolis ethanol extract displayed antioxidant activity.  相似文献

Antioxidant capacity and antimicrobial activities ofLepista nuda (Bull.) Cooke extracts obtained with ethanol were investigated. Four complementary test systems, namely DPPH free radical scavenging, β-carotene/linoleic acid systems, total phenolic compounds and total flavonoid concentration, have been used. Linoleic, acid inhibition values ofL. nuda ethanolic extract, BHA and α-to copherol standards were found to be 84.3% 98.9% and 99.2% respectively in the concentration of 160μg/ml. Total flavonoid amount was 8.21 ± 0.56 μg mg?1 quercetin equivalent while the phenolic compound amount was 48.01 ± 0.29 μg mg?1 pyrocatechol equivalent in the extract. The antimicrobial activity ofL. nuda extract was testedin vitro by using the agar-well diffusion method. TheL. nuda extract showed antibacterial activity againstMicrococcus flavus, Micrococcus luteus, Bacillus cereus, Yersinia enterocolitica, Staphylococcus aureus, Salmonella enteritidis andEscherichia coli. TheL. nuda extract did not exhibit antican didal activity againstCandida albicans. The extracts could be suitable as antimicrobial and antioxidativeagents in the food industry.  相似文献

Objective: The aim of this study was to investigate effect of loss weight on P wave dispersion in obese subjects. Research Methods and Procedures: After a 12‐week weight loss program (diet and medical therapy), a total of 30 (24 women and six men) obese subjects who had lost at least 10% of their original weight were included in the present study. All subjects underwent a routine standard 12‐lead surface electrocardiogram. Electrocardiograms were transferred to a personal computer by a scanner and then magnified 400 times by Adobe Photoshop software (Adobe Systems, Mountain View, CA). P wave dispersion, which is also defined as the difference between the maximum P wave duration and the minimum P wave duration, was also calculated. Results: After a 12‐week weight loss program, BMI (p < 0.001), maximum P wave duration (p < 0.001), and P wave dispersion (p < 0.001) significantly decreased. The mean percentage of weight loss was 13% (10% to 20.3%). The decrease in the level of P wave dispersion (21 ± 10 and 7 ± 12 ms, p < 0.002) was more prominent in Group II (≥12% loss of their original weight) than Group I (<12% loss of their original weight) after the weight loss program. A statistically significant correlation between decrease in the level of P wave dispersion and percentage of weight loss was found (r = 0.624, p < 0.001). Discussion: Substantial weight loss in obese subjects is associated with a decrease of P wave duration and dispersion. Therefore, these observations suggest that substantial weight loss is associated with improvement in atrial repolarization abnormalities in obese subjects.  相似文献

IL-10 can be considered an important agent in the resolution of inflammation. Originally named "cytokine synthesis inhibitory factor" for its ability to inhibit IFN-gamma and IL-2 production in Th2 cells, it is secreted by monocytes, macrophages, mast cells, T and B lymphocytes, and dendritic cells (DCs). IL-10 production and release by monocytic cells in response to allergic challenge is upregulated by TNF-alpha, and by negative feedback regulation of itself. However, it is also secreted by T regulatory cells (Tregs), under the control of IL-2. Importantly in the context of asthma, IL-10 inhibits eosinophilia, by suppression of IL-5 and GM-CSF, by direct effects on eosinophil apoptosis, and effects on cell proliferation through down-regulation of IL-1. A number of its cytokine suppressive characteristics are now thought to occur through its upregulation of suppressor of cytokine signaling (SOCS)-3. IL-10 is also a suppressor of nitric oxide (NO) production, which may have ramifications for its role in airway inflammatory diseases. Initial clinical trials have demonstrated relative safety and few clinically adverse events at doses of recombinant human IL-10 below 50 microg/kg, with mixed success in treatment of patients with inflammatory bowel disease and psoriasis. However, both steroid therapy and allergen specific immunotherapy are known to elevate endogenous IL-10 levels, which may account for their efficacy, suggesting that further study of IL-10 as a target for treatment of airway inflammatory diseases such as asthma and COPD is warranted.  相似文献

Abstract. Plant traits which may give an indication of a plant's strategy for nutrient acquisition and regeneration are known for numerous grassland species. This study aimed to establish whether there is any relationship between two plant traits: specific leaf area (SLA) and number of reproductive tillers, and sward structural characteristics which influence herbage intake by grazers (bulk density and digestibility, leaf:stem ratio). Comparison is made for nutrient‐rich (Dactylis glomerata) and nutrient‐poor (Festuca rubra) grass species. We hypothesized that these traits are responsive to environmental gradients and also act on the processes of the ecosystem. Both grasses were compared with two P‐fertilizer rates in two localities (200 and 1300 m a.s.l.) which differed in their temperature:radiation ratios. For the vegetative phase SLA was well correlated with sward characteristics: D. glomerata, which has the higher SLA, has the lower bulk density and higher digestibility. The values of SLA and vegetation bulk density varied according to growing conditions (P‐rate and temperature:radiation ratio), but the ranking of the species remained the same because the phenotypic plasticity that exists for plant traits was also observed for sward structure and composition. That suggested the possibility of grouping natural grassland species for their relevant characteristics for grazers according to SLA values. Over the reproductive phase, the proportion of stems was well correlated to the percentage of reproductive tillers. However, the percentage of reproductive tillers was a very plastic trait for both species, depending on the growing conditions, and resulting in a density‐dependent effect, particularly for F. rubra. The species studied were too plastic and too similar in their regenerative strategy so that there is no unique relationship between percentage of reproductive tillers and stem proportion, regardless of the species and the growing conditions. The number of reproductive tillers is not a suitable plant trait which could be used to rank species for leaf and stem proportions in the sward.  相似文献

Background and Aims: Fodder provision in species-rich grasslands, i.e. herbage growth,proportion of leaf, and leaf and stem digestibility, is difficultto predict for short periods of time, such as between two defoliationsor less. The value of two methods based on plant traits forevaluating these agronomic properties was examined. Methods: One method is based on plant trait measurements on the plantcommunity (leaf dry matter content, plant height, floweringdate); the other is on vegetation composition expressed as plantfunctional types (acquisitive versus conservative PFTs) establishedby measuring leaf dry matter content on pure grass stands. Theexperiment consisted of 18 fields with three different defoliationregimes (combinations of cutting and grazing) and two levelsof fertilization. To establish a growth curve over the firstgrowth cycle, herbage was sampled about 10 times in spring. Key Results: Coefficients of correlation between agronomic properties ofthe vegetation and its functional composition were higher whenthe latter was assessed through PFT and an indicator of theplant nutrient status (Ni) instead of measured plant traits.The date at which the ceiling yield occurred for the standingherbage mass or only the leaf component, which varied by upto 500 degree-days between treatments, and the leaf proportion,depended entirely on the PFT, and largely so for the leaf digestibility.The standing herbage mass at the time of ceiling yield dependedonly on Ni, or mainly so in the case of the daily herbage growthrate. Similar plant digestibility between plant communitieswas found at flowering time, although there were big differencesin PFT composition. The shape of the growth curve was flatterwhen there was great functional diversity in the plant community. Conclusions: The PFT composition and the Ni were more reliable than the plantfunctional traits measured in the field for evaluating herbagegrowth pattern and digestibility in spring.  相似文献
