This study was aimed to isolate Hg-resistant bacteria from contaminated sediments of the Orbetello Lagoon in Italy and to assess their possible use as biofilms in bioremediation processes. Enrichment cultures prepared from contaminated sediments in the presence of 0.05 mM of mercury and under aerobic conditions allowed the isolation of five heterotrophic bacterial strains. 16S rDNA gene sequencing assigned the isolated strains to the genera Pseudomonas and Psychrobacter. For the first time mercury-resistant bacterial strains belonging to the genus Psychrobacter were evidenced. Minimum inhibitory concentrations in the presence of HgCl2 and of CH3HgCl showed high levels of resistance. EC50 values for the isolated bacterial strains in the presence of HgCl2 and of CH3HgCl confirmed the adaptation to the metal. Hg-resistant strains ORHg1, ORHg4 and ORHg5 showed the capacity to volatilize inorganic and organic mercury to elemental mercury, and formed biofilms on pumice particles, and were shown to play a role in the removal of mercury from sediment leachates. This study reports isolation and characterization of new Hg-resistant bacterial strains and provides novel insight into their possible use in bioremediation processes of mercury polluted sediments.  相似文献

The production of a substrate containing destoned olive mill solid waste for the cultivation of Agaricus bisporus (Lange) Imbach on an industrial scale was studied. A standard mushroom compost (C) mainly made from straw and poultry manure was compared with the experimental compost (EC) containing the same ingredients as (C) but with added olive mill solid waste (10.6% w/w). Microbial indicators such as counts of heterotrophic microbes and actinomycetes were higher in EC than in C. In addition, compost selectivity as indicated by higher mushroom yield and biological efficiency of EC was higher than that of C. Market quality of the mushrooms produced in both C and EC were comparable. These findings support our work that olive mill solid waste can be used safely in thermal composting process to produce a selective substrate for industrial-scale cultivation of A. bisporus. This study also demonstrates an environmentally sustainable system to manage solid waste from olive oil extraction processes thus overcoming environmental pollution brought about by irrational disposal of the waste on farm lands.  相似文献

The feasibility of using olive mill waste (OMW) as an ingredient in the substrate used for cultivation of Agaricus bisporus (Lange) Sing. was studied in a large-scale cultivation trial, concerning 2500 m2 of mushroom growing area, at a specialized mushroom farm. Standard commercial cultivation technique involving compost preparation, spawning, casing and harvesting was used. The performance indicators such as mushroom yield, biological efficiency, market quality as well as horticultural value of the spent compost showed that the compost prepared with OMW was superior to the control compost in all the categories. The OMW-amended substrate supported higher populations of beneficial microorganisms especially, actinomycetes which enabled the breakdown of the compost ingredients. It is suggested that OMW is a suitable ingredient for the preparation of mushroom substrate. We have demonstrated that conversion of OMW (a liability) into value-added mushroom substrate (an asset) is an effective waste management tool in oleaculture.  相似文献

Despite the importance of the adrenergic activity and of the metabolism of magnesium in some important cardiovascular pathologies, very little is known about how intracellular ionized magnesium (Mg i 2+ ) is regulated by catecholamines. We made an in-vitro study of the variations in the concentration of ionized magnesium in human lymphocytes using the fluorescent probe furaptra in response to different catecholamines. We also made an ex-vivo study of the changes in intracellular ionized magnesium in lymphocytes in 20 subjects with essential arterial hypertension, 10 treated with 120 mg/d of propranolol and 10 with placebo. Norepinephrine and isoproterenol significantly decrease Mg i 2+ and this effect is blocked by β-blockers but not by α-blockers. The EC 50 of the effect of norepinephrine is within the range of concentrations physiologically present in plasma. The substitution of extracellular sodium with choline blocks the decrease in intracellular ionized magnesium induced by norepinephrine, which leads us to suppose that the magnesium-reducing effect of catecholamines is a result of the activation of a Na+-Mg2+ exchanger. We were not able to demonstrate any change in intracellular ionized magnesium after 1 and 17 days of active treatment in essential hypertensives. The impossibility of demonstrating ex vivo the mechanism of catecholamine-mediated regulation that is evident in vitro is perhaps due to our experimental conditions or to substances which in vivo inhibit the action of the catecholamines on magnesium, such as insulin and/or glucose.  相似文献

Reduction of vanadate to vanadyl by a strain of Saccharomyces cerevisiae   总被引:1,自引:0,他引:1  
Three strains of Saccharomyces cerevisiae, SC-1, DBVPG 6173 and DBVPG 6037, were studied for vanadate resistance in complex Sabouraud medium since they did not thrive in different minimal media (yeast nitrogen base with and without amino acids). The strain SC-1 was resistant up to 16 mm of vanadate, whereas the strains DBVPG 6173 and DBVPG 6037 were inhibited by 8 mm and 4 mm vanadate, respectively. The vanadate resistance in strain SC-1 was constitutive and due to the reduction of this oxyanion to vanadyl, which was detected by EPR spectroscopy and visible spectroscopy. The transformation of vanadate to vanadyl took place during the exponential growth phase; 10 mm of vanadate was reduced to vanadyl outside the cells since the oxyanion was not detected in the cell biomass and only a negligible concentration of vanadyl (25 nmoles mg cells dry weight) was found in the biomass. The other two vanadate-sensitive yeast strains only accumulated vanadate and did not reduce the oxyanion to vanadyl.  相似文献

Mercury chloride toxicity was investigated in two strains of Chlorella and in a strain of Scenedesmus isolated from polluted areas in Tuscany (Italy). No Hg resistance was found in the autotrophic microorganisms isolated, but Scenedesmus sp. strain AR-2489, isolated from the Arno river, was able to grow at concentrations of up to 5 mg ml1 of Hg. This concentration was twice that which inhibited growth of the two Chlorella strains and Scenedesmus acutus 8M, the reference strain from a culture collection. Photosynthesizing cells of Scenedesmus sp. AR-2489 showed reduced Hg uptake, with the highest percentage of Hg removal from the medium. Loss of Hg was not due to Hg(0) volatilization, as shown by a comparison test with the broad-spectrum Hg-resistant Pseudomonas putida FB1. The metabolic differences between Scenedesmus sp. strain AR-2489 and Siacatus strain 8M were: (1) higher growth rate (doubling time of 6.0h versus 10.6 h); (2) higher O2 production rate (maximum 2 mmol h mg dry weight); and (3) higher intracellular pH during growth. The latter was imaged with a green fluorescence molecular probe (BCEFC–AM) and observed by scanning confocal laser microscopy (SCLM). The distribution of red-autofluorescence chlorophyll-a showed that strain AR-2489 had a rougher and hence more extended specific chloroplast surface than strain 8M. Hg tolerance in strain AR-2489 was related to the rapid increase in dissolved O in the medium and in intracellular pH; this caused a loss of soluble mercury transformed to insoluble mercury hydroxide, which is thermodynamically more stable at alkaline pH in highly oxygenated systems.  相似文献

Bacillus sp. ORAs2 and Pseudomonas sp. ORAs5, two arsenic-resistant bacterial strains previously isolated from sediments of the Orbetello Lagoon, Italy, were tested for their adaptation to mixed contaminants on the level of membrane fatty acid composition. The two bacterial strains were characterized by high levels of arsenic resistance, and Pseudomonas sp. ORAs5 was also shown to be solvent-tolerant. The bacterial strains were exposed to mixtures of two toxic compounds: arsenic at fixed concentrations and toluene in variable amounts or, alternatively, toluene at constant values along with arsenic added at variable concentrations. Both strains react to the contaminants by changing the composition of their membrane fatty acids. Bacillus sp. strain ORAs2 showed a correlation between growth rate decreases and fatty acids degree of saturation increases in both cases, although pointedly in the presence of 1, 2, and 3 mM of toluene and different additions of arsenic, counteracting membranes fluidity induced by toxic compounds. In Pseudomonas sp. ORAs5, adaptive changes in membrane composition was observed both in terms of increases in the degree of saturation and in the trans/cis ratio of unsaturated fatty acids in the presence of varying toluene and constant arsenic concentrations, whereas only minor changes occurred with increasing arsenic and constant toluene concentrations. Thus, on the level of membrane composition, Bacillus sp. ORAs2 showed a higher potential for adaptation to the presence of mixed pollutants, suggesting its probable suitability for bioremediation purposes.  相似文献

It is well known that in systems of surname transmission through the paternal line, surnames simulate neutral gene alleles belonging to the Y chromosome. This property of surnames was used to analyze the genetic structure of Albanian populations. Two large samples of surnames belonging to two different periods of time were analyzed. The analysis of indicators of population structure showed that geographical distance has an important effect on surname distribution. It seems that isolation by distance and genetic drift have been still important factors in the determination of the genetic structure of the Albanian population.  相似文献

Total cholesterol, triglycerides and HDL-, HDL2-, HDL3-cholesterol were evaluated in 81 subjects, 49 of them practised moderate physical activity, 32 were controls. Total cholesterol and triglycerides were evaluated enzymatically. The Abell method was applied to analyse HDL-cholesterol and the HDL3 subclass. A significant difference was observed between the plasmatic triglycerides of the active group and the control, indicating that even moderate physical activity can be associated with lower concentrations of triglycerides. The correlation coefficients calculated for the HDL-cholesterol and the HDL2-cholesterol subclass, respectively, with some anthropometric and physiological variables were, in most of the cases, not significant. The highest number of significant correlations was found in the HDL3-cholesterol subclass, but in that of the control group only.  相似文献
