Rust in bean (Phaseolus vulgaris L.), caused byUromyces appendiculatus (Pers.) Unger var.appendiculatus [ =U. phaseoli (Reben) Wint.], is a major disease problem and production constraint in many parts of the world. The predominant form of genetic control of the pathogen is a series of major genes which necessitate the development of efficient selection strategies. Our objective was focused on the identification of RAPD (random amplified polymorphic DNA) markers linked to a major bean rust resistance gene block enabling marker-based selection and facilitating resistance gene pyramiding into susceptible bean germplasm. Using pooled DNA samples of genotyped individuals from two segregating populations, we identified two RAPD markers linked to the gene block of interest. One such RAPD, OF10970 (generated by a 5-GGAAGCTTGG-3 decamer), was found to be closely linked (2.15±1.50 centi Morgans) in coupling with the resistance gene block. The other identified RAPD, OI19460 (generated by a 5-AATGCGGGAG-3 decamer), was shown to be more tightly linked (also in coupling) than OF10970 as no recombinants were detected among 97 BC6F2 segregating individuals in the mapping population. Analysis of a collection of resistant and susceptible cultivars and experimental lines, of both Mesoamerican and Andean origin, revealed that: (1) recombination between OF10970 and the gene block has occurred as evidenced by the presence of the DNA fragment in several susceptible genotypes, (2) recombination between OI19460 and the gene block has also occurred indicating that the marker is not located within the gene block itself, and (3) marker-facilitated selection using these RAPD markers, and another previously identified, will enable gene pyramiding in Andean germplasm and certain Mesoamerican bean races in which the resistance gene block does not traditionally exist. Observations of variable recombination among Mesoamerican bean races suggested suppression of recombination between introgressed segments and divergent recurrent backgrounds.Research supported by the Michigan Agricultural Research Station and the USDA-ARS. The purpose of this case study was to optimise design, operation and maintenance guidelines, and to assess the water treatment potential of a storm water pond system after 15 months of operation. The system was based on a combined silt trap, attenuation pond and vegetated infiltration basin. This combination was used as the basis for construction of a roof water runoff system from a single domestic property. United Kingdom Building Research Establishment and Construction Industry Research and Information Association, and German Association for Water, Wastewater and Waste design guidelines were tested. These design guidelines failed because they did not consider local conditions. The infiltration function for the infiltration basin was logarithmic. Algal control techniques were successfully applied, and treatment of rainwater runoff from roofs was found to be largely unnecessary for recycling (e.g., watering plants). However, seasonal and diurnal variations of biochemical oxygen demand, dissolved oxygen and pH were recorded.  相似文献

The aim of this study was to investigate the treatment efficiency of passive vertical-flow wetland filters containing different macrophytes (Phragmites and/or Typha) and granular media with different adsorption capacities. Gravel, sand, granular activated carbon, charcoal and Filtralite (light expanded clay) were used as filter media. Different concentrations of lead and copper sulfate were added to polluted urban stream inflow water to simulate pretreated mine wastewater. The relationships between growth media, microbial and plant communities as well as the reduction of predominantly lead, copper and five-day biochemical oxygen demand (BOD5) were investigated. An analysis of variance showed that concentration reductions (mg l(-1)) of lead, copper and BOD5 were significantly similar for the six experimental wetlands. Microbial diversity was low due to metal pollution and similar for all filters. There appears to be no additional benefit in using adsorption media and macrophytes to enhance biomass performance during the first 10 months of operation.  相似文献

Resistance (R) genes containing nucleotide-binding site (NBS)-leucine rich repeats (LRR) are the most prevalent types of R gene in plants. The objective of this study was to develop PCR-based R-gene analog polymorphism (RGAP) markers for common bean (Phaseolus vulgaris L). Twenty degenerate primers were designed from the conserved kinase-1a (GVGKTT) and hydrophobic domains (GLPLAL) of known NBS-LRR type R-genes and from EST databases. Sixty-six of the 100 primer combinations tested yielded polymorphism. Thirty-two RGAP markers were mapped in the BAT 93/Jalo EEP558 core mapping population for common bean. The markers mapped to 10 of 11 linkage groups with a strong tendency for clustering. In addition, the RGAP markers co-located, on six linkage groups, with 15 resistance gene analogs (RGAs) that were previously mapped in other populations of common bean. The distance between the priming sites in NBS-LRR type R-genes is around 500 bp. Of the 32 RGAP markers, 19 had sizes larger and 13 less than 500 bp. RGAP markers mapped close to known R-genes on B11, and to QTLs for resistance on B1, B2, B6, B7, B8, B10, and B11. RGAP appears to provide a useful marker technique for tagging and mapping R-genes in segregating common bean populations, discovery of candidate genes underlying resistance QTL, and future cloning of R-genes in common bean.  相似文献

This paper comprises the scientific justification for the Farm Constructed Wetland (FCW) Design Manual for Northern Ireland and Scotland. Moreover, this document addresses an international audience interested in applying wetland systems in the wider agricultural context. Farm constructed wetlands combine farm wastewater (predominantly farmyard runoff) treatment with landscape and biodiversity enhancements, and are a specific application and class of integrated constructed wetlands (ICW), which have wider applications in the treatment of other wastewater types such as domestic sewage. The aim of this review paper is to propose guidelines highlighting the rationale for FCW, including key water quality management and regulatory issues, important physical and biochemical wetland treatment processes, assessment techniques for characterizing potential FCW sites and discharge options to water bodies. The paper discusses universal design, construction, planting, maintenance and operation issues relevant specifically for FCW in a temperate climate, but highlights also catchment-specific requirements to protect the environment.  相似文献

A self-organizing map (SOM) model was applied as a prediction tool for the performance of an integrated constructed wetland (ICW) agroecosystem treating agricultural runoff to protect receiving watercourses. By utilizing the SOM model, the time-consuming to measure expensive biochemical oxygen demand outflow concentrations were predicted well by other inexpensive variables, which were quicker and easier to measure. Correct predictions for the outflow biochemical oxygen demand concentrations were between 89% and 100%. This novel approach allows for the real time control of the outflow water quality of the ICW and potentially also of other treatment system applications. Moreover, the missing values and outliers from the large but incomplete ICW data set were replaced accurately by most likely values determined by the SOM model. This was important because the proportions of unusable entries for chemical oxygen demand, suspended solids and biochemical oxygen demand were very high: 41%, 54%, and 61%, respectively.  相似文献

The development and use of RAPD markers for applications in crop improvement has recently generated considerable interest within the plant breeding community. One potential application of RAPDs is their use for tagging simply-inherited (monogenic) pest-resistance genes and enabling more efficient identification and selection of genotypes carrying specific combinations of resistance genes. In this report, we propose and describe the use of heterogeneous inbred populations as sources of near-isogenic lines (NILs) for targeting RAPD markers linked to major pest resistance genes. The development of these NILs for RAPD marker analyses involved a sequence of line and mass selection during successive generations of inbreeding. DNA bulks derived from the NILs were used to identify a RAPD marker (designated OK14620, generated by 5-CCCGCTACAC-3 decamer) that was tightly linked (2.23±1.33 centiMorgans) to an important rust [Uromyces appendiculatus (Pers.) Unger var. appendiculatus] resistance gene (Ur-3) in common bean (Phaseolus vulgaris L.). The efficiency of this approach was demonstrated by a low rate of false-positives identified, the tightness of the linkage identified, and the ability to detect polymorphism between genomic regions that are representative of the same gene pool of common bean. This method of deriving NILs should find application by researchers interested in utilizing marker-assisted selection for one or more major pest resistance genes. Cell survival in complex, vascularized tissues, has been implicated as a major bottleneck in advancement of therapies based on cardiac tissue engineering. This limitation motivates the search for small, inexpensive molecules that would simultaneously be cardio-protective and vasculogenic. Here, we present peptide sequence QHREDGS, based upon the fibrinogen-like domain of angiopoietin-1, as a prime candidate molecule. We demonstrated previously that QHREDGS improved cardiomyocyte metabolism and mitigated serum starved apoptosis. In this paper we further demonstrate the potency of QHREDGS in its ability to enhance endothelial cell survival, metabolism and tube formation. When endothelial cells were exposed to the soluble form of QHREDGS, improvements in endothelial cell barrier functionality, nitric oxide production and cell metabolism (ATP levels) in serum starved conditions were found. The functionality of the peptide was then examined when conjugated to collagen-chitosan hydrogel, a potential carrier for in vivo application. The presence of the peptide in the hydrogel mitigated paclitaxel induced apoptosis of endothelial cells in a dose dependent manner. Furthermore, the peptide modified hydrogels stimulated tube-like structure formation of encapsulated endothelial cells. When integrin αvβ3 or α5β1were antibody blocked during cell encapsulation in peptide modified hydrogels, tube formation was abolished. Therefore, the dual protective nature of the novel peptide QHREDGS may position this peptide as an appealing augmentation for collagen-chitosan hydrogels that could be used for biomaterial delivered cell therapies in the settings of myocardial infarction.  相似文献

This review paper summarises the environmental effects of runoff from wood handling sites including log yards. The characteristics of site runoff and the corresponding effects on the receiving watercourses are presented for worldwide case studies, highlighting the urgent need to address the water pollution problem associated with the wood industry. The methods used to reduce the negative environmental impact of the runoff, such as constructed wetlands, soil infiltration and chemical oxidation, are evaluated. The principal environmental problem of runoff is usually the high concentration of organic substances originating from the wood and bark, some of which are toxic to aquatic life. Phosphorus is also a problem according to some studies. The toxicity of the runoff varies greatly, and depends on the species of tree stored, the amount of water the wood has been in contact with and the degree of runoff treatment.  相似文献

Our objective was to develop a rapid and accurate procedure to genotype common bean plants for the bc-1 2 allele, which conditions resistance to bean common mosaic and bean common mosaic necrosis viruses. A segregating F2 population was derived from the cross between pinto bean breeding lines P94207-43 (bc-1 2//bc-1 2) and P94207-189 (bc-1//bc-1). A quantitative PCR assay based on the detection of fluorescent labeled amplicons was developed to distinguish between homozygous (bc-1 2//bc-1 2), heterozygous (bc-1 2//bc-1) and null (bc-1//bc-1) F2 genotypes. Remnant F1 plants were used as a comparative reference sample. PCR results among this sample fit a normal distribution, and 99% and 95% confidence intervals for heterozygotes were determined. F2 plants for which no amplification was detected were classified as null (bc-1//bc-1) genotypes. F2 plants that fell within the confidence intervals for heterozygotes were classified as heterozygotes (bc-1 2//bc-1), while plants that fell outside the right tail of the heterozygote confidence intervals were classified as homozygotes (bc-1 2//bc-1 2). F2 plants were also genotyped for the bc-1 2 allele by performing F3 family progeny tests for virus resistance. Agreement between the two methods for genotyping plants was 100% (59/59) when PCR genotyping was based on a 99% heterozygote confidence interval, and 98.3% (58/59) when based on a 95% heterozygote confidence interval. This assay will accelerate breeding for virus resistance in bean by facilitating discrimination among plants that are heterozygous or homozygous for the bc-1 2 allele. The experimental design may be generally applicable towards developing other assays for the codominant interpretation of dominant markers in diploid plants.  相似文献
