The human neuroblastoma cell line SK-N-BE can be induced to differentiate by retinoic acid (RA) or by alpha-difluoromethylornithine (DFMO). The former inducer produces neurite outgrowth, 60% reduction of growth rate, overexpression of neural antigens, and enhanced gamma-aminobutyric acid (GABA) and acetylcholinesterase levels. In contrast, DFMO causes cell body elongation, complete growth inhibition, and higher binding of antibodies directed against neuroectodermal antigens. Polyamine metabolism is also differently affected by the two agents. In particular a large spermine catabolism is induced by RA, while DFMO treatment leads to a small increase in the level of this compound. The neural differentiation induced by RA is accompanied by a marked increase in transglutaminase activity and its induction is paralleled by a transient increase of putrescine and spermidine. The putrescine and spermidine depletion determined by DFMO is accompanied instead by a large inhibition of transglutaminase activity. The inhibiting effect of DFMO treatment on transglutaminase is reversed by the addition of 1 mM putrescine to the culture medium. In the presence of both RA and DFMO a mixed morphological and biochemical pattern is observed. The possibility that the expression of transglutaminase associated to cellular differentiation may be modulated by the level of its substrates is also discussed.  相似文献

Rhizobium leguminosarum bv. trifolii strain TA1 is an aerobic, motile, Gram-negative, non-spore-forming rod that is an effective nitrogen fixing microsymbiont on the perennial clovers originating from Europe and the Mediterranean basin. TA1 however is ineffective with many annual and perennial clovers originating from Africa and America. Here we describe the features of R. leguminosarum bv. trifolii strain TA1, together with genome sequence information and annotation. The 8,618,824 bp high-quality-draft genome is arranged in a 6 scaffold of 32 contigs, contains 8,493 protein-coding genes and 83 RNA-only encoding genes, and is one of 20 rhizobial genomes sequenced as part of the DOE Joint Genome Institute 2010 Community Sequencing Program.  相似文献

There is accumulating evidence that acetylcholinesterase has secondary noncholinergic functions, related to adhesion, differentiation, and the deposition of beta-amyloid in Alzheimer's disease. We have observed that the specific acetylcholinesterase peripheral anionic site inhibitors, BW284c51 and propidium iodide, abrogated cell-substrate adhesion in three human neuroblastoma cell lines. The active-site inhibitors, eserine and edrophonium, in contrast, had no effect. Certain anti-AChE antibodies were also shown to inhibit adhesion. Of these, the most effective were a monoclonal (E8) and a polyclonal having cholinesterase-like catalytic activity. These were raised against an acetylcholinesterase-inhibitor complex, implying that the epitope is associated with active-site structures. Two other monoclonal antibodies (E62A1 and E65E8) partially inhibited adhesion. The epitopes of these antibodies have been shown to overlap the peripheral anionic site of acetylcholinesterase. Competition ELISA between the monoclonal antibodies and inhibitors indicated competition between E8, E62A1, and E65E8 and the peripheral-site inhibitors BW284c51 and propidium, but not with the active-site inhibitors eserine and edrophonium. Fluorescence titration between antibodies and propidium confirmed these results. We conclude that the adhesion function of acetylcholinesterase is located at the peripheral anionic site. This has implications, not only for our understanding of neural development and its disorders, but also for the treatment of neuroblastoma, the leukemias, and Alzheimer's disease.  相似文献

By crossing Huntington's disease (HD) R6/1 transgenic mice with 'tissue' transglutaminase (TG2) knock-out mice, we have demonstrated that this multifunctional enzyme plays an important role in the neuronal death characterising this disorder in vivo. In fact, a large reduction in cell death is observed in R6/1, TG2(-/-) compared with R6/1 transgenic mice. In addition, we have shown that the formation of neuronal intranuclear inclusions (NII) is potentiated in absence of the 'tissue' transglutaminase. These phenomena are paralleled by a significant improvement both in motor performances and survival of R6/1, TG2(-/-) versus R6/1 mice. Taken together these findings suggest an important role for tissue transglutaminase in the regulation of neuronal cell death occurring in Huntington's disease.  相似文献
