Rabies is a fatal neurologic disease caused by lyssavirus infection. Bats are important natural reservoir hosts of various lyssaviruses that can be transmitted to people. The epidemiology and pathogenesis of rabies in bats are poorly understood, making it difficult to prevent zoonotic transmission. To further our understanding of lyssavirus pathogenesis in a natural bat host, an experimental model using straw-colored fruit bats (Eidolon helvum) and Lagos bat virus, an endemic lyssavirus in this species, was developed. To determine the lowest viral dose resulting in 100% productive infection, bats in five groups (four bats per group) were inoculated intramuscularly with one of five doses, ranging from 100.1 to 104.1 median tissue culture infectious dose (TCID50). More bats died due to the development of rabies after the middle dose (102.1 TCID50, 4/4 bats) than after lower (101.1, 2/4; 101.1, 2/4) or higher (103.1, 2/4; 104.1, 2/4) doses of virus. In the two highest dose groups, 4/8 bats developed rabies. Of those bats that remained healthy 3/4 bats seroconverted, suggesting that high antigen loads can trigger a strong immune response that abrogates a productive infection. In contrast, in the two lowest dose groups, 3/8 bats developed rabies, 1/8 remained healthy and seroconverted and 4/8 bats remained healthy and did not seroconvert, suggesting these doses are too low to reliably induce infection. The main lesion in all clinically affected bats was meningoencephalitis associated with lyssavirus-positive neurons. Lyssavirus antigen was detected in tongue epithelium (5/11 infected bats) rather than in salivary gland epithelium (0/11), suggesting viral excretion via the tongue. Thus, intramuscular inoculation of 102.1 TCID50 of Lagos bat virus into straw-colored fruit bats is a suitable model for lyssavirus associated bat rabies in a natural reservoir host, and can help with the investigation of lyssavirus infection dynamics in bats.  相似文献

An archaeological excavation in the Tungelroyse Beek valley revealed the remains of two red deer specimens (Cervus elaphus) of Early Mesolithic age that possibly were the victims of hunter-gatherers. The find of animal remains of this age is unique in the Netherlands. In this respect, a sediment core taken close to the remains was investigated, i.e. to reconstruct the vegetation and landscape development of the site and to find more evidence for human activity at this site during the Early Mesolithic. The sediment core shows a typical Early Holocene palynological sequence from the Younger Dryas into the Middle Atlantic, which is supported by AMS dating. The microscopic charcoal record shows peaks in fire activity during the Younger Dryas and Friesland phase, probably wildfire related. Records of spores of coprophilous fungi indicate that the Tungeroyse Beek valley was a favourable place for large herbivores (game) to visit during the investigated period. However, around the age of the oldest red deer remains, no significant peak in fire activity or spores of coprophilous fungi is visible in the investigated record. The pollen diagram does not show disturbed or open vegetation around this age either. This study therefore suggests the impact of Early Mesolithic people on their environment was very low.  相似文献

Dexmedetomidine is a highly selective adrenergic receptor agonist, which has a dose-dependent sedative hypnotic effect. Furthermore, it also has pharmacological properties, and the ability to inhibit sympathetic activity and improve cardiovascular stability during an operation. However, its protective effect on patients with severe craniocerebral injury in the perioperative period remains unclear. Eighty adult male SD rats were used and divided into two groups (n = 40, each group): dexmedetomidine injury group (experimental group), and sodium chloride injury group (control group). Models of severe craniocerebral injury were established in these two groups using the modified Feeney’s free-fall method. As soon as the establishment of models was succeed, rat in the experimental group received 1 μg of dexmedetomidine (0.1 ml), while each rat in the control group was given 0.1 ml of 0.9% sodium chloride. Blood was sampled from an incision at the femoral vein to detect TNF-α and IL-2 levels at 1, 12, 24,36,48 and 72 h after establishing the model in the two groups. After severe craniocerebral injury, TNF-α levels of rats were lower in every stage and at different degrees in the experimental group than in the control group (P < 0.05), while IL-2 levels were lower in the experimental group to different extents (P < 0.05). Dexmedetomidine protects the brain of rats with severe craniocerebral injury by reducing the release of inflammatory mediators.  相似文献

Harbour porpoises (Phocoena phocoena) stranding in large numbers around the southern North Sea with fatal, sharp-edged mutilations have spurred controversy among scientists, the fishing industry and conservationists, whose views about the likely cause differ. The recent detection of grey seal (Halichoerus grypus) DNA in bite marks on three mutilated harbour porpoises, as well as direct observations of grey seal attacks on porpoises, have identified this seal species as a probable cause. Bite mark characteristics were assessed in a retrospective analysis of photographs of dead harbour porpoises that stranded between 2003 and 2013 (n = 1081) on the Dutch coastline. There were 271 animals that were sufficiently fresh to allow macroscopic assessment of grey seal-associated wounds with certainty. In 25% of these, bite and claw marks were identified that were consistent with the marks found on animals that had tested positive for grey seal DNA. Affected animals were mostly healthy juveniles that had a thick blubber layer and had recently fed. We conclude that the majority of the mutilated harbour porpoises were victims of grey seal attacks and that predation by this species is one of the main causes of death in harbour porpoises in The Netherlands. We provide a decision tree that will help in the identification of future cases of grey seal predation on porpoises.  相似文献

Summary The migration pattern of germinal center cells of the rabbit appendix was studied and compared with that of appendix dome cells, spleen cells, thymus cells and thoracic duct lymphocytes. To discriminate T-and B-cell migration pathways, normal or T-cell-depleted rabbits were used as donors. Cell suspensions were labeled in vitro with 3H-leucine followed by intravenous transfer. The migration of labeled cells in lymphoid organs was studied using autoradiography, particular attention being paid to the spleen of the recipient. B-cells from the appendix dome, spleen and thoracic-duct lymph migrate to primary follicles or the corona of secondary follicles via thymus-dependent areas of peripheral lymphoid organs. In contrast, a B-cell subpopulation from the germinal centers of the appendix migrates to the center of splenic primary follicles and into germinal centers. The migration of germinal center cells to splenic follicle centers is not enhanced by specific antigens. The migration properties of B-cells, possibly changing during differentiation, may be instrumental in the two types of immune reactions, i.e., plasma-cell reaction and germinal-center reaction.  相似文献

The species Urtica dioica L., Plantago major ssp. major L., Plantago lanceolata L., Hypochaeris radicata L. ssp. radicata and Hypochaeris radicata ssp. ericetorum Van Soest were grown under high and low nutrient conditions (1/4 Hoagland and 2% of 1/4 Hoagland further called the 100% and 2% treatment, containing 3.75 mM NO-3 and 0.075 mM NO-3, respectively). After a certain period half of the plants were transferred from low to high or high to low nutrients, yielding the 100%/2% and the 2%/100% treatments. The kinetics of nitrate uptake in the range of system I of the five species grown under the different nutrient conditions were measured during a three week experimental period. The nitrate uptake of all the species showed the characteristic features of Michaelis-Menten kinetics. Under low nutrient conditions the apparent Vmax of U. dioica expressed per g dry root was lower than under high nutrient conditions. For H. radicata ssp. radicata and for H. radicata ssp. ericetorum the reverse was found. The Vmax values of P. major ssp. major were almost the same for the two treatments. The apparent Vmax in young plants of P. lanceolata was higher in the 100% treatment than in 2%; whereas the reverse was found in mature plants. The results are explained in relation to the relative growth rate, the shoot to root ratio and the natural environment of the species. The apparent Km values were not influenced by the different treatments. Differences in Km between the species, if any, were very small. It is suggested that the Vmax is a more important parameter for the distribution of plant species in the field than the Km. The rate of nitrogen accumulation was calculated from growth data and the contents of nitrate and reduced nitrogen. It is concluded that the Vmax of system I for nitrate uptake in most cases was sufficient to explain the observed growth rates.  相似文献

Plantago major L. ssp. major , a grassland species from a relatively nutrient-rich habitat, was grown in nutrient-rich and nutrient-poor culture solutions. Half of the plants were transferred from high to low or from low to high nutrient conditions. The rate of dry matter accumulation in both shoots and roots decreased slowly upon transfer of plants to low nutrient conditions and the shoot to root ratio was unaffected. The rate of structural growth of both roots and shoots increased upon transfer from low to high nutrient conditions and the shoot to root ratio, if calculated from non-structural-carbohydrate-free dry weights, increased.
Photosynthesis was largely independent of the nutrient supply. Root respiration, particularly the activity of the alternative oxidative pathway, decreased with increasing age. This decrease was ascribed to a decreased shoot to root ratio, which reduced the relative amount of carbohydrates translocated to the roots and thus the amount available for the alternative pathway. It is calculated that in young as well as in old plants grown in full nutrient solution 48% of the daily produced photosynthates was translocated to the roots.
This is at variance with data on P. lanceolata , where a decreasing proportion of the daily produced photosynthates was translocated to the roots when the plants grew older. It is concluded that shoot growth plus shoot respiration consumed a constant amount of the daily produced photosynthates in P. major and that the rest was left for translocation. It is further calculated that in P. major plants grown in full nutrient solution c . 25% and c . 2% of the daily produced photosynthates in young and old plants, respectively, was respired in a way that is not involved in production of energy that is utilized in growth and maintenance ('inefficient root respiration').
The results are discussed in comparison with those of P. lanceolata , a species from a relatively nutrient-poor habitat.  相似文献

Various aspects of the nitrogen metabolism of Plantago major L. ssp. major , a grassland species from a relatively nutrient-rich habitat, were investigated with plants that were grown in nutrient-rich and nutrient-poor culture solutions. In addition the influence of a change in the nutrient supply, both from high to low and from low to high nutrient conditions was studied. The data on P. major major showed that the activities of nitrate reductase (NR) and the reduced nitrogen content were all rather independent of the nutrient supply. These findings are in contrast with those of P. lanceolata , a grassland species from a relatively nutrient-poor habitat, where both the activities of NR, GDH and GS and the reduced nitrogen content were correlated with the level of the nutrient supply. The rigidity of the nitrogen metabolism of P. major major may be of advantage if the nutrient supply is limited only for relatively short periods.  相似文献

Plantago lanceolata L., a grassland species from a relatively nutrient-poor habitat, was grown in nutrient-rich and in nutrient-poor culture solutions. Half of the plants were trensferred from high to low or from low to high nutrient conditions. Shoot growth was immediately reduced upon transfer to low nutrient conditions, whilst it reacted more slowly upon transfer of plants to high nutrient conditions. Root growth was less dependent on the supply of nutrients, but it was slightly reduced upon transfer of plants to high nutrient conditions.
Photosynthesis was largely independent of the nutrient supply, apart from an initial increase upon transfer of plants to low nutrient conditions. Photosynthesis decreased with age in all treatments, and this decrease was not due to mutual shading. The decrease of photosynthetic rate was not accompanied by a decreased relative growth rate: it was compensated by a more efficient root respiration, since the activity of the alternative nonphosphorylating pathway continuously decreased in plants grown in a high nutrient environment.
It is concluded that the alternative pathway was of significance in removal of carbohydrates, which could not be utilized for growth, energy production, etc. , due to a temporary or structural imbalance between assimilate production and requirement. The alternative pathway also appeared to allow P. lanceolata plants to adapt to a changed environment as regards mineral nutrition.
The experimental value for root growth respiration of P. lanceolata grown under high nutrient conditions was compared with a theoretical value, calculated from the biochemical composition of plant dry matter and the known energy costings for biosynthetic and transport processes. A good correlation between the experimental and theoretical value of root growth respiration was found if it was assumed that ion uptake required c . 1.0 molecule of ATP per ion per membrane passage.  相似文献

Various aspects of nitrogen metabolism of Plantago lanceolata L., a grassland species from a relatively nutrient poor habitat, were investigated under high nutrient conditions or low nutrient conditions. In addition the responses after switching the plants from high nutrient conditions to low nutrient conditions and vice versa were studied. The activities of nitrate reductase (NR), glutamate dehydrogenase (GDH) and glutamine synthetase (GS) were determined. In the roots they were correlated with the level of the nutrient supply. Not only NR but also GDH and GS increased upon a switch from low nutrients to high nutrients and decreased upon a switch from high nutrients to low nutrients. Reduced nitrogen content of both roots and shoots was also correlated with the nutrient supply and changed rapidly after a switch. The flexibility of the nitrogen metabolism of Plantago lanceolata is discussed in relation to its ecological habitat.  相似文献
