We demonstrate that the combined application of Langmuir-Blodgett and self-assembly techniques allows the fabrication of patterns with contrasting surface properties on gold substrates. The process is monitored using fluorescence microscopy and surface plasmon spectroscopy and microscopy. These structures are suitable for the investigation of biochemical processes at surfaces and in ultrathin films. Two examples of such processes are shown. In the first example, the structures are addressed through the binding of a monoclonal antibody to a peptide. This demonstrates the formation of self-assembled monolayers by cysteine-bearing peptides on gold, and the directed binding of proteins to the structured layers. A high contrast between specific and unspecific binding of proteins is observed by the patterned presentation of antigens. Such films possess considerable potential for the design of multichannel sensor devices. In the second example, a structured phospholipid layer is produced by controlled self-assembly from vesicle solution. The structures created--areas of phospholipid bilayer, surrounded by a matrix of phospholipid monolayer--allow formation of a supported bilayer which is robust and strongly bound to the gold support, with small areas of free-standing bilayer which very closely resemble a phospholipid cell membrane.  相似文献

We quantified the vigilance levels of elk (Cervus elaphus) preyedon by wolves (Canis lupus) in Yellowstone National Park, betweenJanuary and May in 2005 and 2006, and used Akaike's informationcriterion to compare a set of 38 regression models for vigilancelevels. These models combined up to 9 predictor variables of3 types: characteristics of the prey group (herd size and composition),characteristics of the predator (wolf pack size, distance away,and the presence/absence of a kill), and characteristics ofthe local environment (distance to woodland edges, snow depth,and snow cover). The set of models spanned a range of complexityfrom simple univariate models to complex combinations with upto 3 variables of each type. Complex models incorporating thecharacteristics of the wolf pack, the structure of the elk herd,and the environmental conditions had higher information contentthan simple models. Although univariate models of vigilancedetect significant relationships, they have low informationcontent relative to multivariate models. These results showthat elk assesses factors of several types when assessing riskand deciding how much time to allocate to vigilance. In particular,we found that all well-supported models of vigilance includedseveral "prey" variables and several "predator" variables. Thisresult highlights the need to consider information about predatorswhen trying to explain variation in vigilance levels in prey.  相似文献

In the protogynous stoplight parrotfish (Sparisoma viride), large males defend territories that encompass the home-ranges of several mature females. However, high-quality habitat is in short supply, such that smaller, competitively inferior males do not defend territories. We investigated the role of 11-ketotestosterone (11KT) and testosterone (T) in the regulation of territorial behavior in a wild population of a protogynous reef fish, the stoplight parrotfish, at Glover's Reef, Belize. Radioimmunoassay of plasma samples from individuals of known social status revealed that nonterritorial males have lower levels of T and 11KT than territorial males. Nonterritorial males allowed access to vacant territories underwent pronounced increases in T and 11KT. When sampled 1 week after territory acquisition, levels of T and 11KT in new territorial males were significantly higher than the levels in established territorial males, but by 3 weeks after territory acquisition, there was no significant difference. We further investigated the hypothesis that such short-term increases in androgen levels are a response to intense male-male interactions during territory establishment. Simulated territorial intrusion promoted increased plasma levels of both T and 11KT while access to vacant territories without neighboring territorial males did not. These findings suggest that the endocrine system plays a role in fine-tuning the levels of territorial aggression exhibited by male stoplight parrotfish. We discuss these results in light of recent theory in behavioral endocrinology.  相似文献

A method has been developed for isolating differentiated type II cells from human lung of 18-24-week gestation. The procedure involves an initial 4-day culture of lung explants in the presence of dexamethasone (10 nM) and triiodothyronine (2 nM). Type II cells (and fibroblasts) are isolated by trypsin digestion of the explants, two differential adherence steps and incubation overnight in primary culture. This method provides a high yield of type II cells ((50 +/- 15) X 10(6) cells/g wet weight of explant) with a purity of 85 +/- 5% in 16 experiments. The type II cells contain numerous perinuclear granules which stain darkly with toluidine blue and Papanicolaou stain; electron microscopy showed these inclusions to be lamellar bodies with tightly stacked, well defined lamellae. Type II cells, but not fibroblasts, were positive by immunofluorescence histology for surfactant apoprotein and binding of Maclura pomifera lectin which binds to the surface of type II but not type I cells in vivo. The rate of both [3H]acetate and [3H]choline incorporation into phosphatidylcholine (PC) was several-fold greater in type II cells than fibroblasts; the saturation of PC was 36.2 and 25.9%, respectively. Release of saturated PC was stimulated by terbutaline, the ionophore A23187, and tetradecanoyl phorbol acetate in type II cells but not fibroblasts. We conclude that differentiated type II cells can be isolated in relatively high yield and purity from hormone-treated explants of fetal human lung.  相似文献

We have examined the effect of explant culture and hormones on the major surfactant associated protein of Mr 28,000-36,000 (SP 28-36) in human fetal lung. Explants of 16- to 23-week gestation lung were maintained for up to 5 days in culture. Polyclonal antibodies raised to SP 28-36 purified from alveolar proteinosis lung lavage were used in immunofluorescence experiments (n = 11). There was no specific fluorescence seen in frozen sections of preculture tissue. In explants cultured without serum or hormones, fluorescence was seen in most epithelial cells lining potential airspaces. In cultures treated with 10 nM dexamethasone and 2 nM T3 much brighter fluorescence was seen in virtually all epithelial cells. Immunofluorescence studies on cell monolayers prepared from explants confirmed that SP 28-36 is found in the cytoplasm of type II cells but not in fibroblasts. The pattern of fluorescence was consistent with the presence of SP 28-36 on rough endoplasmic reticulum. SP 28-36 mRNA was measured in isolated cell populations using a 32P-labeled cDNA probe. mRNA levels were manyfold higher in type II cell preparations (purity 78-92%) than in fibroblasts (purity 81-97%). A competitive enzyme linked assay was developed to quantify SP 28-36. The SP 28-36 content of five lungs before culture (17-23 weeks) was less than 0.02 microgram/mg DNA. During explant culture without hormones the SP 28-36 content increased exponentially. Exposure to dexamethasone accelerated the increase in SP 28-36 content. T3, alone or in the presence of dexamethasone, did not influence SP 28-36 content. We conclude that SP 28-36 content is very low in human fetal lung before 24 weeks gestation. Explant culture and treatment with dexamethasone synchronize development of type II cells from epithelial precursors, and induce synthesis of SP 28-36 in type II cells. These findings provide evidence of concomitant regulation by glucocorticoids of the phospholipid synthetic enzymes and the major protein of pulmonary surfactant.  相似文献

Although theory and widespread evidence show that the evolution of egg size is driven primarily by offspring and maternal fitness demands, an additional explanation invokes sperm limitation as a selective force that could also influence egg size optima. Levitan proposed that constraints from gamete encounter in external fertilization environments could select for enlargement of ova to increase the physical size of the fertilization target. We test this theory using in vitro fertilization experiments in an externally fertilizing fish. Sockeye salmon (Onchorhyncus nerka) females show considerable between-individual variation in ovum size, and we explored the consequences of this natural variation for the fertilization success of individual eggs under conditions of sperm limitation. By engineering consistent conditions where in vitro fertilization rate was always intermediate, we were able to compare the sizes of fertilized and unfertilized eggs across 20 fertilization replicates. After controlling for any changes in volume through incubation, results showed that successfully fertilized eggs were significantly larger than the eggs that failed to achieve fertilization. Under conditions without sperm limitation, fertility was unaffected by egg size. Our findings therefore support Levitan''s theory, demonstrating empirically that some element of egg size variation could be selected by fertilization demands under sperm limitation. However, further research on sperm limitation in natural spawnings is required to assess the selective importance of these results.  相似文献

The reproductive seasonality of Puntius schwanenfeldii from the Perak River, Malaysia was investigated over a 19 month period. Females greater than 160g had mature or rematuring ovaries, and males of all sizes had mature testes throughout the study period. Analysis of female reproductive condition was based upon the presence of post ovulatory follicles (POF), the gonadosomatic index (GSI) and the volume fraction of stage IV oocytes (IV%), and GSI and presence of tubercles for males. Recrudescence followed immediately after ovulation, which allowed the use of objective thresholds to identify recently spawned females during the GSI and IV% recovery period. GSI and IV% were the best indicators of reproductive patterns as a result of their recovery period being longer than for POF. Male tubercles and GSI showed some statistically significant variation, but no clear relation with observed changes in female indicators. Reproduction was apparently aseasonal, and this pattern is likely an effect of flow regulation by dams which have virtually eliminated the dominant flood period and most seasonal flow variation. Hypothesized shifts in the reproductive seasonality due to flow regulation are consistent with predictions based on life history theory under conditions of altered environmental seasonality.  相似文献

Fatigue failure of bones of the metacarpo-phalangeal (fetlock, MCP) joint is common in thoroughbred racehorses. Stresses within the fetlock joint cartilages are affected by the morphology of the third metacarpal bone (MC3) and proximal phalangeal bone, and the steepness of the median sagittal ridge of MC3 is believed to be associated with fracture. This study investigated the influence of the steepness of the sagittal ridge on cartilage stress distribution using a finite element model of the joint. Changes to the steepness of the sagittal ridge were made by applying a parabolic function to the mesh, creating four different models with sagittal ridge angles ranging from 95° to 105°. In the fetlock joint of Thoroughbred racehorses, sagittal ridge angles of >100° were associated with higher Von Mises stresses in cartilage at the palmar aspect of the condylar groove than such stresses in joints with sagittal ridge angles of <100°. Stresses were high in the specific region where fractures are known to originate in MC3. This aspect of morphology of the fetlock joint thus appears to play an important role in the magnitude and distribution of cartilage stresses, which, when acting on the underlying hard tissues of the articular calcified cartilage and subchondral bone may play a role in the initiation of fatigue fracture in the third metacarpal bone.  相似文献

The ability of larger males to control resources and monopolise spawnings is thought to explain the benefits of protogyny in some animals. Here we describe the breeding and non-breeding social organisations of a temperate protogynous goby, Coryphopterus nicholsii, in the light of this prediction. Our results confirm that C. nicholsii has a stable social organisation, based on the year-round defence of a territory by both females and males. As predicted, larger individuals of both sexes were more likely to defend territories than smaller ones. These territories always included one or more shelter rocks, which were used as refuges by both sexes throughout the year, and also as nest sites by males in the breeding season. We then use experimental manipulation of shelter rock availability to examine the influence of intraspecific competition on territory establishment by smaller males. Following the removal of resident, non-nesting males, territories were quickly taken over. In addition, artificial reefs and nest sites were quickly colonised. Therefore, it appears that suitable shelter rocks are limited. Furthermore, intraspecific competition is strongly size-specific in C. nicholsii and determines which individuals gain access to shelter rocks. The presence of these social conditions is consistent with the prediction that the ability of larger males to monopolise more than one mate affects the occurrence of protogyny in some animals, and may clarify the occurrence of protogyny in C. nicholsii.  相似文献
