Structures and stability of salt-bridges in aqueous solutions were investigated using a complex formed from the guanidinium (Gdm+) and formate (FmO-) ions as a model system. The Test-particle model (T-model) potentials to describe the interactions in the Gdm+-H2O, FmO(-)-H2O and Gdm+-FmO- complexes were constructed, tested and applied in molecular dynamics (MD) simulations of the aqueous solutions at 298 K. The three-dimensional structures and energetic of the hydrogen bond (H-bond) networks of water in the first hydration shells of the Gdm+ and FmO- ions, as well as the Gdm+-FmO- complex, were visualized and analyzed using various probability distribution (PD) maps. The structures of the average potential energy landscapes at the H-bond networks were employed to characterize the stability and dynamic behavior of water molecules in the first hydration shells of the solutes. It was observed that water molecules in the first hydration shell of the close-contact Gdm+-FmO- complex form associated H-bond networks, which introduce a net stabilization effect to the ion-pair, whereas those in the interstitial H-bond network destabilize and break the solvent-separated Gdm+-FmO- complex. The present results showed that, in order to provide complete insights into the structures and stability of ion-pairs in aqueous solutions, explicit water molecules have to be included in the model calculations.  相似文献

Novel hydrogel polyelectrolyte complexes (PECs) between the N,N,N,-trimethylchitosan-homocysteine thiolactone (TM-HT-chitosan) and two anionic polymers were investigated. The particles of pure thiolated chitosan and its PECs with alginate and carrageenan were fabricated using the electrospray ionization technique. The hydrogel PEC particles were characterized by scanning electron microscopy, dynamic light scattering, Fourier transform infrared microscopy, thermogravimetric analysis, encapsulation efficiency (EE), mucoadhesive property and in vitro drug release behavior. TM-HT-chitosan/alginate particles could be loaded with camptothecin (CPT), employed as a model anti-cancer drug, at an over 70% EE, and revealed both a reduced burst effect and a prolonged release of CPT over 3 days. The resultant TM-HT-chitosan/alginate PEC particles displayed a 5.60-, 1.86- and 1.55-fold stronger mucoadhesive property compared to that of the unmodified chitosan/alginate PEC at pH 1.2, 4.0 and 6.4, respectively, and this was not affected by the CPT loading level.  相似文献

The effects of metal ion and solute conformation change on the structures, energetic and dynamics of water molecules in the first hydration shell of amino acid were studied, using three forms of alanine (Ala) and Li(+)/Ala as model molecules. The theoretical investigations were started with construction of the test-particle model (T-model) potentials for all molecules involved and followed by molecular dynamics (MD) simulations of [Ala](aq) and [Li(+)/Ala](aq) at 298 K. The MD results showed that the hydrogen bond (H-bond) networks of water at the functional groups of Ala are strengthened by the metal ion binding, whereas the rotation of the N-C(alpha) bond from the angle phi=0 degrees to 180 degrees brings about smaller effects which cannot be generalized. It was also shown that the dynamics of water molecule in the first hydration shell of amino acid could be estimated from the total-average potential energy landscapes and the water exchange diagrams. The MD results suggested inclusion of an additional dynamic step in the water exchange process, in which water molecule moves inside a channel within the first hydration shell of solute, before leaving the channel at some point. The theoretical results reported in the present work iterated the necessity to include explicit water molecules in the model calculations.  相似文献

Diatoms have been recognized as a promising raw material for biodiesel production as a result of their thermal stability. However, potent thermotolerant strains of diatoms, such as hot spring diatoms, have not yet been studied. Achnanthidium exiguum AARL D025–2 was originally isolated from a habitat at temperatures ranging from 39 to 45 °C; however, it could only be isolated and maintained at 30 °C. Hence, it has been determined that certain factors have a considerable impact on the thermostability of this diatom. One factor that has not yet been given as much consideration is pH. The pH can be significant for the growing conditions and physiology of algal cells. Thus, this study aims to investigate the effect of pH on thermotolerant diatom cultivation. The diatom A. exiguum AARL D025–2 was cultivated in modified Bold Basal Medium at different pH (7.0, 8.0, 9.0, and 10.0) and at temperatures of 30, 40, and 50 °C. Growth was monitored by direct cell-counting, while fatty acid methyl ester (FAME) composition was also examined. The results showed that an alkaline pH could promote the heat tolerance of this diatom. The optimal culture conditions for biomass and FAME production was pH 9 at 40 °C. The major FAMEs were C16-C18, which may be suitable for the purposes of biodiesel production. The results from this research study will be useful for the future development of thermotolerant diatom cultivation with regard to the study and promotion of alternative energy sources.  相似文献

Background

Age-related changes occur in both the peripheral and central nervous system, yet little is known about the influence of chronic pain on pain sensitivity in older persons. The aim of this study was to investigate pain sensitivity in elders with chronic neck pain compared to healthy elders.

Methods

Thirty elderly women with chronic neck pain and 30 controls were recruited. Measures of pain sensitivity included pressure pain thresholds, heat/cold pain thresholds and suprathreshold heat pain responses. The pain measures were assessed over the cervical spine and at a remote site, the tibialis anterior muscle.

Results

Elders with chronic neck pain had lower pressure pain threshold over the articular pillar of C5-C6 and decreased cold pain thresholds over the cervical spine and tibialis anterior muscle when compared with controls (p < 0.05). There were no between group differences in heat pain thresholds and suprathreshold heat pain responses (p > 0.05).

Conclusion

The presence of pain hypersensitivity in elderly women with chronic neck pain appears to be dependent on types of painful stimuli. This may reflect changes in the peripheral and central nervous system with age.  相似文献

This study deals with five species of the Barbirostris Complex of Anopheles subgenus Anopheles that are known to occur in Thailand. Three new species of the complex, A nopheles dissidens sp. nov. , A nopheles saeungae sp. nov. , and A nopheles wejchoochotei sp. nov. , are characterized and compared with Anopheles barbirostris van der Wulp and Anopheles campestris Reid based on specimens of molecularly identified progeny broods. For practical purposes, the five species are essentially isomorphic and can only be unequivocally identified from diagnostic mitochondrial and ribosomal DNA sequences. Based on overall morphological similarity, An. campestris is considered to be a member of the Barbirostris Complex rather than a separate member of the Barbirostris Subgroup. The molecular data, mitotic karyotypes, bionomics, and distributions of the species are reviewed and discussed. It is concluded that integrated molecular epidemiological studies of the complex throughout the Oriental Region are needed to unambiguously elucidate the individual species and their relation to disease. © 2015 The Linnean Society of London  相似文献

We characterized a collection of 256 Aeromonas hydrophila strains isolated from blood, discharge and stool for their serogroup designation. Of these, 2.3% were untypable and 15.2% were rough strains. Among the typable strains, about 50% comprised serogroups O:11, O:16, O:18, O:34 and O:83. To develop rapid differentiation of Aeromonas from other oxidase-positive bacteria, antisera against Aeromonas were produced to establish a direct, genus-specific, agglutination test. It was found that among 105 isolates of Aeromonas, 102 showed positive results with the agglutination test. The calculated sensitivity and specificity were 97.1% and 90.7%, respectively.  相似文献

Eight species members of the Thai Hyrcanus Group were identified based on the intact morphology and molecular analysis (COI barcoding, 658 bp) of F1-progenies. Five iso-female lines of each species were pooled in order to establish stock colonies. A stenogamous colony of each species was investigated by making 200 and 300 newly emerged adult females and males co-habit in a 30 cm cubic cage for one week. After ovipositon, the spermathecae of females were examined for sperms. The results revealed that Anopheles argyropus, Anopheles crawfordi, Anopheles nitidus, Anopheles pursati, Anopheles sinensis, Anopheles nigerrimus, Anopheles paraliae and Anopheles peditaeniatus yielded insemination rates of 0%, 0%, 0%, 31%, 33%, 42%, 50% and 77%, respectively. Continuous selection to establish stenogamous colonies indicated that An. sinensis, An. pursati, An. nigerrimus, An. paraliae and An. peditaeniatus provided insemination rates of 33–34%, 27–31%, 42–58%, 43–57% and 61–86% in 1, 2, 5, 6 and 20 generations of passages, respectively.  相似文献

To assess differentiation and relationships between Anopheles lesteri and Anopheles paraliae we established three and five iso-female lines of An. lesteri from Korea and An. paraliae from Thailand, respectively. These isolines were used to investigate the genetic relationships between the two taxa by crossing experiments and by comparing DNA sequences of ribosomal DNA second internal transcribed spacer (ITS2) and mitochondrial DNA cytochrome c oxidase subunit I (COI) and subunit II (COII). Results of reciprocal and F1-hybrid crosses between An. lesteri and An. paraliae indicated that they were compatible genetically producing viable progenies and complete synaptic salivary gland polytene chromosomes without inversion loops in all chromosome arms. The pairwise genetic distances of ITS2, COI and COII between these morphological species were 0.040, 0.007-0.017 and 0.008-0.011, respectively. The specific species status of An. paraliae in Thailand and/or other parts of the continent are discussed.  相似文献

The Hyrcanus Group comprises important vectors of malaria because of Plasmodium vivax Grassi and Feletti and filariasis caused by Brugia malayi Brug in many countries of South, Southeast and East Asian regions. In Thailand, eight species members (Anopheles argyropus Swellengrebel, An. crawfordi Reid, An. nigerrimus Giles, An. nitidus Harrison, Scanlon & Reid, An. paraliae Sandosham, An. peditaeniatus Leicester, An. pursati Laveran and An. sinensis Wiedemann) of the Hyrcanus Group have been recognized. Due to morphological overlap, adult females of the Hyrcanus Group in Thailand have been misidentified markedly among the eight species, particularly when using the traumatic scales of wild-caught specimens from epidemiology and control approaches. Therefore, this study first developed a simple and robust multiplex PCR assay, based on second internal transcribed spacer sequences of ribosomal DNA, for differentiating the eight species members of the Thai Hyrcanus Group.  相似文献
