Abstract A new experimental system was devised for the simultaneous measurement of elongation rate and the activity of the spatially separate electrogenic ion pumps of a hypocotyl segment excised from a seedling of Vigna unguiculata L. Walp. under enforced intra-organ perfusion by artificial solutions. The pathway of the perfusion medium was apoplastic space, including xylem vessels as main routes. The elongation rate of the segment was highly dependent on the perfusion pressure applied. It was possible to increase the growth rate under pressurized perfusion by 10-30 times as much as that without perfusion. Elongation rate was also dependent on respiration under perfusion, being retarded reversibly by anoxia a few minutes after the activities of the electrogenic ion pumps were stopped. Perfusion pressure had a little influence on the membrane potential (Vpx) below a breakdown level (c. 130 kPa). Perfusion of mannitol or sorbitol solution of appropriate concentration reduced the elongation rate reversibly. 相似文献

Cells of younger cultures were found to be moreactivethan those of older cultures, with respect both to respirationand photosynthesis.
- The pigment content was found to be higherin the light-growncells than in the dark-grown ones. The ratiophotosyntheticactivity/bacteriochlorophyll was significantlyhigher in thelatter than in the former.

The sea lavender, Limonium wrightii , has six morphs of flower colour variation. The geographical distribution of flower colour morphs is disjunct; the distribution of the pink flower morph is divided into two subregions, and that of the yellow flower morph intervenes between them. The present study aimed to examine the origin of this apparent distribution pattern of flower colour in L. wrightii . Two main hypotheses (i.e. past dispersal events and phenotypic changes by natural selection and/or stochastic processes) have been proposed to account for the origin of leapfrog distribution patterns. To determine which hypothesis was applicable, we conducted a molecular phylogenetic analysis using sequence variation in chloroplast DNA (three regions of intergenic spacers, trnG - trnfM , trnV - trnM , and psbA-trnH ). We sequenced 58 accessions of L. wrightii frin 28 islands in the Ryukyu Archipelago and the Izu-Ogasawara Islands, located south of the Japanese mainland, and 12 accessions of four congeneric species. Within L. wrightii , we obtained four lineages of ten haplotypes. These lineages and haplotypes did not correlate with the different flower colours. These results indicate that the formation processes of populations are complex. The haplotypes of the pink flower morph did not show a sister relationship between the two disjunct subregions, indicating that the disjunct populations of the pink flower morphs are unlikely to share the pink flower colour as a result of common ancestry. Ten polymorphic microsatellite markers were developed in the drywood termite Incisitermes minor (Hagen) by using a genomic DNA extracted from the heads of workers. The microsatellite markers obtained in this study produced between two and seven alleles per locus. The observed and expected heterozygosities among the 10 markers ranged from 0.16 to 0.83 and from 0.43 to 0.84, respectively. These markers were shown to be good molecular tools for identification of the genetic structure and parentage assessment in I. minor. 相似文献

- 1 Hipposideros turpis is traditionally known as a species composed of three subspecies, H. t. turpis, H. t. alongensis and H. t. pendleburyi, distributed disjunctly in south‐west Japan, north‐east Vietnam and south‐west Thailand, respectively. Prior to the present study, the systematic status of forms within the species remained unclear.
- 2 Using morphological (external, bacular, cranial and dental characters), genetic and echolocation data, we demonstrate that turpis, alongensis and pendleburyi represent three distinct species, and that these species are endemic to Japan, Vietnam and Thailand, respectively. They are very distinct genetically and do not even form a monophyletic group.
- 3 We also prove that H. alongensis is composed of two subspecies, H. a. alongensis and H. a. sungi. The latter subspecies is described as new to science. To date, H. a. alongensis appears to be restricted to the Cat Ba Island of Cat Ba National Park, west Ha Long Bay, whereas H. a. sungi ssp. nov. is known from three localities in mainland northeast Vietnam. These two subspecies are distinguished by body size, molecular data and the frequency of the constant‐frequency component of their echolocation signals.

Partially purified homoserine dehydrogenase was prepared frompea seedlings. The optimum pH for this enzyme is approximately 5.4. The Kmvaluesfor ASA and TPNH are 4.6xl04Af and 7.7xl05M, respectively.This enzyme can also utilize DPNH but less effectively thanTPNH. In contrast with yeast homoserine dehydrogenase whichis insensitive to SH reagents, the pea enzyme is inhibitedalmost completely by 104MPCMB and 105MHgCl2, theinhibition being removed by 102M thioglycolate. Homoserinedehydrogenase was found not only in decotylized seedlings, butalso in cotyledons. The significance of this enzyme in homoserine biosynthesis ingerminating pea seeds has been discussed. The criocerine leaf beetle Lema coronata Baly has extremely long genitalia that reach more than twice the body length in both sexes. We observed mating behavior of this species in the laboratory and inspected the male genital morphology using a scanning electron microscope. The males did not perform pre-copulatory courtship and post-copulatory guarding of the mates, and copulation lasted only for about 30 min. The surface of male genitalia is smooth without any special structures at the tip. A fragment of broken male genitalia was detected in the spermathecal duct of one female. We discuss the adaptive significance of male genital damage and the selective factor of elongated genitalia. 相似文献
