In this paper a modular model of the GnRH neuron is presented. For the aim of simplicity, the currents corresponding to fast time scales and action potential generation are described by an impulsive system, while the slower currents and calcium dynamics are described by usual ordinary differential equations (ODEs). The model is able to reproduce the depolarizing afterpotentials, afterhyperpolarization, periodic bursting behavior and the corresponding calcium transients observed in the case of GnRH neurons.  相似文献

In the process of biogenous weathering of Beacon sandstone in the McMurdo Dry Valleys (Ross Desert), Antarctica, periods of microbial growth, on the time scale of 103-104 years, alternate with sudden exfoliation events. The present study addressed the question of whether microbial growth is continuous between exfoliation events or whether each exfoliation is followed by a period of comparatively rapid growth and then an extended period of steady state. The color intensity (Munsell lightness value) of the rock surface is an indicator of relative age of the crust within the exfoliation cycle, permitting measurement of changes in microbial biomass on a geological time scale. Results indicate that microbial growth is continuous and that exfoliation occurs when the microbial biomass reaches the carrying capacity of the cryptoendolithic habitat.  相似文献

The ground pearls comprise the scale insect family Margarodidae s. str., and are subterranean plant sucking parasites on roots of a wide variety of plants. Some ground pearls are serious destructive pests of grape vines, sugar cane, oil palms, cotton or lawn grass around the world. Both female and male development is characterised by an apodous, feeding 2nd-instar nymph, called a cyst. The cyst is primarily circular and often colourful, shiny, metallic or pearl-like, from which the common name “ground pearls” is derived. Other unique features are the strongly developed prothoracic legs for digging, the construction of a protective test from their own liquid excreta in which the cyst is enclosed and a behavioral adaptation by modification of their life-cycle to survive in adverse environmental conditions.

The 10 genera, currently include 105 species, form a monophyletic group with a worldwide distribution. First-instar nymphs and cysts are feeding instars; however, adults male and females lack mouthparts and do not feed. Species of ground pearls reproduce either bisexually or parthenogenetically, parthenogenesis is facultative in Eurhizococcus brasiliensis. Females undergo three, four or five developmental stages, while male have five. During the cyst stage, one, two or three molts may occur. Most species have a single generation each year, although development in Margarodes vitis requires three years including three instars of cysts. This revision redescribes and illustrates the type species of each genus, including Margarodes vitis (Philippi) and provides a key to the genera based on the morphology of adult females. Lectotypes of Dimargarodes medíterraneus (Silvestri), Heteromargarodes americanus Jakubski, Promargarodes sinensis Silvestri and Termitococcus carrotai Silvestri are designated. A nomenclatural change is proposed, the genus Sphaeraspis Giard, 1894 is considered to be a synonym of Margarodes Guilding, 1829. This work also provides information on the history of the group of Archaeococcoids; on the biology, economic importance and distribution of the margarodids.  相似文献

The sub-retinal pigment epithelial deposits that are a hallmark of age-related macular degeneration contain both C3b and millimolar levels of zinc. C3 is the central protein of complement, whereas C3u is formed by the spontaneous hydrolysis of the thioester bridge in C3. During activation, C3 is cleaved to form active C3b, then C3b is inactivated by Factor I and Factor H to form the C3c and C3d fragments. The interaction of zinc with C3 was quantified using analytical ultracentrifugation and x-ray scattering. C3, C3u, and C3b associated strongly in >100 μm zinc, whereas C3c and C3d showed weak association. With zinc, C3 forms soluble oligomers, whereas C3u and C3b precipitate. We conclude that the C3, C3u, and C3b association with zinc depended on the relative positions of C3d and C3c in each protein. Computational predictions showed that putative weak zinc binding sites with different capacities exist in all five proteins, in agreement with experiments. Factor H forms large oligomers in >10 μm zinc. In contrast to C3b or Factor H alone, the solubility of the central C3b-Factor H complex was much reduced at 60 μm zinc and even more so at >100 μm zinc. The removal of the C3b-Factor H complex by zinc explains the reduced C3u/C3b inactivation rates by zinc. Zinc-induced precipitation may contribute to the initial development of sub-retinal pigment epithelial deposits in the retina as well as reducing the progression to advanced age-related macular degeneration in higher risk patients.  相似文献

Background

Psychosocial stress and activation of neutrophil granulocytes are increasingly recognized as major risk factors of coronary artery disease (CAD), but the possible relationship of these two factors in CAD patients is largely unexplored. Activation of neutrophils was reported to be associated with stenting; however, the issue of neutrophil state in connection with percutaneous coronary intervention (PCI) is incompletely understood from the aspect of stress and its hypothalamic-pituitary-adrenal axis (HPA) background. Thus, we aimed to study cortisol- and ACTH-associated changes in granulocyte activation in patients undergoing PCI.

Methodology/Principal Findings

Blood samples of 21 stable angina pectoris (SAP) and 20 acute coronary syndrome (ACS) patients were collected directly before (pre-PCI), after (post-PCI) and on the following day of PCI (1d-PCI). Granulocyte surface L-selectin, CD15 and (neutrophil-specific) lactoferrin were analysed by flow cytometry. Plasma cortisol, ACTH, and lactoferrin, IL-6 were also assayed. In both groups, pre- and post-PCI ratios of lactoferrin-bearing neutrophils were relatively high, these percentages decreased substantially next day; similarly, 1d-PCI plasma lactoferrin was about half of the post-PCI value (all p≤0.0001). Post-PCI ACTH was reduced markedly next day, especially in ACS group (SAP: p<0.01, ACS: p≤0.0001). In ACS, elevated pre-PCI cortisol decreased considerably a day after stenting (p<0.01); in pre-PCI samples, cortisol correlated with plasma lactoferrin (r∼0.5, p<0.05). In 1d-PCI samples of both groups, ACTH showed negative associations with the ratio of lactoferrin-bearing neutrophils (SAP: r = −0.601, p<0.005; ACS: r = −0.541, p<0.05) and with plasma lactoferrin (SAP: r = −0.435, p<0.05; ACS: r = −0.609, p<0.005).

Conclusions/Significance

Pre- and post-PCI states were associated with increased percentage of activated/degranulated neutrophils indicated by elevated lactoferrin parameters, the 1d-PCI declines of which were associated with plasma ACTH in both groups. The correlation of plasma cortisol with plasma lactoferrin in the extremely stressed ACS before stenting, however, suggests an association of cortisol with neutrophil activation.  相似文献

Proteomics research revealed the impressive complexity of eukaryotic proteomes in unprecedented detail. It is now a commonly accepted notion that proteins in cells mostly exist not as isolated entities but exert their biological activity in association with many other proteins, in humans ten or more, forming assembly lines in the cell for most if not all vital functions.1,2 Knowledge of the function and architecture of these multiprotein assemblies requires their provision in superior quality and sufficient quantity for detailed analysis. The paucity of many protein complexes in cells, in particular in eukaryotes, prohibits their extraction from native sources, and necessitates recombinant production. The baculovirus expression vector system (BEVS) has proven to be particularly useful for producing eukaryotic proteins, the activity of which often relies on post-translational processing that other commonly used expression systems often cannot support.3 BEVS use a recombinant baculovirus into which the gene of interest was inserted to infect insect cell cultures which in turn produce the protein of choice. MultiBac is a BEVS that has been particularly tailored for the production of eukaryotic protein complexes that contain many subunits.4 A vital prerequisite for efficient production of proteins and their complexes are robust protocols for all steps involved in an expression experiment that ideally can be implemented as standard operating procedures (SOPs) and followed also by non-specialist users with comparative ease. The MultiBac platform at the European Molecular Biology Laboratory (EMBL) uses SOPs for all steps involved in a multiprotein complex expression experiment, starting from insertion of the genes into an engineered baculoviral genome optimized for heterologous protein production properties to small-scale analysis of the protein specimens produced.5-8 The platform is installed in an open-access mode at EMBL Grenoble and has supported many scientists from academia and industry to accelerate protein complex research projects.  相似文献
