Eighteen filamentous fungi and six actinomycetes species were screened for their ability to metabolize bisoprolol, a \-blocking drug. All strains of Cunninghamella tested accumulated metabolite M4 = EMD 46193 ([4-(2-hydroxy-3-isopropylaminopropoxy)benzyloxy]ethanol). Among the strains investigated only Gliocladium deliquescens excreted the corresponding carbonic acid M1 = EMD 44025 into the culture medium. Biotransformation of bisoprolol by fungi occurred only during growth in complex medium or with resting cells after cultivation in complex medium. Cultivation of the climbing plant Dioscorea zingiberensis at a light intensity of 100 microE. m(-2) sec(-1) yields three different phenotypes. Most of the plants grow as green phenotype (DzW). Two further forms differ in their leaf shape and leaf color. Whereas one type exhibits a more pointed leaf shape in the upper part of the plant with leaves appearing yellow-green with white stripes or hatchings (DzY), the other type shows a more round leaf shape with an intensive yellow-green color (DzT). These three plant types differ in their diosgenin content not only in their rhizomes but also in the chloroplasts. In the rhizomes the diosgenin content in the green form is 0.4%, in the DzY-form 0.6% and in the DzT-form even 1.3% of the dry weight. Furthermore, even in chloroplasts of the green DzW-form and of the DzY-form the presence of diosgenin was demonstrated. It occurs there as the epimeric form yamogenin. The DzT-form contains no yamogenin in its chloroplasts. Besides this, these plant forms differ in their chlorophyll and carotenoid content and in their fatty acid composition. Carotenoids increase from 1.3% of total lipids in the green phenotype to 3.3% in the DzY- and to 4.2% in the DzT-form. This increase refers to beta-carotene as well as to lutein and neoxanthin. The chlorophyll content in the green type is 8.1% and lower in the DzY-form with 7%. The highest chlorophyll content is found in the DzT-form with 12%. Fatty acids in the DzY-form and in the DzT-form have a more unsaturated character than in the green phenotype. The content of the monoenoic acid trans-hexadecenoic acid is considerably lower in both phenotypes when compared to the green phenotype. In both phenotypes the quantity of fatty acids with 16 carbon atoms is reduced, whereas fatty acids with 18 carbon atoms occur in higher concentration. Cultivation of the green phenotype (DzW) at the three light intensities of 10, 100 and 270 microE x m(-2) x sec(-1) leads to changes of the diosgenin content in rhizomes, to an increase of leaf dry weight, to a reduction of the grana structure in chloroplasts and therewith to a decrease of the chlorophyll content. The total lipid content is highest under the cultivation at 100 microE x m(-2) x sec(-1) and reduced by 30% at 10 and 270 microE x m(-2) x sec(-1). Carotenoids, however, are highest in shaded plants (10 microE x m(-2) x sec(-1)) and plants grown under high light conditions of 270 microE x m(-2) x sec(-1). At 100 microE x m(-2) x sec(-1) a decrease of saturated fatty acids is observed in comparison to plants grown under shaded conditions.  相似文献

We analysed chloroplast lipids of Nicotiana tabacum var. John William's Broadleaf, cultivated under an increased PCO(2) of 700 p.p.m. Glycolipids and phospholipids remain constant under these conditions, whereas the carotenoid content undergoes a quantitative change. The saturation degree of fatty acids increases due to an increase in palmitic acid and decreases in hexadecatrienoic acid and linolenic acid.  相似文献

The localization of protochorophyllide (Pchlide) and of NADPH-protochlorophyllide oxidoreductase (POR, EC 1.6.99.1) within (etio)chloroplasts has been investigated at selected stages of greening of barley seedlings. Pchlide pigment and POR protein contents were evaluated in different plastid membrane fractions by fluorescence spectroscopy and immunoblot analysis using a monospecific polyclonal antibody raised against the purified enzyme. Fluorescence analysis showed the presence of Pchlide in both the envelope and thylakoid membranes. During greening, the Pchlide content, expressed on a total protein basis, decreased in thylakoid membranes, whereas it increased in the envelope membranes. POR proteins were detected mainly in thylakoid membranes at early greening stages. In contrast, the weak amount of POR proteins was associated more specifically with envelope membranes of mature chloroplasts. Whatever the greening stage, thylakoid-bound Pchlide and POR proteins were more abundant in the thylakoid regions which remained unsolubilized after mild Triton treatment used as standard procedure to prepare PS II particles. This suggests the preferential association of Pchlide and POR to the appressed regions of thylakoids. We have found that in petroleum-ether extracted tobacco thylakoids, plastoquinone A (PQ-A) and plastoquinone C (PQ-C) had similar efficiency in restoration of oxygen-evolving activity, while plastoquinone B (PQ-B), which is a fatty acid ester of PQ-C, was about 50% less effective. This indicates that apart from PQ-A, PQ-C and to a smaller extent PQ-B may function as electron acceptors of Photosystem II (PS II). The DCMU inhibition curves for PQ-C and PQ-B were biphasic and an initial slow decline was followed by a sharp decrease in oxygen evolution yield with a 50% inhibition (I50) at 0.25 M DCMU. In the case of PQ-A (I50 = 0.20 M DCMU), the activity decreased gradually without the sharp transition. The corresponding inhibition curve for unextracted thylakoids, where all the native prenylquinones are present, shows an intermediate shape between PQ-A and PQ-C but with a higher I50, equal to 0.32 M, suggesting that the contribution of PQ-C as an electron acceptor of Photosystem II might be significant in thylakoid membranes with natural prenyllipid composition. -Tocopherol quinone showed no activity in the restoration of oxygen evolution in extracted thylakoids, indicating that it cannot accept electrons from PS II. The fatty acid composition of PQ-B isolated from maple leaves showed a high degree of saturated fatty acids like myristic and palmitic acid, and its unique composition indicates that it is a natural component of the thylakoid membrane.  相似文献

Zusammenfassung 1. Es wird eine Methode zur Herstellung, Montierung und Färbung von epidermalen Häutchenpräparaten zur dreidimensionalen Analyse des Epidermis-Aufbaues beschrieben. Eine Kombination mit der Histoautoradiographie ist bedingt möglich. Damit ist eine Basis für weitere Untersuchungen mit neuer Fragestellung geschaffen. 2. Die untersuchten Objekte Mäuseohr und Rückenhaut zeigen eine bisher unbekannte, auffallende Musterbildung mit Zuordnung bestimmter interfollikulärer basaler Gruppen zu Zellen des Stratum spinosum und des Stratum corneum. Es handelt sich um rosettenähnliche Gebilde aus mehreren peripheren Basalzellen und meist einer zentralen Spinosumzelle. 3. Die Bedeutung der Befunde für die normale wie für die pathologische Regeneration und Neoplasie der Epidermis ist offen. Es werden Beziehungen zum Regenerationsmodus und damit zur Proliferationskinetik der Epidermis diskutiert. In diesem Zusammenhang sind die Untersuchungen auch für die Humanpathologie relevant.

Summary 1. A method for the preparation, mounting, and staining of thin preparations of mouse epidermis is described, which allows a three-dimensional analysis of epidermal structure. A limited combination of this technique with histo-autoradiography is possible, thereby presenting a broader basis for further investigations. 2. Both ear and back skin show a striking, previously unrecognized pattern of organization, in which definite interfollicular basal cell groups are oriented towards cells of the stratum spinosum and stratum corneum. A rosettelike picture of many peripheral basal cells and, in most instances, a central spinosum cell is found. 3. The significance of our findings for normal and pathological regenerations as well as neoplasia of the epidermis remains open. We discuss relationships to the mode of regeneration and thus the kinetics of proliferation in the epidermis. Our investigations are also relevant for human pathology in this regard.

The kernels of Trichosanthes kirilowii seeds contain a green oil which makes up for 62% of their dry matter. This oil consists up to 95% of triglycerides, 2% of glycolipids, 1.3% of phospholipids and 1.8% of chlorophylls. As fatty acid components the triglycerides, glycolipids and phospholipids contain the unsaturated fatty acids linoleic and oleic acid and the saturated palmitic acid. In the triglycerides 19% of the C18:3 acid occur with the configuration delta9 cis, delta11 trans, delta13 cis. This acid is called trichosanic acid and is absent in glycolipids and phospholipids which contain instead another C18:3 fatty acid, which has conjugated double bounds and occurs with an amount of 21% and 3%, respectively. Typically, these oil seeds contain in addition up to 30% of their dry matter proteins and up to 2.5% mono- and oligosaccharides. The monosaccharides consist of rhamnose, galactose and glucose and the oligosaccharides represent a mixture of tri- and tetrasaccharides.  相似文献
