19-hydroxycoronaridin und 19-hydroxyibogamin,zwei antibiotisch wirksame alkaloide vom ibogamin-typ

During the separation of an extract from Tabernaemontana glandulosa, small amounts of a new alkaloid were obtained for which the structure 19-hydroxycoronaridine was deduced. This new alkaloid and 19-hydroxyibogamine, which can be obtained from it, show marked antibiotic activity in the agar plate diffusion test.     

Empirical examination of the threshold model of neuron firing

An elementary model of neuronal activity involves temporal and spatial summation of postsynaptic currents that are elicited by presynaptic spikes and that, in turn, elicit postsynaptic potentials at a trigger zone; when the potential at the trigger zone exceeds a threshold level, a postsynaptic spike is generated. This paper describes three methods of estimating the summation function, that is, the function of time that converts the synaptic current into potential at the trigger zone: namely, maximum likelihood, cross-correlation analysis and cross-spectral analysis. All three methods, when applied to input-output data collected on various neurons of Aplysia californica, give comparable results. As estimated, the summation function involved in the explored cells has an early positive-going swing that is large and brief. The natural variability of vital rates and associated statistics

The first concern of this work is the development of approximations to the distributions of crude mortality rates, age-specific mortality rates, age-standardized rates, standardized mortality ratios, and the like for the case of a closed population or period study. It is found that assuming Poisson birthtimes and independent lifetimes implies that the number of deaths and the corresponding midyear population have a bivariate Poisson distribution. The Lexis diagram is seen to make direct use of the result. It is suggested that in a variety of cases, it will be satisfactory to approximate the distribution of the number of deaths given the population size, by a Poisson with mean proportional to the population size. It is further suggested that situations in which explanatory variables are present may be modelled via a doubly stochastic Poisson distribution for the number of deaths, with mean proportional to the population size and an exponential function of a linear combination of the explanatories. Such a model is fit to mortality data for Canadian females classified by age and year. A dynamic variant of the model is further fit to the time series of total female deaths alone by year. The models with extra-Poisson variation are found to lead to substantially improved fits.     

Recommended protocols based on a survey of current practice in genotoxicity testing laboratories, IV. Chromosome aberration and sister-chromatid exchange in Chinese hamster ovary, V79 Chinese hamster lung and human lymphocyte cultures

A recommended protocol has been developed for chromosomal aberration and sister-chromatid exchange assays in CHO, V79 and human lymphocyte cultures. The protocol was based on the responses to a detailed questionnaire completed by North-American and European governmental, university, and contract laboratories using these tests. This report identifies those modifications to previously described methods that are used on a regular basis and clarifies confusing or inconsistent practices. These protocols can be modified for use in other types of cells.     

Metabolic products of microorganisms

In the course of a screening for new metabolites from fungi we isolated a substance with antimicrobial activity from cultures of Aspergillus duricaulis (CBS 481.65) (Tü 679). It was antagonized by putrescine, spermidine, spermine, arginine, citrulline, lysine, ornithine, in higher concentration by asparagine and glutamine too. The effect of ethylenediaminetetraacetate on the susceptibility of Streptomyces viridochromogenes (Tü 57) and Bacillus subtilis ATCC 6051 to this antibiotic has been studied.The substance was characterized and identified as cyclopaldic acid.Abbreviations MIC minimum inhibitory concentration - tlc thinlayer chromatography Metabolic products of microorganisms. 166. Chitin synthase from Coprinus cinereus (Schaeff. ex Fr.) S. F. Gray (= C. lagopus sensu Buller) was used as a model for chitin synthase from insects. The effect of dimilin (difluorobenzuron), captan (trichloromethylsulfonyl fungicide), kitazin P (organophosphorus ester fungicide) and parathion (organophosphorus insecticide) on the fungal enzyme was compared with the effect of nikkomycin (nucleoside-peptide antibiotic).Metabolic products of microorganisms. 180. Recommended protocols based on a survey of current practice in genotoxicity testing laboratories: III. Cell transformation in C3H/10T1/2 mouse embryo cell, BALB/c 3T3 mouse fibroblast and Syrian hamster embryo cell cultures

A standardized protocol and guidelines for the performance of cell transformation testing in mouse embryo (C3H/10T1/2), mouse fibroblast (BALB/c 3T3) and Syrian hamster embryo (SHE) cells have been developed. The protocol is based primarily on current laboratory practices as determined by responses to a detailed questionnaire completed by North American and European governmental, university and contract laboratories involved with cell transformation experimentation. This report identifies those modifications to previously described methodologies which are being used on a regular basis and also serves to clarify confusing or inconsistent practices.     

Recommended protocols based on a survey of current practice in genotoxicity testing laboratories: I. Unscheduled DNA synthesis assay in rat hepatocyte cultures

A protocol based primarily on current laboratory practices in the performance of the unscheduled DNA synthesis (UDS) assay with primary rat hepatocyte cultures has been developed. These guidelines were developed using tabulated responses to a detailed questionnaire completed by North American and European governmental, university and contract laboratories involved with the UDS test. This report identifies those modifications to previously described methodologies which are used on a regular basis and also serves to clarify confusing or inconsistent practices. Although this protocol pertains specifically to the use of primary rat hepatocyte cultures, it can be modified to incorporate other types of cells in which certain aspects remain the same.     

A note on the estimation of evoked response

The traditional average evoked response is compared and contrasted with several alternate estimates, derived from frequency domain considerations, for a model of evoked responses superimposed upon a stationary noise series. Further, a means of constructing approximate confidence intervals for the values of an evoked response is indicated.
