Abstract Larvae of the tobacco hornworm moth Manduca sexta starved for the first 3 days of the last (fifth) stadium undergo a supernumerary moult. If they are provided with sucrose during the starvation period, they develop into normal pupae although pupation is delayed. The activities of the corpora allata (CA) from normal, starved, and sucrose fed larvae were followed through the fifth stadium with a radiochemical assay for Juvenile Hormone (JH) biosynthesis. An attempt was made to correlate CA-activity with CA cell number, size, and protein content.
In CA of normally fed larvae the rate of JH synthesis declined to undetectable levels by day 4 which was also the time of exposure of the dorsal vessel. In CA of starved larvae, the rate of JH synthesis at first decreased but began to increase on day 3 and reached a peak value by day 7 , at which time head capsule slippage occurred. In CA of sucrose fed larvae, the rate of biosynthesis declined as in normal larvae but the decline was extended over a longer period. Exposure of the dorsal vessel was delayed in the same manner and occurred on days 7–9. The major JH in all cases was JH-II.
The CA comprise c. 150 cells in the early fifth stadium, and this number remained constant during the fifth stadium in all three feeding regimens. In normal larvae, CA size and protein content increased several-fold during the stadium whereas in starved and sucrose-fed larvae they increased slowly and in agreement with the altered timing of developmental events. In none of the groups was the CA activity pattern correlated with morphometric changes of the CA. The rates of JH biosynthesis were not closely correlated with published JH titre curves. The in vivo mechanisms for regulation of JH production remain to be elucidated.  相似文献

2, 4-Dichlorophenoxyacetic acid caused a shortening of rootsand shoots when mature seeds of Sorghum bicolor (L.) Moenchcv. BT3197 were germinated on Murashige and Skoog (MS) mediumthat contained 2,4-D. Shoot growth was restored with cytokinins.A callus formed at the nodal region, the further differentiationof which was determined by the ratio of 2,4-D and cytokininsin the initial culture medium. A high auxin to cytokinin ratiopromoted primarily root differentiation while a high cytokininto auxin ratio promoted multiple bud development. Isolated shootapical meristem with the subtending node produced embryogeniccallus at low cytokinin levels and green buds on high cytokininlevels when cultured in the presence of 2,4-D. It is concludedthat cells potentially capable of differentiation into roots,somatic embryos or axillary buds are present in the first nodalregion. The outcome of competition between Campoletis chlorideae Uchida and Eriborus argenteopilosus (Cameron) (Hymenoptera: Ichneumonidae), two indigenous larval parasitoids of the noctuid pest, Spodoptera litura (Fabricius), was investigated in the laboratory by way of two experiments. In the individual exposure experiment, the host larvae were exposed to the parasitoids either alone or one after the other at different time intervals and were considered to be parasitised when the parasitoid was observed to sting the host larva. When they stung the host larva singly, the parasitism rates by C. chlorideae and E. argenteopilosus was either similar to or higher than the parasitism recorded by each parasitoid in the different combination/interaction treatments. C. chlorideae cocoons were formed from majority of the host larvae, which were stung by both parasitoid species, one after the other, in most oviposition orders and delays between ovipositions. E. argenteopilosus appeared to be the dominant parasitoid only in those treatments where it was the first one to parasitise and the delay in C. chlorideae parasitism was more than 18 h. and it never dominated the interactions, when it was the second one to parasitise. This implied that C. chlorideae had an almost consistent advantage over E. argenteopilosus. In the mass exposure experiment, the two parasitoid species (either alone or together) were allowed to forage and parasitise the host larvae in cages. When the two parasitoid species were allowed to forage in the same host patch simultaneously, the performance of C. chlorideae when it was alone was statistically similar to its performance in the presence of E. argenteopilosus. However, E. argenteopilosus performed better when it could forage alone. The results of both experiments revealed that C. chlorideae was the more competitive of the two species.  相似文献

This paper deals with the effects of benzamide, a chromosomal RNA inhibitor, on embryonic development of the housefly Musca domestica nebulo. Eggs exposed to benzamide immediately after oviposition continued to develop until the blastema stage, but further development was arrested. Formation of cell boundaries and nucleoli failed to occur and the nuclei at the posterior pole did not differentiate into pole cells. This suggests that synthesis of new RNA is needed for development beyond the blastema stage. Treatment of eggs at different stages of development showed that as development progressed embryos became less sensitive to the drug. Introduction of benzamide into eggs during the pre-blastema period caused irreversible arrest of development, whereas, treatment during post-blastema stages resulted in reversible inhibition of development. The cortical cytoplasm thus appears to induce a significant change in the cleavage nuclei, which alters their sensitivity to the drug.  相似文献

Abstract. The role of Juvenile Hormone (JH) during reproductive development and diapause was investigated in the boll weevil, Anthonomus grandis Boheman (Coleoptera: Curculionidae). JH sythesized by corpora allata (CA) in vitro of A.grandis was identified as JH-UJ by high-performance liquid chromatography and by conversion to the methoxyhydrin. Optimal conditions for the use of a short-term assay in vitro were established to examine profiles of CA activity. In addition, rates of JH degradation by JH-specific esterase were determined. Patterns of CA and JH-esterase activity during reproductive development and the diapause state were established with laboratory-reared reproductive weevils and diapausing weevils collected as larvae and pupae in the field after the cotton-growing season. The results indicate that JH production is elevated in reproductive females whereas males and winter field-collected females show no CA activity. Vitellogenin concentrations in haemolymph and rates of oviposition were studied in relation to CA activity and JH degradation. An attempt to induce diapause in the laboratory failed.  相似文献

Haploid plants were generated from anther-culture of Nicotianatabacum cv. Burley Ky 14. Plants that developed from pollenwithout an intervening callus stage were grown to maturity.Ploidy of the anther-derived plants were verified by chromosomecounts in root tip squashes. Crystalline ribulose 1,5-bisphosphatecarboxylase was purified from leaves of mature haploid plantsgrown in a growth chamber. Crystals appeared to be similar tothose from the diploid parental line.
