Photoactivation of Oxygen-evolving System in Dark-grown Spruce Seedlings   总被引:4,自引:0,他引:4  
Plastids prepared from dark-grown spruce seedlings showed a negligible activity of photosystem II, and no fluorescence variation was observed during actinic illumination. The photosystem II reaction centre, however, was present in primary thylakoids. Exposure of such seedlings to continuous light induced the development of photosystem II activity via three stages (rapid, lagged and gradual), and the variable fluorescence appeared. The rapid development of photosystem II may be attributed to the activation of the oxygen-evolving system, possibly the manganese-catalyzing site, and the lagged and gradual developments may be closely related to the formation of thylakoid membranes and their assembly to grana.  相似文献

The Hill reaction of chloroplasts was inhibited by washing themwith 0.8 M Tris buffer. This inhibition was further promotedby adding ferricyanide in the washing medium. When a reducingreagent, such as the 2,6-dichlorophenol indophenol (DCPIP)-ascorbatesystem or the hydroquinone (HQJ-ascorbate system, had been addedto the Tris buffer, Hill reaction activity was unaffected. Hill reaction activity of Tris-washed chloroplasts recoveredup to 70% of the initial level by re-washing the chloroplastswith a preparation medium containing theabove reducing reagents. Photobleaching of carotenoid and chlorophyll is characteristicof Tris-washed chloroplasts. However, reactivated chloroplastsshowed no photobleaching as in the case with intact chloroplasts. The latent O2-evolving center in chloroplasts isolated from spruce [Picea abies (L.) Karst.] seedlings grown in the dark was readily activated by pre-illuminating the chloroplast suspension with weak white light. The photoactivation depended on pH with the optimum at pH 7–8, and was strongly stimulated by ascorbic acid. The optimal stimulation was also obtained at pH 7–8. The temperature dependence of the photoactivation suggested the involvement of some dark reaction in the activation process.  相似文献
