Long-term assessments of species assemblages are valuable tools for detecting species ecological preferences and their dispersal tracks, as well as for assessing the possible effects of alien species on native communities. Here we report a 50-year-long study on population dynamics of the four species of land flatworms (Platyhelminthes, Tricladida, Terricola) that have colonized or become extinct in a 70-year-old Atlantic Forest regrowth remnant through the period 1955–2006. On the one hand, the two initially most abundant species, which are native to the study site, Notogynaphallia ernesti and Geoplana multicolor have declined over decades and at present do not exist in the forest remnant. The extinction of these species is most likely related with their preference for open vegetation areas, which presently do not exist in the forest remnant. On the other hand, the neotropical Geoplaninae 1 and the exotic Endeavouria septemlineata were detected in the forest only very recently. The long-term study allowed us to conclude that Geoplaninae 1 was introduced into the study area, although it is only known from the study site. Endeavouria septemlineata, an active predator of the exotic giant African snail, is originally known from Hawaii. This land flatworm species was observed repeatedly in Brazilian anthropogenic areas, and this is the first report of the species in relatively well preserved native forest, which may be evidence of an ongoing adaptive process. Monitoring of its geographic spread and its ecological role would be a good practice for preventing potential damaging effects, since it also feeds on native mollusk fauna, as we observed in lab conditions. The effect of environmental factors on the post-illumination burst of CO2 (PIB) and O2 inhibition of apparent photosynthesis (APS) in wheat (Triticum aestivum L.) was studied in an open gas exchange system utilizing the mathematics of non-steady-state systems. Two components of inhibition by O2 are suggested: one is caused by photorespiration as measured from the maximum rate of the PIB, and the second is direct inhibition as taken as APS2%O2— (APSx%O2+ PIBx%O2) where X is the oxygen concentration. A primary PIB which occurred from 16–28 s after the darkening of the foliage was attributed to photorespiration. No primary PIB was observed at 2% O2. At a CO2 concentration of 100 μ/1 in the atmosphere (about 2.5 μM based on leaf intercellular concentration) and at 30°C and 145 nE/cm2 nE/cm2·s, APS decreased curve-linearly with increasing O2 and reached an O2 compensation point of 560 μM (48% by volume), above which there was a net loss of CO2 in the light. The PIB increased with increasing O2 and became saturated at about 500 μM O2 but decreased above 900 μM O2. Direct inhibition of photosynthesis by O2 increased with increasing O2 concentration. Decreasing CO2 concentration had an effect on the magnitude of the PIB similar to that of increasing O2. At 30°C and 21% O2, the PIB increased with decreasing CO2 down to the CO2 compensation point (I) of 1.4 μM (47 μM/l). Below Γ, both PIB and CO2 evolution into the air in the light (at 21% O2) increased and then decreased at CO2 below 0.8 μM. The ratio of the PIB to APS2% o O2 increased linearly with increasing O2/CO2 ratio where O2 was held constant at 21% and CO2 was varied from 1.4 to 8.5 μM, while direct inhibition of photosynthesis expressed as a proportion of APS2%O2 remained constant over this range. At low CO2 concentration photorespiration as estimated by the PIB is the major part of O2 photosynthesis, while at atmospheric CO2 levels, direct inhibition is the major component. The PIB and APS at 2% and 21% O2 increased hyperbolically with increasing irradiance and all became light-saturated at about 65 nE/cm2 s. The percentage total O2 inhibition of photosynthesis remained constant with increasing irradiance as did the relative contribution of direct O2 inhibition or photorespiration (PIB) to total O2 inhibition. The PIB and APS at 21% O2 had similar temperature optima of 30°C when experimental conditions were adjusted to provide a constant internal O2/CO2 solubility ratio at varying temperatures. However, with a constant external CO2 concentration, the temperature optimum for the PIB shifted upward to 35°C while that for APS at 21% O2 remained at 30°C, which may be due to an increased O2/CO2 concentration in the leaf with increasing temperature.  相似文献

The acyl carrier protein (ACP), an essential protein cofactor for fatty acid synthesis, has been isolated from two cyanobacteria: the filamentous, heterocystous, Anabaena variabilis (ATCC 29211) and the unicellular Synechocystis 6803 (ATCC 27184). Both ACPs have been purified to homogeneity utilizing a three-column procedure. Synechocystis 6803 ACP was purified 1800-fold with 67% yield, while A. variabilis ACP was purified 1040-fold with 50% yield. Yields of 13.0 micrograms ACP/g Synechocystis 6803 and 9.0 micrograms ACP/g A. variabilis were achieved. Amino acid analysis indicated that these ACPs were highly charged acidic proteins similar to other known ACPs. Sequence analysis revealed that both cyanobacterial ACPs were highly conserved with both spinach and Escherichia coli ACP at the phosphopantetheine prosthetic group region. Examining the probability of alpha-helix and beta-turn regions in various ACPs, showed that cyanobacterial ACPs were more closely related to E. coli ACP than spinach ACP I. Immunoblot analysis and a competitive binding assay for ACP illustrated that both ACPs bound poorly to spinach ACP I antibody. SDS/PAGE and native PAGE of Synechocystis 6803 ACP and A. variabilis ACP showed that cyanobacteria ACPs co-migrated with E. coli ACP and had relative molecular masses of 18,100 and 17,900 respectively. Both native and urea gel analysis of acyl-ACP products from fatty acid synthase reactions demonstrated that bacterial ACPs and plant ACP gave essentially the same metabolic products when assayed using either bacterial or plant fatty acid synthase. A. variabilis and Synechocystis 6803 ACP could be acylated using E. coli acyl ACP synthetase.  相似文献

Miniplasmids with the P1 copN22 mutation have a copy number about seven times that of the wild type. Selection for reduced copy number from this plasmid led to the isolation of second-site pseudorevertants, called poc mutants. DNA sequence analysis showed that all six independent poc mutants have a single base change in the same codon of the repA gene. This implicates the amino acid at this location, either directly or indirectly, in interactions important for copy number control.  相似文献

Saccharomyces cerevisiae has recently been engineered to use acetate, a primary inhibitor in lignocellulosic hydrolysates, as a cosubstrate during anaerobic ethanolic fermentation. However, the original metabolic pathway devised to convert acetate to ethanol uses NADH-specific acetylating acetaldehyde dehydrogenase and alcohol dehydrogenase and quickly becomes constrained by limited NADH availability, even when glycerol formation is abolished. We present alcohol dehydrogenase as a novel target for anaerobic redox engineering of S. cerevisiae. Introduction of an NADPH-specific alcohol dehydrogenase (NADPH-ADH) not only reduces the NADH demand of the acetate-to-ethanol pathway but also allows the cell to effectively exchange NADPH for NADH during sugar fermentation. Unlike NADH, NADPH can be freely generated under anoxic conditions, via the oxidative pentose phosphate pathway. We show that an industrial bioethanol strain engineered with the original pathway (expressing acetylating acetaldehyde dehydrogenase from Bifidobacterium adolescentis and with deletions of glycerol-3-phosphate dehydrogenase genes GPD1 and GPD2) consumed 1.9 g liter−1 acetate during fermentation of 114 g liter−1 glucose. Combined with a decrease in glycerol production from 4.0 to 0.1 g liter−1, this increased the ethanol yield by 4% over that for the wild type. We provide evidence that acetate consumption in this strain is indeed limited by NADH availability. By introducing an NADPH-ADH from Entamoeba histolytica and with overexpression of ACS2 and ZWF1, we increased acetate consumption to 5.3 g liter−1 and raised the ethanol yield to 7% above the wild-type level.  相似文献

Artificial reefs continue to be added as habitat throughout the world, yet questions remain about how reef design affects fish diversity and abundance. In the present study, the effects of reef density were assessed for fish communities and sizes of economically valuable Lutjanus campechanus 13 km off Port Mansfield, Texas, at a reef composed of more than 4000 concrete culverts. The study spanned from May to June in 2013 and 2014, and sites sampled included natural reefs, bare areas, and varying culvert patch density categories, ranging from 1–190 culverts. Abundances of adults and species evenness of juvenile populations differed between the years. Fish communities did not significantly differ among density categories; however, highest species richness and total abundances were observed at intermediate culvert densities and at natural reefs. Whereas the abundance of L. campechanus did not differ among density categories, mean total lengths of L. campechanus were greatest at the lower density. Our findings suggest that reefs should be deployed with intermediate patch density of 71–120 culverts in a 30-m radius to yield the highest fish abundances.  相似文献
