The concept of "stimulus-secretion coupling" suggested by Douglas and co-workers to explain the events related to monamine discharge by the adrenal medulla (5, 7) may be applied to other endocrine tissues, such as adrenal cortex (36), pancreatic islets (4), and magnocellular hypothalamic neurons (6), which exhibit a similar ion-dependent process of hormone elaboration. In addition, they share another feature, that of joining neighbor cells via membrane junctions (12, 26, and Fletcher, unpublished observation). Given this, and the reports that hormone secretion by the pars distalis also involves a secretagogue-induced decrease in membrane bioelectric potential accompanied by a rise in cellular [Ca++] (27, 34, 41), it was appropriate to test the possibility that cells of the anterior pituitary gland are united by junctions.  相似文献

The purpose of this experiment was to investigate how early lactating cows adjust their metabolism and production to acute, but moderate changes in the energy density of the diet. Sixty dairy cows were randomly assigned to one of four treatments: two change-over groups (HNH and NHN) and two control groups (HHH and NNN), where H and N refer to a high and normal energy density in the total mixed ration (TMR), respectively. The experimental period covered the first 9 weeks post calving, which was split up in three 3-week periods. Thus, cows assigned to HNH or NHN shifted TMR in weeks 4 and 7 after calving while cows assigned to HHH or NNN were fed the same TMR for all 9 weeks. Results from cows on treatment HNH were compared with group HHH while cows on treatment NHN were compared with group NNN. When the diet changed from N to H and H to N, cows increased and decreased their dry-matter intake (DMI), respectively compared with control groups. Cows adjusted milk yield accordingly to changes in DMI, although not always significantly. Energy-corrected milk yield was not significantly affected by any of the changes in the energy density of the diet but generally showed same tendencies as milk yield. Non-esterified fatty acids (NEFA), beta-hydroxybutyrate in blood and milk and triacylglycerol and glycogen content in the liver were not significantly affected by changes in the energy density of the diet, except from NEFA at one change. Glucose increased more when the diet changed from N to H and increased less when the diet changed from H to N, compared with control groups, although not always significantly. Collectively, these results suggest that cows adjust their DMI and partly milk yield according to the energy density of the diet and therefore only limited effects were observed in physiological parameters.  相似文献

Intramammary infusion of lipopolysaccharide (LPS) in cows induces udder inflammation that partly simulates mastitis caused by infection with Gram‐negative bacteria. We have used this animal model to characterize the quantitiative response in the milk proteome during the time course before and immediately after the LPS challenge. Milk samples from three healthy cows collected 3 h before the LPS challenge were compared with milk samples collected 4 and 7 h after the LPS challenge, making it possible to describe the inflammatory response of individual cows. Quantitative protein profiles were obtained for 80 milk proteins, of which 49 profiles changed significantly for the three cows during LPS challenge. New information obtained in this study includes the quantified increase of apolipoproteins and other anti‐inflammatory proteins in milk, which are important for the cow's ability to balance the immune response, and the upregulation of both complement C3 and C4 indicates that more than one complement pathway could be activated during LPS‐induced mastitis. In the future, this analytical approach may provide valuable information about the differences in the ability of individual cows to resist and recover from mastitis.  相似文献

Background

Population movements along the Thailand-Cambodia border, particularly among highly mobile and hard-to-access migrant groups from Cambodia and Myanmar, are assumed to play a key role in the spread of artemisinin resistance. Data on treatment-seeking behaviours, knowledge and perceptions about malaria, and use of preventive measures is lacking as characteristics of this population prevent them from being represented in routine surveillance and the lack of a sampling frame makes reliable surveys challenging.

Methods

A survey of migrant populations from Cambodia and Myanmar was implemented in five selected rural locations in Thailand along the Thai-Cambodian border using respondent driven sampling (RDS) to determine demographic characteristics of the population, migratory patterns, knowledge about malaria, and health-care -seeking behaviours.

Results

The majority of migrants from Myanmar are long-term residents (98%) with no plans to move back to Myanmar, understand spoken Thai (77%) and can therefore benefit from health messages in Thai, have Thai health insurance (99%) and accessed public health services in Thailand (63%) for their last illness. In comparison, the majority of Cambodian migrants are short-term (72%). Of the short-term Cambodian migrants, 92% work in agriculture, 18% speak Thai, 3.4% have Thai health insurance, and the majority returned to Cambodia for treatment (45%), self-treated (11%), or did not seek treatment for their last illness (27%).

Conclusion

Most highly mobile migrants along the Thai-Cambodia border are not accessing health messages or health treatment in Thailand, increasing their risk of malaria and facilitating the spread of potentially resistant Plasmodium falciparum as they return to Cambodia to seek treatment. Reaching out to highly mobile migrants with health messaging they can understand and malaria diagnosis and treatment services they can access is imperative in the effort to contain the spread of artemisinin-resistant P. falciparum.  相似文献

Background and methods

Human metapneumovirus (hMPV) is a recently discovered respiratory virus associated with bronchiolitis, pneumonia, croup and exacerbations of asthma. Since respiratory viruses are frequently detected in patients with acute exacerbations of COPD (AE-COPD) it was our aim to investigate the frequency of hMPV detection in a prospective cohort of hospitalized patients with AE-COPD compared to patients with stable COPD and to smokers without by means of quantitative real-time RT-PCR.

Results

We analysed nasal lavage and induced sputum of 130 patients with AE-COPD, 65 patients with stable COPD and 34 smokers without COPD. HMPV was detected in 3/130 (2.3%) AE-COPD patients with a mean of 6.5 × 105 viral copies/ml in nasal lavage and 1.88 × 105 viral copies/ml in induced sputum. It was not found in patients with stable COPD or smokers without COPD.

Conclusion

HMPV is only found in a very small number of patients with AE-COPD. However it should be considered as a further possible viral trigger of AE-COPD because asymptomatic carriage is unlikely.  相似文献

This animal simulation model, named e-Cow, represents a single dairy cow at grazing. The model integrates algorithms from three previously published models: a model that predicts herbage dry matter (DM) intake by grazing dairy cows, a mammary gland model that predicts potential milk yield and a body lipid model that predicts genetically driven live weight (LW) and body condition score (BCS). Both nutritional and genetic drives are accounted for in the prediction of energy intake and its partitioning. The main inputs are herbage allowance (HA; kg DM offered/cow per day), metabolisable energy and NDF concentrations in herbage and supplements, supplements offered (kg DM/cow per day), type of pasture (ryegrass or lucerne), days in milk, days pregnant, lactation number, BCS and LW at calving, breed or strain of cow and genetic merit, that is, potential yields of milk, fat and protein. Separate equations are used to predict herbage intake, depending on the cutting heights at which HA is expressed. The e-Cow model is written in Visual Basic programming language within Microsoft ExcelR. The model predicts whole-lactation performance of dairy cows on a daily basis, and the main outputs are the daily and annual DM intake, milk yield and changes in BCS and LW. In the e-Cow model, neither herbage DM intake nor milk yield or LW change are needed as inputs; instead, they are predicted by the e-Cow model. The e-Cow model was validated against experimental data for Holstein–Friesian cows with both North American (NA) and New Zealand (NZ) genetics grazing ryegrass-based pastures, with or without supplementary feeding and for three complete lactations, divided into weekly periods. The model was able to predict animal performance with satisfactory accuracy, with concordance correlation coefficients of 0.81, 0.76 and 0.62 for herbage DM intake, milk yield and LW change, respectively. Simulations performed with the model showed that it is sensitive to genotype by feeding environment interactions. The e-Cow model tended to overestimate the milk yield of NA genotype cows at low milk yields, while it underestimated the milk yield of NZ genotype cows at high milk yields. The approach used to define the potential milk yield of the cow and equations used to predict herbage DM intake make the model applicable for predictions in countries with temperate pastures.  相似文献

In this paper, the effect of clinical symptoms of uterine inflammation on progesterone profile characteristics was quantified in dairy cows. A continuous scale based on visual observation of vaginal discharge (the previously developed D-index) was used to describe the clinical symptoms. Progesterone profiles in milk were used to describe the ovarian cycles, and to determine the distinguishing features of these profiles, a multivariate statistical procedure (principal component analysis) was performed.Significant negative effects of the D-index were seen during the first and second postpartum ovarian cycles. The D-index had a significant effect on the shape of progesterone profiles and the length of the ovarian cycles but it only accounted for a small proportion of the variation in these ovarian cycle features. The D-index was not a significant risk factor for the length of postpartum anovulatory period in the present study.  相似文献

Properties of the sliding disintegration response of demembranated tetrahymena cilia have been studied by measuring the spectrophotomeric response or turbidity of cilia suspensions at a wavelength of 350 nm relative to changes in the dynein substrate (MgATP(2-)) concentration. The maximum decrease in turbidity occurs in 20 muM ATP, and 90 percent of the decrease occurs in approximately 5.9 s. At lower ATP concentrations (1-20 muM), both the velocity and magnitude of the turbidity decreases are proportional to ATP concentration. The velocity data for 20 muM ATP permit construction of a reaction velocity curve suggesting that changes in turbidity are directly proportional to the extent and velocity of disintegration. At ATP concentrations more than 20 muM (50muM to 5mM), both velocity and magnitude of the turbidimetric response are reduced by approximately 50 percent. This apparent inhibition results in a biphasic response curve that may be related to activation of residual shear resistance or regulatory components at the higher ATP concentrations. The inhibitory effects of elevated ATP can be eliminated by mild trypsin proteolysis, whereupon the reaction goes to completion at any ATP concentration. The turbidimetric responses of the axoneme-substrate suspensions are consistent with the extent and type of axoneme disintegration revealed by electron microscope examination of the various suspensions, suggesting that the turbidimetric assay may prove to be a reliable means for assessing the state of axoneme integrity.  相似文献

Prediction of nutrient partitioning is a long-standing problem of animal nutrition that has still not been solved. Another substantial problem for nutritional science is how to incorporate genetic differences into nutritional models. These two problems are linked as their biological basis lies in the relative priorities of different life functions (growth, reproduction, health, etc.) and how they change both through time and in response to genetic selection. This paper presents recent developments in describing this biological basis and evidence in support of the concepts involved as they relate to nutrient partitioning. There is ample evidence that at different stages of the reproductive cycle various metabolic pathways, such as lipolysis and lipogenesis, are up or down regulated. The net result of such changes is that nutrients are channelled to differing extents to different organs, life functions and end-products. This occurs not as a homeostatic function of changing nutritional environment but rather as a homeorhetic function caused by the changing expression of genes for processes such as milk production through time. In other words, the animal has genetic drives and there is an aspect of nutrient partitioning that is genetically driven. Evidence for genetic drives other than milk production is available and is discussed. Genetic drives for other life functions than just milk imply that nutrient partitioning will change through lactation and according to genotype - i.e. it cannot be predicted from feed properties alone. Progress in describing genetic drives and homeorhetic controls is reviewed. There is currently a lack of good genetic measures of physiological parameters. The unprecedented level of detail and amounts of data generated by the advent of microarray biotechnology and the fields of genomics, proteomics, etc. should in the long-term provide the necessary information to make the link between genetic drives and metabolism. However, gene expression, protein synthesis etc, have all been shown to be environmentally sensitive. Thus, a major challenge in realising the potential afforded by this new technology is to be able to be able to distinguish genetically driven and environmentally driven effects on expression. To do this we need a better understanding of the basis for the interactions between genotypes and environments. The biological limitations of traditional evaluation of genotype × environment interactions and plasticity are discussed and the benefits of considering these in terms of trade-offs between life functions is put forward. Trade-offs place partitioning explicitly at the centre of the resource allocation problem and allow consideration of the effects of management and selection on multiple traits and on nutrient partitioning.  相似文献
