The long-chain fatty acyl-CoA content of various biological materials, i.e., baker's yeast and mammalian liver, has been determined under standard and several other metabolic conditions, using optimized methods for cell disruption, separating acid-soluble and acid-insoluble CoA from each other, and assaying. After studying the optimization of the extraction of long-chain acyl-CoA compounds and the purification of the extracts, acyl-CoA fractions from several biological sources have been isolated and characterized on behalf of their fatty acid residues by gas-liquid chromatography of the methyl ester derivatives.  相似文献

We propose a new method for estimating gill net selectivity which estimates the probabilities leading to retention by analyzing both the fish morphology and the mesh geometry. This method estimates the number of fish approaching and contacting gill nets of different mesh sizes as an intermediate step towards computing the selectivity. Instead of assuming an underlying probability distribution as in indirect methods, we split the entire interaction between a fish and the gill net into several stages, each with its own probability. All the necessary parameters to compute these probabilities can be obtained from measurements of the fish, knowledge of the mesh geometry, and catch data from different mesh sizes. The framework offers three pathways for computing the total number of fish contacting the gill nets and has the capability to use both wedged and entangled fish in the analysis. As a proof of concept, the method is applied to catch data for cod (G. morhua) and Dolly Varden (S. malma) to estimate the number of fish contacting the gill nets in both cases. By estimating the number of fish contacting the gill net in addition to the selectivity, this method provides an important step towards deriving estimates of fish density in a particular fishery from gill net measurement.  相似文献

Ultrastructural and immunohistochemical analysis of six specimens revealed for the first time a small number of Langerhans cells in non-neoplastic bronchial epithelium. These cells were usually found either interspersed perpendicularly between columnar epithelial cells or just above the basal lamina with extending cytoplasmic processes. Usually few Birbeck granules, especially located in the Golgi region, were present throughout the cytoplasm. Immunohistochemical studies were performed on semi-thin sections with a polyclonal anti-S-100 protein. The population of S-100 positive cells represented about 1% of all the epithelial cells. Not all ultrastructurally identified Langerhans cells were shown to be positive for S-100 antigen. Our results suggest that Langerhans cells could be a constant cellular constituent for the normal bronchial epithelium. The exact function of Langerhans cells in the respiratory epithelium remains to be investigated, but they may have an immunologic function, such as antigen presentation to T lymphocytes.  相似文献

Wheezing during infancy has been linked to early loss of pulmonary function. We prospectively investigated the relation between bronchial hyperresponsiveness (BHR) and progressive impairment of pulmonary function in a cohort of asthmatic infants followed until age 9 years. We studied 129 infants who had had at least three episodes of wheezing. Physical examinations, baseline lung function tests and methacholine challenge tests were scheduled at ages 16 months and 5, 7 and 9 years. Eighty-three children completed follow-up. Twenty-four (29%) infants had wheezing that persisted at 9 years of age. Clinical outcome at age 9 years was significantly predicted by symptoms at 5 years of age and by parental atopy. Specific airway resistance (sRaw) was altered in persistent wheezers as early as 5 years of age, and did not change thereafter. Ninety-five per cent of the children still responded to methacholine at the end of follow-up. The degree of BHR at 9 years was significantly related to current clinical status, baseline lung function, and parental atopy. BHR at 16 months and 5 years of age did not predict persistent wheezing between 5 and 9 years of age, or the final degree of BHR, but it did predict altered lung function. Wheezing that persists from infancy to 9 years of age is associated with BHR and to impaired lung function. BHR itself is predictive of impaired lung function in children, strongly pointing to early airway remodeling in infantile asthma.  相似文献

Comparative ultrastructural analysis of the conducting and non-conducting phloem cells in the common straight-grained silver birch (Betula pendula var. pendula) and the Karelian birch (B. pendula var. carelica) with abnormal patterned wood was carried out, leading to the conclusion that there is an elevated sucrose content in the conducting phloem of the Karelian birch. A connection between sucrose levels and formation of abnormalities in the development of conducting tissues in the Karelian birch trunk was surmised. Experiments in which exogenous sucrose was applied to the silver birch trunk tissues have demonstrated the effects of different sucrose concentrations (0 g L−1, 10 g L−1, 25 g L−1, 50 g L−1, 100 g L−1) on the formation of xylem and phloem structural elements, and they yielded the types of tissue development that correspond to the abnormal tissue development in the Karelian birch trunk.  相似文献

The TonB protein transduces energy from the proton gradient across the cytoplasmic membrane of Gram-negative bacteria to TonB-dependent outer membrane receptors. It is a critically important protein in iron uptake, and deletion of this protein is known to decrease virulence of bacteria in animal models. This system has been used for Trojan horse antibiotic delivery. Here, we describe the high-resolution solution structure of Escherichia coli TonB residues 103-239 (TonB-CTD). TonB-CTD is monomeric with an unstructured N terminus (103-151) and a well structured C terminus (152-239). The structure contains a four-stranded antiparallel beta-sheet packed against two alpha-helices and an extended strand in a configuration homologous to the C-terminal domain of the TolA protein. Chemical shift perturbations to the TonB-CTD (1)H-(15)N HSCQ spectrum titrated with TonB box peptides modeled from the E.coli FhuA, FepA and BtuB proteins were all equivalent, indicating that all three peptides bind to the same region of TonB. Isothermal titration calorimetry measurements demonstrate that TonB-CTD interacts with the FhuA-derived peptide with a K(D)=36(+/-7) microM. On the basis of chemical shift data, the position of Gln160, and comparison to the TolA gp3 N1 complex crystal structure, we propose that the TonB box binds to TonB-CTD along the beta3-strand.  相似文献

BACKGROUND: Blood histamine levels are decreased after severe allergic reactions and in various chronic diseases. AIMS: To study blood histamine levels in infants and children with acute infectious and non-infectious, non-allergic, disease. METHODS: Blood histamine levels were investigated by a fluorometric method in infants and children admitted to hospital with bronchiolitis, non-wheezing bronchitis, acute infections of the urinary tract, skin and ear-nose-throat, gastroenteritis, or hyperthermia of unknown aetiology. Results of blood histamine levels and white blood cell counts were compared with those obtained for children recovering from benign non-infectious, non-allergic illnesses. RESULTS: As compared with control children, white blood cell numbers were significantly increased in children with acute infections of the urinary tract, skin and ear-nose-throat, and were significantly decreased in children with gastroenteritis. Blood histamine levels were significantly lower in children with gastroenteritis and hyperthermia than in children with other diseases and control children. It was not possible to correlate blood histamine levels and the number of blood basophils. CONCLUSIONS: BHL are significantly decreased in infants and children with acute gastroenteritis and hyperthermia of unknown aetiology. The mechanisms responsible for the decrease in blood histamine levels in children with gastroenteritis and hyperthermia are discussed.  相似文献
