The centromeric region of Costus spiralis is characteristically composed of a small heterochromatic DAPI(+) band flanked by a discrete decondensed region. High concentrations of serine 10 of histone H3 (H3S10ph) around the DAPI(+) band in pachytene chromosomes and the location of this heterochromatin at the chromosome region directed towards the poles during metaphase-anaphase I confirm its integration into the centromeric region. Antibodies against both typical components of euchromatin histones (histone H4 acetylated at lysine 5 (H4K5ac) and histone H3 dimethylated at lysine 4 (H3K4me2)) and heterochromatin (dimethylated lysine 9 of H3 (H3K9me2) and anti-5-methylcytosine (5-mC)) were used to characterize the centromeric chromatin of this species during meiosis. In pachytene chromosomes, the decondensed terminal euchromatin of the chromosome arms were seen to be richer in H4K5ac and H3K4me2 histones, while the more condensed proximal region was relatively stronger labeled with anti-H3K9me2 and anti-5-methylcytosine (5-mC). The centromeric region itself, including the DAPI(+) band, was poor in all of these chromatin modifications, but it was highly enriched in H4K5ac at pachytene. Before and after this stage, the centromeric region was poorly labeled with anti-H4K5ac. Hypomethylation and hyperacetylation of any kind of heterochromatin has rarely been reported, and it may be related to the dominant role of the centromere domain over the heterochromatin repeats. 相似文献

Background
Molecular genetic studies on rare tumour entities, such as bone tumours, often require the use of decalcified, formalin-fixed, paraffin-embedded tissue (dFFPE) samples. Regardless of which decalcification procedure is used, this introduces a vast breakdown of DNA that precludes the possibility of further molecular genetic testing. We set out to establish a robust protocol that would overcome these intrinsic hurdles for bone tumour research.Findings
The goal of our study was to establish a protocol, using a modified DNA isolation procedure and quality controls, to select decalcified samples suitable for array-CGH testing. Archival paraffin blocks were obtained from 9 different pathology departments throughout Europe, using different fixation, embedding and decalcification procedures, in order to preclude a bias for certain lab protocols. Isolated DNA samples were subjected to direct chemical labelling and enzymatic labelling systems and were hybridised on a high resolution oligonucleotide chip containing 44,000 reporter elements. Genomic alterations (gains and losses) were readily detected in most of the samples analysed. For example, both homozygous deletions of 0.6 Mb and high level of amplifications of 0.7 Mb were identified.Conclusions
We established a robust protocol for molecular genetic testing of dFFPE derived DNA, irrespective of fixation, decalcification or sample type used. This approach may greatly facilitate further genetic testing on rare tumour entities where archival decalcified, formalin fixed samples are the only source. 相似文献4.
Eukaryotic chromosomes are organized into two large and distinct domains, euchromatin and heterochromatin, which are cytologically
characterized by different degrees of chromatin compaction during interphase/prophase and by post-synthesis modifications
of histones and DNA methylation. Typically, heterochromatin remains condensed during the entire cell cycle whereas euchromatin
is decondensed at interphase. However, a fraction of the euchromatin can also remain condensed during interphase and appears
as early condensing prophase chromatin. 5S and 45S rDNA sites and telomere DNA were used to characterize these regions in
metaphase and interphase nuclei. We investigated the chromosomal distribution of modified histones and methylated DNA in the
early and late condensing prophase chromatin of two species with clear differentiation between these domains. Both species,
Costus spiralis and Eleutherine bulbosa, additionally have a small amount of classical heterochromatin detected by CMA/DAPI staining. The distribution of H4 acetylated
at lysine 5 (H4K5ac), H3 phosphorylated at serine 10 (H3S10ph), H3 dimethylated at lysine 4 or 9 (H3K4me2, H3K9me2), and 5-methylcytosine
was compared in metaphase, prophase, and interphase cells by immunostaining with specific antibodies. In both species, the
late condensing prophase chromatin was highly enriched in H4K5ac and H3K4me2 whereas the early condensing chromatin was very
poor in these marks. H3K9me2 was apparently uniformly distributed along the chromosomes whereas the early condensing chromatin
was slightly enriched in 5-methylcytosine. Signals of H3S10ph were restricted to the pericentromeric region of all chromosomes.
Notably, none of these marks distinguished classical heterochromatin from the early condensing euchromatin. It is suggested
that the early condensing chromatin is an intermediate type between classical heterochromatin and euchromatin. 相似文献

Oxysterols comprise a very heterogeneous group derived from cholesterol through enzymatic and non-enzymatic oxidation. Among them, 7-ketocholesterol (7-KC) is one of the most important. It has potent effects in cell death processes, including cytoxicity and apoptosis induction. Mesenchymal stem cells (MSCs) are multipotent cells characterized by self-renewal and cellular differentiation capabilities. Very little is known about the effects of oxysterols in MSCs. Here, we describe the short-term cytotoxic effect of 7-ketocholesterol on MSCs derived from human adipose tissue. MSCs were isolated from adipose tissue obtained from two young, healthy women. After 24 h incubation with 7-KC, mitochondrial hyperpolarization was observed, followed by a slight increase in the level of apoptosis and changes in actin organization. Finally, the IC50 of 7-KC was higher in these cells than has been observed or described in other normal or cancer cell lines. 相似文献

The composition and trophic structure of reef fish communities in two natural and two artificial reefs along the coast of Paraíba State in north‐eastern Brazil were investigated. A total of 114 species of fish belonging to 47 families were recorded during 120 stationary visual surveys, slightly less than half (46·55%) of which were recorded at all four surveyed localities. Most species are widely distributed on the western Atlantic coast, but several are endemic to Brazil. The greatest diversity and equitability indexes were recorded at the reefs of Sapatas and Cabeço dos Cangulos, whereas the greatest richness and abundance were found at the Queimado shipwreck. The Alvarenga shipwreck reef had the least richness, diversity and equitability. The four localities studied had very similar ichthyofaunas, especially in relation to species composition. The reefs along the Paraíba coast are considered priority conservation areas by the Brazilian Ministry of the Environment, and the information generated by this study will be useful for comparison with other reefs in the region and can be directly applied to programmes seeking to protect and manage these environments. 相似文献

Cinnamic acid is a known allelochemical that affects seed germination and plant root growth and therefore influences several metabolic processes. In the present work, we evaluated its effects on growth, indole-3-acetic acid (IAA) oxidase and cinnamate 4-hydroxylase (C4H) activities and lignin monomer composition in soybean (Glycine max) roots. The results revealed that exogenously applied cinnamic acid inhibited root growth and increased IAA oxidase and C4H activities. The allelochemical increased the total lignin content, thus altering the sum and ratios of the p-hydroxyphenyl (H), guaiacyl (G), and syringyl (S) lignin monomers. When applied alone or with cinnamic acid, piperonylic acid (PIP, a quasi-irreversible inhibitor of C4H) reduced C4H activity, lignin and the H, G, S monomer content compared to the cinnamic acid treatment. Taken together, these results indicate that exogenously applied cinnamic acid can be channeled into the phenylpropanoid pathway via the C4H reaction, resulting in an increase in H lignin. In conjunction with enhanced IAA oxidase activity, these metabolic responses lead to the stiffening of the cell wall and are followed by a reduction in soybean root growth. 相似文献

Species belonging to the Alismataceae (Echinodorus) and Limnocharitaceae (Hydrocleys and Limnocharis) families were analysed by banding with CMA/DAPI fluorochromes, C/CMA/DAPI banding, and in situ hybridization (FISH) with probes that recognise 45S rDNA. All species of Echinodorus presented 2n = 22, but only in E. lanceolatus were DAPI+ telomeric bands in seven chromosome pairs observed. A bimodal karyotype and GC-rich heterochromatin preferably located in two smaller acrocentric pairs that generally corresponded to the number of sites of 45S rDNA. A similar pattern of bands was observed in both Limnocharis species (2n = 20), but the two differed with respect to 45S rDNA, with L. laforestii showing only two sites. Hydrocleys nymphoides and H. martii had a chromosome number of 2n = 16, but the position of the GC-rich heterochromatin associated with the satellite differed among chromosomal types. In this work, the cytotaxonomic implications of these patterns are discussed and correlated with previous data from the literature. 相似文献
