The aim of the present study was to evaluate phytohormone production by symbiotic and saprophytic actinomycetes isolated from
the actinorhizal plant Ochetophila trinervis which had previously proved to stimulate nodulation by Frankia. Three saprophytic strains out of 122, isolated from the rhizosphere of this plant with multiple enzymatic activities were
selected for plant growth experiments in pots: Streptomyces sp. (BCRU-MM40), Actinoplanes sp. (BCRU-ME3) and Micromonospora sp. (BCRU-MM18). For experiments, the symbiotic N2-fixing strain Frankia (BCU110501), isolated from nodules of the same actinorhizal plant was used. Phytohormone production was evaluated in supernatant
of non-inoculated and inoculated culture media in exponential growth phase. Indole 3-acetic acid (IAA) and gibberellic acid
(GA3) were analyzed by gas chromatography-mass spectrometry (GC–MS), while zeatine (Z) production was determined by gas chromatography-flame
ionization detector and high performance liquid chromatography (HPLC fluorescent and UV). The levels of the three phytohormones
produced by the saprophytic rhizoactinomycetes were higher than that produced by the symbiotic Frankia strain. Zeatine biosynthesis was higher (μg ml−1) than IAA and GA3 (ng ml−1), and Micromonospora strain produced the highest levels of these phytohormones. Although O. trinervis has been shown to be intercellularly infected by Frankia without mediation of root hair deformation, when plants were co-inoculated with actinomycetes’ culture, some root hair deformation

A large kindred of hereditary deaf affected with a progressive sensorineural loss that begins during childhood with the low audiologic frequencies is described. Deafness progresses slowly through adolescence, when losses of up to 70 decibels are often detected. Affected adults present profound losses at all frequencies. Genetically, this deafness is transmitted as a simple, dominant, and autosomal mutation. No associated abnormalities have been detected in studies involving medical examinations, care histories, quantitation of several blood serum components, electrocardiograms, electrophoretograms, and karyotypes. 相似文献

The bee tribe Eucerini is a large monophyletic taxon occurring in all continents, except Oceania and Antarctica, but far more diverse in the Americas and, especially, in the Neotropics. The phylogenetic relationships within its subtribe Eucerina, which contains the bulk of Eucerini diversity, is poorly understood, this being especially true for the relationships among its Neotropical representatives, which have been poorly represented in all phylogenetic studies, including Eucerini, to date. This leads to a generalized lack of confidence on the monophyly of the groups currently accepted as genera and subgenera. Here, a phylogenetic study based on three molecular markers (COI, 18S, and 28S, totalling about 1700 bp) and 58 morphological characters is presented as a contribution to the understanding of the relationships of the so‐called Thygater‐Trichocerapis group, and especially of the genus Thygater Holmberg, which has not previously been extensively sampled. Representatives of Trichocerapis, including its monotypic subgenus T. (Dithygater), are included for the first time in a phylogenetic study. The main results were: (i) support for the monophyly of the Thygater‐Trichocerapis group; (ii) support for the monophyly of Thygater; (iii) recognition of two main clades in Thygater, each one containing the type species of one of the previously recognized subgenera; and (iv) additional support for the position of Alloscirtetica as sister to all remaining Eucerina. Based on these results a redefinition of the scope of the two subgenera of Thygater is proposed, with changes in the subordination of three of its species, T. (Nectarodiaeta) chaetaspis comb.n. , T. (Nectarodiaeta) paranaensis comb.n. and T. (Thygater) mexicana comb.n. 相似文献

Cotyledon explants of radiata pine contain principally lipid and protein reserve materials, which decline during shoot initiation. This process was followed phytochemically and ultrastructurally. Fatty acid and sterol analyses indicated that there were both quantitative and qualitative changes in the different classes of lipid. The most pronounced changes were an increase in the linolenic acid content of the polar lipids and the appearance of stigmasterol during shoot initiation. There was also a continued increase in chlorophyll and carotenoid levels, which paralleled chloroplast development. It appears that the changes observed were similar to those that occur in cotyledons during normal seedling development. 相似文献

This study investigated the toxic effects of essential oils isolated from Siparuna guianensis against Aedes aegypti, Culex quinquefasciatus (eggs, larvae, pupae, and adult) and Aedes albopictus (C6/36) cells. The oviposition-deterring activity, egg viability, and repellence activity in the presence of different essential oils concentrations were determined. The essential oils showed high toxicity to all developmental stages of A. aegypti and C. quinquefasciatus. Furthermore, the oils also showed high repellent activity towards the adult stage of mosquitoes (0.025 to 0.550 μg/cm2 skin conferred 100% repellence up to 120 min) and in contact with cultured insect cells (C6/36) induced death possibly by necrosis. The results presented in this work show the potential of S. guianensis essential oils for the development of an alternative and effective method for the natural control of mosquitoes in homes and urban areas. 相似文献

The exact function of MAGE-C1/CT7 and MAGE-C2/CT10 is not yet understood in multiple myeloma (MM). However, the homologs MAGE-C1/CT7 and MAGE-C2/CT10 genes encode highly immunogeneic cancer/testis antigens (CTAs) and can be potential targets for T cell-based immunotherapy. MAGE-C1/CT7 and MAGE-C2/CT10 mRNA expression were investigated in MM patients, solitary plasmacytomas, monoclonal gammopathies of undetermined significance (MGUS) and bone marrow (BM) aspirates from healthy donors by RT-PCR. MAGE-C1/CT7 and MAGE-C1/CT10 were expressed in 67 and 59 % of the 46 MM analyzed patients. At least one of the genes was expressed in 76 % of MM cases. Solitary plasmacytoma also showed MAGE-C1/CT7 and MAGE-C2/CT10 expression. MAGE-C1/CT7 and MAGE-C2/CT10 were not expressed in normal BM samples, showing restricted expression of these CTA genes in MM, solitary plasmacytoma and MGUS. In the present study, we found high expression of the homologs MAGE-C1/CT7 and MAGE-C2/CT10 in monoclonal gammopathies and speculate whether these genes might represent a valuable therapeutic option for myeloma, in particular for combined immunotherapy. 相似文献

With the recent development of retinal prostheses, it is important to develop reliable techniques for assessing the safety of these devices in preclinical studies. However, the standard fixation, preparation, and automated histology procedures are not ideal. Here we describe new procedures for evaluating the health of the retina directly adjacent to an implant. Retinal prostheses feature electrode arrays in contact with eye tissue. Previous methods have not been able to spatially localize the ocular tissue adjacent to individual electrodes within the array. In addition, standard histological processing often results in gross artifactual detachment of the retinal layers when assessing implanted eyes. Consequently, it has been difficult to assess localized damage, if present, caused by implantation and stimulation of an implanted electrode array. Therefore, we developed a method for identifying and localizing the ocular tissue adjacent to implanted electrodes using a (color-coded) dye marking scheme, and we modified an eye fixation technique to minimize artifactual retinal detachment. This method also rendered the sclera translucent, enabling localization of individual electrodes and specific parts of an implant. Finally, we used a matched control to increase the power of the histopathological assessments. In summary, this method enables reliable and efficient discrimination and assessment of the retinal cytoarchitecture in an implanted eye. 相似文献
