Phosphatidic acid (PA) is an important signalling lipid involved in various stress‐induced signalling cascades. Two SnRK2 protein kinases (SnRK2.4 and SnRK2.10), previously identified as PA‐binding proteins, are shown here to prefer binding to PA over other anionic phospholipids and to associate with cellular membranes in response to salt stress in Arabidopsis roots. A 42 amino acid sequence was identified as the primary PA‐binding domain (PABD) of SnRK2.4. Unlike the full‐length SnRK2.4, neither the PABD‐YFP fusion protein nor the SnRK2.10 re‐localized into punctate structures upon salt stress treatment, showing that additional domains of the SnRK2.4 protein are required for its re‐localization during salt stress. Within the PABD, five basic amino acids, conserved in class 1 SnRK2s, were found to be necessary for PA binding. Remarkably, plants overexpressing the PABD, but not a non‐PA‐binding mutant version, showed a severe reduction in root growth. Together, this study biochemically characterizes the PA–SnRK2.4 interaction and shows that functionality of the SnRK2.4 PABD affects root development.  相似文献

Abstract: Rögla is the northernmost locality yielding Mesozoic plant fossils in Scania, southern Sweden, and is one of the northernmost Rhaetian assemblages in Europe. The assemblage consists of over 500 specimens collected 50–60 years ago, of which 139 yielded identifiable plant remains referable to 15 plant species; another 19 specimens are tentatively assigned to four species because of their fragmentary preservation. The flora includes sphenophytes, ferns, cycads, bennettitaleans, seed ferns of uncertain alliance, conifers and some leaf remains that are tentatively assigned to ginkgophytes based on their epidermal anatomy. The species‐level composition of the assemblage is consistent with a Rhaetian age and is similar to well‐known floras from nearby Höganäs and Bjuv, except for the absence of cycads belonging to Nilssonia, which are very common in most other Scanian floras. The fossil assemblage is interpreted to derive from multi‐storey vegetation occupying moist habitats on a coastal plain. Strong affinities are evident with the coeval floras of Jameson Land, Greenland, reinforcing the concept of a distinctive North Atlantic floristic sub‐province at the close of the Triassic.  相似文献

Combined analysis of three experiments showed that when lamb carcases with initial bacterial numbers of between logi103.29 and 4.22/cm2 were spray washed, statistically significant reductions in bacterial numbers of log10O.5 were obtained when the spray wash water temperature was > 57°C, and reductions of log101.0 were obtained when the temperature was ≥ 80°C. Reductions at all temperatures were enhanced by log100.66 when the water contained 30 µg/ml chlorine, but increasing the concentration to 450 µg/ml reduced bacterial numbers only by a further log100–29. At highly contaminated sites increasing the duration of spraying from 30 to 120 s significantly increased the reductions obtained when water containing added chlorine was used. Reductions in bacterial numbers after spray washing with pressures of 3.5, 5.6. 7.7 kg/cm2 were not significantly different.  相似文献

An ecological study of the nodulation of Trifolium repens var. grassland Huai by genetically marked Rhizobium trifolii was carried out in two Irish soils, a neutral mineral and an acidic peat. An indigenous population of 2 x 104 R. trifolii /g was found in the mineral soil. In the peat soil, 4 x 101 R. trifolii /g was found in the uninoculated peat. This number increased to 4.5 x 105 R. trifolii /g, however, eight weeks after the peat soil was neutralized, supplemented with nutrients and sown with uninoculated clover seed. Indigenous R. trifolii strains from the mineral soil were effective whereas strains from the peat soil were ineffective on the host plant T. repens under plant room conditions. The introduced strains were inoculated on to clover seed at the rate of 1 x 105 R. trifolii /seed. In the mineral soil, the introduced inoculum failed to establish at any period during the growing season. In the peat soil, the percentage establishment of the introduced inoculum varied from 40-50% of nodules selected eight weeks after sowing to 70-90% of nodules selected at the end of the growing season.  相似文献

Abstract: We assessed age-dependent survival, site-fidelity, and, together with data on prey and reproduction, trends in the population of ferruginous hawks (Buteo regalis) breeding in western Canada. Our analysis included 7,129 ferruginous hawks banded near Hanna, Alberta, and Kindersley-Alsask, Saskatchewan, from 1972 to 2003. We estimated annual adult survival rate to be 0.708 (SE = 0.024) and first year survival for nestlings was 0.545 (SE = 0.147). Resighting probability was modeled as a constant for nestlings (0.009, SE = 0.010), but it varied among years for adults consistent with our sampling efforts. Band reporting rate was at 0.022 (SE = 0.007) for both nestlings and adults. Fidelity to the study site was 1.00 (SE = 0.000) for adults and 0.035 (SE = 0.014) for nestlings. Nesting density ranged from 3.1 to 14.0 pairs/100 km2 and averaged 9.8 pairs/100 km2. We observed an average clutch size of 3.2 (SE=0.06) and brood sizes of 2.71 (SE=0.07) near Hanna and 2.79 (SE = 0.99) at Kindersley-Alsask. Richardson's ground squirrels (Spermophilus richardsonii) made up 95% of ferruginous hawk prey. Prey availability was positively correlated with number of offspring near Hanna and Kindersley-Alsask. We believe the lower than expected adult survival did not result in population decrease; rather, declines in reproduction resulting from declines in the abundance of ground squirrels better explain an observed 4.5-fold decline in nesting densities during the study. The results suggest that ferruginous hawk management should address prey in addition to habitat management, and that management needs are regional in scope with particular emphasis on the breeding range within the northern Great Plains.  相似文献

1. We derived models of nutrient release [nitrogen (N) and phosphorus (P)] by fish based on studies that directly measured the release rates from 56 species across a broad range of fish mass, feeding histories and temperature. 2. We developed four separate models of nutrient release from multiple regression analysis: detritivore release rates of N and P, and non‐detritivore release rates of N and P. 3. Fish mass explained most of the variance (78–92%) in release rates. 4. Our predicted rates of release of P by fish (g ha?1 day?1) were similar to observed rates in the literature from other lakes. 5. The influence of a shift in diet (planktivory to detritivory) by a single species (gizzard shad, Dorosoma cepedianum, a facultative detritivore) on nutrient release rates was estimated. During periods of detritivory, gizzard shad accounted for on average 39% (<1–96%) of all nutrients released by the fish assemblage, and increased total fish assemblage release rates on average by 59% (<1–331%) compared to when gizzard shad were modelled as planktivores. 6. These models provide a rapid means for predicting the release of nutrients by fish assemblages and may facilitate more comprehensive comparisons of nutrient cycling by fish with other internal pathways.  相似文献

Abstract: Direct and indirect effects of industrial development have contributed, in part, to the threatened status of boreal ecotype caribou (Rangifer tarandus caribou) in Alberta and Canada. Our goal was to develop a model that would allow managers to identify landscape-scale targets for industrial development, while ensuring functional habitat for sustainable caribou populations. We examined the relationship between functional habitat loss resulting from cumulative effects of natural and anthropogenic disturbance, and the rate of population change (Λ) for 6 populations of boreal caribou in Alberta, Canada. We defined functional habitat loss according to 2 variables for which we had a priori reasons to suspect causative associations with Λ: 1) percentage area of caribou range within 250 m of anthropogenic footprint, and 2) percentage of caribou range disturbed by wildfire within the last 50 years. Multiple regression coefficients for both independent variables indicated significant effects on Λ. The 2-predictor model explained 96% (R2) of observed variation in Λ among population units (F2,3 = 35.2, P = 0.008). The model may be used to evaluate plans for industrial development in relation to predicted wildfire rates and goals for caribou population growth rates.  相似文献

Rock‐dwelling, endolithic micro‐organisms can create tubular microcavities (TMCs) by the dissolution of rock substrates. Microtunnels can also conceivably be formed by abiotic processes, and collectively, these structures are here termed tubular microcavities. A textural record of life in subseafloor environments is provided by biological TMCs, and it is imperative to distinguish these from abiological tunnels. To this end, the morphologies and petrographic context of tunnels formed by chemical solution, physical abrasion, and biological processes are here described. Biological TMCs in volcanic glass are restricted to sites that were connected to early fluid circulation. Their shapes, distribution, and the absence of intersections exclude an origin by chemical dissolution of pre‐existing heterogeneities such as, radiation damage trails, gas‐escape structures, or fluid inclusion trails. Rather their characteristics are best explained by microbial dissolution, involving perhaps, cellular extensions that provide a mechanism of localizing and directing microtunnel formation as observed in terrestrial soils. Biological TMCs are contrasted with ambient inclusion trails (AITs) found in cherts and authigenic minerals. These differ in exhibiting longitudinal striae, a constant diameter, and polygonal cross‐section, sometimes with terminal inclusions. The origin(s) of AITs remain unclear but they are hypothesized to form by migration of crystalline or organic inclusions in sealed substrates, in contrast to biotic TMCs that form in open systems. We present diagnostic morphological and petrographic criteria for distinguishing these different types of TMCs. Moreover, we argue that AIT‐type processes are not viable in volcanic glass because of the absence of crystalline millstones, localized chemical solution agents, and elevated fluid pressures, necessary to drive this process.  相似文献
