Possibilities of a new principle for the homogeneous enzyme immunoassay utilizing the systems of surfactant reversed micelles in organic solvents have been demonstrated taking thyroxine determination as an example. The catalytic activity of an enzyme, solubilized in such systems, is determined by the ratio of geometric dimensions of the micellar matrice and the enzyme molecule. The addition of antibodies against thyroxine to the peroxidase-thyroxine conjugate, solubilized in the system of reversed micelles of aerosol OT in octane, leads to the formation of the immune complex whose size differs substantially from that of the initial enzyme-antigen conjugate. This induces changes in the peroxidase catalytic activity. The addition of free thyroxine to the system stimulates the conjugate release from the immune complex and, consequently, the reduction of the peroxidase activity to the initial level. Sensitivity of the analysis in reversed micellar systems can be regulated by changing the surfactant hydration degree. Substances of different nature (both hydrophobic and hydrophylic) can be solubilized in reverse micellar systems under standard conditions, which allows determination of water insoluble antigens.  相似文献

The effect of essential fatty acids (EFA), given orally as dietary supplements, on the responsiveness in vitro of peripheral blood lymphocytes (PBL), to the mitogen concanavalin A have been studied in 10 patients with localized and 14 patients with advanced colorectal cancer. The degree of lymphocyte activation was assessed by measuring the amount of tritiated [3H]thymidine incorporated into newly synthesised lymphocyte DNA. The results were expressed as stimulation indices. T cell responses to concanavalin A stimulation showed a significant reduction of stimulation indices following EFA supplementation, in both the localized (P=0.026) and advanced (P=0.016) tumour groups, when compared with pretreatment activity in vitro. Mixing experiments, using EFA-supplemented and non-EFA-supplemented lymphocytes with concanavalin A, suggest no enhancement of T suppressor cell activity. Cell surface marker analysis (fluorescence-activated cell sorting for CD phenotyping) revealed a reduction of absolute numbers of CD4+ and CD8+ lymphocytes following EFA supplementation. The stimulation indices returned to presupplementation values 3 months following cessation of EFA intake. There was no significant change of these indices in the control (no EFA supplementation) advanced tumour group tested. This study suggests that EFA supplementation in patients with colorectal cancer selectively reduces circulating PBL. and T cell subset (including suppressor cells) numbers and/or activity. The V3 loop on gp120 from human immunodeficiency virus type 1 (HIV-1) is a focus of many research groups involved in anti-AIDS drug development because this region of the protein is a principal target for neutralizing antibodies and a major determinant for cell tropism and syncytium formation. In this study, the nucleotide sequences of the env gene region coding the V3 loop were determined by DNA sequencing methods for four novel HIV-1 strains that circulate in the countries of Eastern Europe, such as Russia, Belarus, Ukraine, etc. Based on the empirical data obtained, the 3D structures of the V3 loops associated with these viral modifications were generated by computer modeling and then compared to discover similarities in the spatial arrangement of this functionally important site of gp120. Despite the HIV-1 genetic variety, several regions of the V3 loop that contain residues critical for cell tropism were shown to be structurally invariant, which may explain its exceptional role in a co-receptor usage. These data together with those on the biological activity of the V3 individual residues clearly show that these conserved structural motifs of gp120 represent potential HIV-1 weak points most suitable for therapeutic intervention.  相似文献

It was shown that bidistilled modified water induces a marked decrease in the intensity of intrinsic fluorescence of bovine serum albumin and increases the binding of this protein to the fluorescent probe 1.8 ANS. These effects can be interpreted as a denaturing action of bidistilled modified water on the protein and a change in its conformational state, which is probably caused by changes in the microenvironment of the protein molecule. In addition, a substantial increase in the intrinsic fluorescence of bidistilled modified water, as compared with that of distilled water, was found.  相似文献

The effect of the degree of hydration (W0) of reversed micelles of Aerosol OT (AOT) and its mixture with Triton X-45 in heptane on the peroxidase activity of horse spleen ferritin in the oxidation of various substrates by hydrogen peroxide and organic hydroperoxides and on the activity of solubilized or immobilized immunocomplexes of horseradish peroxidase-cortisol conjugates (HP-COR) was studied. The peroxidase activity versus W0 plot has maxima at W0 8-14 and 19-22, which cannot be attributed to dissociation of immunocomplexes into its components or of ferritin into its subunits. The possibility of the stabilization of the conformers of oligomeric proteins by reversed micelles and the effect of the self-association of micelles on the peroxidase activity of the HP-COR immunocomplexes and ferritin were discussed. A procedure for the isolation of the iron-containing cluster from the ferritin molecule without reduction of the Fe3+ ions was suggested.  相似文献

Comparative study of virulence of B. anthracis strains harbouring pXO1 and pXO2 plasmids in mice and guinea pigs showed that among six B. anthracis strains, three were 100-1000 times less virulent for guinea pigs. Genetic construction of B. anthracis strains using transduction and conjugation transfer of resident plasmids permitted us to rule out the effects of modified pXO1 and pXO2 replicons and to prove the existence of nonidentified chromosome locuses responsible for the development of an infectious process in anthrax, along with plasmid determinants of virulence.  相似文献

Kinetics of Catalase Inactivation Induced by Ultrasonic Cavitation   总被引:1,自引:0,他引:1  
Kinetic patterns of sonication-induced inactivation of bovine liver catalase (CAT) were studied in buffer solutions (pH 4.0–11.0) within the temperature range from 36 to 55o. Solutions of CAT were exposed to LF (20.8 kHz) ultrasound (specific power, 48–62 W/cm2). The kinetics of CAT inactivation was characterized by effective first-order rate constants (s–1) of total inactivation (k in), thermal inactivation (*k in), and ultrasonic inactivation (k in(us)). In all cases, the following inequality was valid: k in > *k in. The value of k in(us) increased with the ultrasound power (range, 48–62 W/cm2) and exhibited a strong dependence on the pH of the medium. On increasing initial concentration of CAT (0.4–4.0 nM), k in(us) decreased. The three rate constants were minimum within the range pH 6.5–8.0; their values increased considerably at pH < 6.0 and pH > 9.0. At 36–55o, the temperature dependence of k in(us) was characterized by an activation energy (E act) of 19.7 kcal/mol, whereas the value of E act for CAT thermoinactivation was equal to 44.2 kcal/mol. Bovine and human serum albumins (BSA and HSA, respectively) inhibited sonication-induced CAT inactivation; complete prevention was observed at concentrations above 2.5 g/ml. Dimethyl formamide (DMFA), a scavenger of hydroxyl radicals (O ), prevented sonication-induced CAT inactivation at 10% (k in and *k in increased with the content of DMFA at concentrations in excess of 3%). The results obtained indicate that free radicals generated in the field of ultrasonic cavitation play a decisive role in the inactivation of CAT, which takes place when its solutions are exposed to low-frequency ultrasound. However, the efficiency of CAT inactivation by the radicals is determined by (1) the degree of association between the enzyme molecules in the reaction medium and (2) the composition thereof.  相似文献

A method for isolating extracellular glucose oxidase from the fungus Penicillium funiculosum 46.1 using ultrafiltration membranes was developed. Two samples of the enzyme with a specific activity of 914–956 IU were obtained. The enzyme exhibited a high catalytic activity at pH above 6.0. The effective rate constant of glucose oxidase inactivation at pH 2.6 and 16°C was 2.74 × 10–6 s–1. This constant decreased significantly as the pH of the medium increased (4.0–10.0). The temperature optimum for glucose oxidase–catalyzed -D-glucose oxidation was in the range 30–65°C. At temperatures below 30°C, the activation energy for -D-glucose oxidation was 6.42 kcal/mol; at higher temperatures, this parameter was equal to 0.61 kcal/mol. Kinetic parameters of glucose oxidase–catalyzed -D-glucose oxidation depended on the initial concentration of the enzyme in the solution. Glucose oxidase also catalyzed the oxidation of 2-deoxy-D-glucose, maltose, and galactose.  相似文献
