CAM requires a substantial investment of resources into storage carbohydrates to account for nocturnal CO2 uptake, thereby restricting carbohydrate partitioning to other metabolic activities, including dark respiration, growth and acclimation to abiotic stress. Flexible modulation of carbon flow to the different competing sinks under changing environmental conditions is considered a key determinant for the growth, productivity and ecological success of the CAM pathway. The aim of the present study was to examine how shifts in carbohydrate partitioning could assure maintenance of photosynthetic integrity and a positive carbon balance under conditions of increasing water deprivation in CAM species. Measurements of gas exchange, leaf water relations, malate, starch and soluble sugar (glucose, fructose and sucrose) contents were made in leaves of the CAM bromeliad Aechmea ‘Maya’ over a 6‐month period of drought and subsequently over a 2‐month period of recovery from drought. Results indicated that short‐term influences of water stress were minimized by elevating the level of respiratory recycling, and carbohydrate pools were maintained at the expense of export for growth while providing a comparable nocturnal carbon gain to that in well‐watered control plants. Longer term drought resulted in a disproportionate depletion of key carbohydrate reserves. Sucrose, which was of minor importance for providing substrate for the dark reactions under well‐watered conditions, became the major source of carbohydrate for nocturnal carboxylation as drought progressed. Flexibility in terms of the major carbohydrate source used to sustain dark CO2 uptake is therefore considered a crucial factor in meeting the carbon and energy demands under limiting environmental conditions. Recovery from CAM‐idling was found to be dependent on the restoration of the starch pool, which was used predominantly for provision of substrate for nocturnal carboxylation, while net carbon export was limited. The conservation of starch for the nocturnal reactions might be adaptive with regard to responding efficiently to a return of water stress.  相似文献

This paper establishes a framework within which a rapid and pragmatic assessment of river ecosystems can be undertaken at a broad, subcontinental scale, highlighting some implications for achieving conservation of river biodiversity in water‐limited countries. The status of river ecosystems associated with main rivers in South Africa was assessed based on the extent to which each ecosystem had been altered from its natural condition. This requires consistent data on river integrity for the entire country, which was only available for main rivers; tributaries were thus excluded from the analyses. The state of main river ecosystems in South Africa is dire: 84% of the ecosystems are threatened, with a disturbing 54% critically endangered, 18% endangered, and 12% vulnerable. Protection levels were measured as the proportion of conservation target achieved within protected areas, where the conservation target was set as 20% of the total length of each river ecosystem. Sixteen of the 112 main river ecosystems are moderately to well represented within protected areas; the majority of the ecosystems have very low levels of representation, or are not represented at all within protected areas. Only 50% of rivers within protected areas are intact, but this is a higher proportion compared to rivers outside (28%), providing some of the first quantitative data on the positive role protected areas can play in conserving river ecosystems. This is also the first assessment of river ecosystems in South Africa to apply a similar approach to parallel assessments of terrestrial, marine, and estuarine ecosystems, and it revealed that main river ecosystems are in a critical state, far worse than terrestrial ecosystems. Ecosystem status is likely to differ with the inclusion of tributaries, since options may well exist for conserving critically endangered ecosystems in intact tributaries, which are generally less regulated than main rivers. This study highlights the importance of healthy tributaries for achieving river conservation targets, and the need for managing main rivers as conduits across the landscape to support ecological processes that depend on connectivity. We also highlight the need for a paradigm shift in the way protected areas are designated, as well as the need for integrated river basin management plans to include explicit conservation visions, targets, and strategies to ensure the conservation of freshwater ecosystems and the services they provide.  相似文献

Understanding tolerance of thermal extremes by pest insects is essential for developing integrated management strategies, as tolerance traits can provide insights into constraints on activity and survival. A major question in thermal biology is whether thermal limits vary systematically with microclimate variation, or whether other biotic or abiotic factors can influence these limits in a predictable manner. Here, we report the results of experiments determining thermal limits to activity and survival at extreme temperatures in the stalk borer Eldana saccharina Walker (Lepidoptera: Pyralidae), collected from either Saccharum spp. hybrids (sugarcane) (Poaceae) or Cyperus papyrus L. (Cyperaceae) and then reared under standard conditions in the laboratory for 1–2 generations. Chill‐coma temperature (CTmin), critical thermal maximum (CTmax), lower lethal temperatures (LLT), and freezing temperature between E. saccharina collected from the two host plants were compared. CTmin and CTmax of E. saccharina moths collected from sugarcane were significantly lower than those from C. papyrus (CTmin = 2.8 ± 0.4 vs. 3.9 ± 0.4 °C; CTmax = 44.6 ± 0.1 vs. 44.9 ± 0.2 °C). By contrast, LLT of moths and freezing temperatures of pupae did not vary with host plant [LLT for 50% (LT50) of the moth population, when collected from sugarcane: ?3.2 ± 0.5 °C, from C. papyrus: ?3.9 ± 0.8 °C]. Freezing temperatures of pupae collected from C. papyrus were ?18.0 ± 1.0 °C and of those from sugarcane ?17.5 ± 1.8 °C. The E. saccharina which experienced the lowest minimum temperature (in C. papyrus) did not have the lowest CTmin, although the highest estimate of CTmax was found in E. saccharina collected from C. papyrus and this was also the microsite which reported the highest maximum temperatures. These results therefore suggest that host plant may strongly mediate lower critical thermal limits, but not necessarily LLT or freezing temperatures. These results have significant implications for ongoing pest management and thermal biology of these and other insects.  相似文献

The riparian zone and instream habitat integrity of the Luvuvhu River were assessed based on a qualitative rating of the impacts of major disturbance factors such as water abstraction, flow regulation, bed and channel modification, etc. A system was devised to assess the impact of these factors on the relative frequency and variability of habitats on a spatial and temporal scale gauged against habitat characteristics that could have been expected to occur under conditions not anthropogenically influenced. It was found that deterioration of habitat integrity can be ascribed primarily to water abstraction. This has resulted in the cessation of surface flow in a naturally perennial river during the dry season and during droughts with consequent tree deaths and a loss of fast flowing instream habitat types in the main stem of the river. The relatively small high rainfall area in the catchment, the highly variable rainfall pattern and the occurrence of sporadic severe droughts exacerbate the impact of water abstraction on the instream and riparian habitats with expected detrimental consequences for the associated biota. The effect of water abstraction is particularly severe in the lower part of the river which flows through the Kruger National Park as no perennial tributaries join the Luvuvhu River in this section. Other factors which affect the habitat integrity of the river are the removal of indigenous riparian vegetation in some river sections, encroachment by exotic vegetation, bank erosion and stream bed modification.  相似文献

Aim Invasive alien species (IAS) pose a significant threat to biodiversity. The Convention on Biological Diversity’s 2010 Biodiversity Target, and the associated indicator for IAS, has stimulated globally coordinated efforts to quantify patterns in the extent of biological invasion, its impact on biodiversity and policy responses. Here, we report on the outcome of indicators of alien invasion at a global scale. Location Global. Methods We developed four indicators in a pressure‐state‐response framework, i.e. number of documented IAS (pressure), trends in the impact of IAS on biodiversity (state) and trends in international agreements and national policy adoption relevant to reducing IAS threats to biodiversity (response). These measures were considered best suited to providing globally representative, standardized and sustainable indicators by 2010. Results We show that the number of documented IAS is a significant underestimate, because its value is negatively affected by country development status and positively by research effort and information availability. The Red List Index demonstrates that IAS pressure is driving declines in species diversity, with the overall impact apparently increasing. The policy response trend has nonetheless been positive for the last several decades, although only half of countries that are signatory to the Convention on Biological Diversity (CBD) have IAS‐relevant national legislation. Although IAS pressure has apparently driven the policy response, this has clearly not been sufficient and/or adequately implemented to reduce biodiversity impact. Main conclusions For this indicator of threat to biodiversity, the 2010 Biodiversity Target has thus not been achieved. The results nonetheless provide clear direction for bridging the current divide between information available on IAS and that needed for policy and management for the prevention and control of IAS. It further highlights the need for measures to ensure that policy is effectively implemented, such that it translates into reduced IAS pressure and impact on biodiversity beyond 2010.  相似文献

The standard treatment of tuberculosis (TB) takes six to nine months to complete and this lengthy therapy contributes to the emergence of drug-resistant TB. TB is caused by Mycobacterium tuberculosis (Mtb) and the ability of this bacterium to switch to a dormant phenotype has been suggested to be responsible for the slow clearance during treatment. A recent study showed that the replication rate of a non-virulent mycobacterium, Mycobacterium smegmatis, did not correlate with antibiotic susceptibility. However, the question whether this observation also holds true for Mtb remains unanswered. Here, in order to mimic physiological conditions of TB infection, we established a protocol based on long-term infection of primary human macrophages, featuring Mtb replicating at different rates inside the cells. During conditions that restricted Mtb replication, the bacterial phenotype was associated with reduced acid-fastness. However, these phenotypically altered bacteria were as sensitive to isoniazid, pyrazinamide and ethambutol as intracellularly replicating Mtb. In support of the recent findings with M. smegmatis, we conclude that replication rates of Mtb do not correlate with antibiotic tolerance.  相似文献

Fly larvae can be used effectively to reduce various organic waste types and produce value-added products, including protein as an ingredient in livestock feeds and oil for biodiesel production. However, fly development on different waste types may cause differences in growth rate and the body composition, which can further be influenced by fly species and their stocking rate. This study explored the impact of different waste types (kitchen waste, abattoir waste and swine manure) and larval stocking rate on growth and body composition of four blowfly species, Chrysomya chloropyga (Wiedemann), Chrysomya megacephala (Fabricius), Chrysomya putoria (Wiedemann) and Lucilia sericata (Meigen). First-instar larvae (20, 50 or 100), less than 3 hr old, were placed on 100 g of each waste type. Pre-pupal mass at commencement of post-feeding larval dispersal, time to onset of dispersal, survival and nutrient reserves were determined for each species, stocking rate and waste type. Our results revealed that larvae fed kitchen and abattoir waste had significantly higher dry mass, crude protein and lipid content compared with those fed swine manure. Higher survival rate was observed with increasing larval stocking rate. We provide important information to guide the mass production of high-quality nutrient-rich larvae and recommend C. putoria, which is versatile and effective on a range of waste products, as well as high in protein and lipids. The implications for waste management are discussed.  相似文献
