Molecular analysis of a reciprocal translocation t(5;11)(q11;p13) in a WAGR patient

Summary Most patients with the complex association aniridia — predisposition to Wilms' tumor (WAGR syndrome) present with a de novo constitutional deletion of band 11p13. We report a patient with WAGR syndrome and a reciprocal translocation between chromosomes 5 and 11 t(5;11)(q11;p13). High resolution banding cytogenetic analysis and molecular characterization using 11p13 DNA markers showed a tiny deletion encompassing the gene for CAT but sparing the gene for FSHB. This suggests that syndromes associated with apparently balanced translocations may be due to undetectable loss of material at the breakpoint(s) rather than to breakage in the gene itself.     

FVB mice transgenic for the H-2Db gene become resistant to persistent infection by Theiler's virus.

The DA strain of Theiler's virus causes a persistent infection of the white matter of the spinal cord with chronic inflammation and primary demyelination. Inbred strains of mice differ greatly in their susceptibility to this disease. It has been shown that both viral persistence and demyelination are controlled mainly by a gene located in the H-2D region. This raised the possibility that the H-2D gene itself controls viral persistence, which in turn determines demyelination. In the present work we introduced the H-2Db gene of resistant C57BL/6 mice into the genome of susceptible H-2q FVB mice and showed that the FVB mice become resistant to persistence of the infection and did not develop inflammatory lesions.     

Isolation and characterization of cytochrome b5 from Candida tropicalis grown on alkane

Cytochrome b₅ from Candida tropicalis grown on alkane has been solubilized in three different ways (sodium cholate, trypsin, osmotic wash). After solubilization of the microsomal membrane with sodium cholate, the purification of cytochrome b₅ was achieved by DEAE-cellulose chromatography, hydroxylapatite chromatography, a second DEAE-cellulose chromatography and a Sephadex G-75 gel filtration. The purified protein had an apparent molecular weight of 16 000 ± 1 000. After solubilization by trypsin treatment or osmotic wash, the purification procedure yielded a protein with an apparent molecular weight of 12 000 ± 1 000. Though the purified proteins presented molecular weights depending on the technique of solubilization, they exhibited identical optical properties, a great stability with respect to temperature and pH, and were all autooxidable. Redox titrations revealed differences in their midpoint potential values, which were 35 ± 5 mV for the b₅ purified after cholate solubilization, —59 ± 5 mV for the b₅ purified after trypsin treatment and —65 ± 5 mV for the b₅ purified after osmotic wash.     

Microsomal cytochromes of Candida tropicalis grown on alkanes

A comparison of methods used in isolating microsomes and in measuring microsomal cytochrome P-450 demonstrated that separation following protoplast lysis gave the best results. By this latter technique a high amount of cytochrome P-450 (0.2–0.3 nmol/mg) was recovered but cytochrome P-420, considered as the denatured form, was absent.The alkanes specifically induce cytochromes P-450 and b₅ localized on the microsomes. The denaturation in vivo of cytochrome P-450 into cytochrome P-420 even occurs during storage at 1 °C. This degradation is increased during preparation of subcellular fractions if no preventive measures are taken.
