BackgroundVisceral leishmaniasis (VL) is a multifactorial disease, where the host genetics play a significant role in determining the disease outcome. The immunological role of anti-inflammatory cytokine, Interleukin 10 (IL10), has been well-documented in parasite infections and considered as a key regulatory cytokine for VL. Although VL patients in India display high level of IL10 in blood serum, no genetic study has been conducted to assess the VL susceptibility / resistance. Therefore, the aim of this study is to investigate the role of IL10 variations in Indian VL; and to estimate the distribution of disease associated allele in diverse Indian populations.MethodologyAll the exons and exon-intron boundaries of IL10 were sequenced in 184 VL patients along with 172 ethnically matched controls from VL endemic region of India.
Result and Discussion
Our analysis revealed four variations; rs1518111 (2195 A>G, intron), rs1554286 (2607 C>T, intron), rs3024496 (4976 T>C, 3’ UTR) and rs3024498 (5311 A>G, 3’ UTR). Of these, a variant g.5311A is significantly associated with VL (χ2=18.87; p =0.00001). In silico approaches have shown that a putative micro RNA binding site (miR-4321) is lost in rs3024498 mRNA. Further, analysis of the above four variations in 1138 individuals from 34 ethnic populations, representing different social and linguistic groups who are inhabited in different geographical regions of India, showed variable frequency. Interestingly, we have found, majority of the tribal populations have low frequency of VL (‘A’ of rs3024498); and high frequency of leprosy (‘T’ of rs1554286), and Behcet’s (‘A’ of rs1518111) associated alleles, whereas these were vice versa in castes. Our findings suggest that majority of tribal populations of India carry the protected / less severe allele against VL, while risk / more severe allele for leprosy and Behcet’s disease. The financial returns to Kenyan tourism demonstrate the importance of the country's tourist potential to its economic development. Protected areas and their inhabitants are the principal focus of the tourist industry, the nations's main foreign exchange earner, and a source of wonder and value for a global population of non-users. It might be expected that such assets would be accorded some degree of security with sufficient funding to safeguard current and potential economic benefits. Yet park use is haphazard, and there is frequently little coincidence between those that benefit and those that pay for the continued existence of such areas. Growing economic and demographic pressures which threaten to swamp protected areas only emphasize the implicit subsidy currently paid by Kenyans to support conservation for the benefit of the world at large. In this climate the case for conservation depends on the measurement and capture of economic benefits. Using a contingent valuation survey of expressed preference this study estimates the consumer surplus attached to current non-consumptive use of protected areas by foreign visitors at some $450 million per annum. This sum alone is more than double the best available estimate of opportunity cost and appears to justify current resource use. The estimate is additional to current financial returns from tourism and makes no allowance for other direct and indirect benefits and potential returns from consumptive uses. Measured consumer surplus contains some margin of willingness to pay that could be captured through the current fee structure. Moreover, park fees represent the most accessible market mechanism to finance revenue sharing and additional park investment before potential recourse to emerging global market institutions. 相似文献

A crown-of-thorns starfish control program was conducted at Grub Reef (central Great Barrier Reef) in an area (0.64 km2) which encompassed 53 individual patch reefs. During a two week period, 15 divers injected 3175 starfish with copper sulphate. The program was considered unsuccessful. Although starfish abundance had declined significantly after the control efforts, biological surveys indicated that a relatively large number of starfish remained. The surveys also indicated a general decline in the number of starfish along the reef perimeter, outside the control area. The total cost of the control program was $35 per starfish. These results have important implications for the implementation of future control programs and highlight the need to undertake before and after biological surveys to assess the effectiveness of the control efforts. 相似文献

Consistent individual differences in animal performance drive individual fitness under variable environmental conditions and provide the framework through which natural selection can operate. Underlying this concept is the assumption that individuals will display consistent levels of performance in fitness-related traits and interest has focused on individual variation and broad sense repeatability in a range of behavioural and physiological traits. Despite playing a central role in maintenance and growth, and with considerable inter-individual variation documented, broad sense repeatability in rates of protein synthesis has not been assessed. In this study we show for the first time that juvenile flounder Platichthys flesus reared under controlled environmental conditions on the same plane of nutrition for 46 days maintain consistent whole-animal absolute rates of protein synthesis (As). By feeding meals containing 15N-labelled protein and using a stochastic end-point model, two non-terminal measures of protein synthesis were made 32 days apart (d14 and d46). As values (mass-corrected to a standard mass of 12 g) showed 2- to 3-fold variation between individuals on d14 and d46 but individuals showed similar As values on both days with a broad sense repeatability estimate of 0.684 indicating significant consistency in physiological performance under controlled experimental conditions. The use of non-terminal methodologies in studies of animal ecophysiology to make repeat measures of physiological performance enables known individuals to be tracked across changing conditions. Adopting this approach, repeat measures of protein synthesis under controlled conditions will allow individual ontogenetic changes in protein metabolism to be assessed to better understand the ageing process and to determine individual physiological adaptive capacity, and associated energetic costs of adaptation, to global environmental change. 相似文献

Microsomes isolated from developing seeds of Helianthus annuus were prepared in a medium which ensured that endoplasmic reticulum (ER)-bound polysomes remained attached to the ER during homogenization. The microsomes were then incubated with the substrates necessary to sustain the synthesis of triacylglycerols (TAGs). Microsomes that contained high activities of the enzymes involved in the synthesis of TAGs (the enzymes of the Kennedy pathway) accumulated TAGs synthesized in vitro , resulting in a decrease in their buoyant density. These light membrane fractions could therefore be separated on discontinuous sucrose density gradients from microsomes containing low activities of the enzymes of the Kennedy pathway. Analysis of the microsome fractions by 1H-NMR spectroscopy showed that the TAGs synthesized in the microsomes in vitro were tumbling isotropically in an environment similar to that of the TAGs in oil bodies. Western blot analysis revealed that microsomes which synthesized large amounts of TAGs in vitro were also substantially enriched in oleosins. In addition, labelling studies indicated that the oleosins newly synthesized in vitro by ‘run-on' translation of ER-bound polysomes also localized to light membrane fractions. This indicates that oleosins are specifically enriched in regions of the ER involved in the biogenesis of the oil body. 相似文献
