This study examines the relationship of profundal oxygen concentrations in 55 shallow Florida lakes to humic color, trophic
state, and lake size during different seasons. The data set represented a broad range of color and trophic state. The percent
saturation of dissolved oxygen remained relatively constant during the fall (mean 78.4%), winter (mean 81.3%), and spring
(mean 82.5%), but declined markedly during summer (mean 65.2%). Chlorophyll a concentrations were highest during the winter
(mean 2.52 mg m–3) and lowest during the fall (mean 1.17 mg m–3), while color peaked during the fall (mean 30.1 mg Pt l–1) and was lowest during the summer (mean 12.7 mg Pt l–1). The relative importance of lake size, chlorophyll a, and color in explaining variation in percent oxygen saturation was
examined using multiple regression. Percent oxygen saturation was negatively correlated with color during the winter, spring,
and summer, and positively correlated with lake size in the winter and spring. However, percent oxygen saturation showed no
relationship with chlorophyll a during any season. These results suggest that colored Florida lakes are naturally oxygen depleted
and that profundal oxygen values have little relationship to lake trophic state.

Meprin A and B are highly regulated, secreted, and cell-surface metalloendopeptidases that are abundantly expressed in the kidney and intestine. Meprin oligomers consist of evolutionarily related alpha and/or beta subunits. The work herein was carried out to identify bioactive peptides and proteins that are susceptible to hydrolysis by mouse meprins and kinetically characterize the hydrolysis. Gastrin-releasing peptide fragment 14-27 and gastrin 17, regulatory molecules of the gastrointestinal tract, were found to be the best peptide substrates for meprin A and B, respectively. Peptide libraries and a variety of naturally occurring peptides revealed that the meprin beta subunit has a clear preference for acidic amino acids in the P1 and P1' sites of substrates. The meprin alpha subunit selected for small (e.g. serine, alanine) or hydrophobic (e.g. phenylalanine) residues in the P1 and P1' sites, and proline was the most preferred amino acid at the P2' position. Thus, although the meprin alpha and beta subunits share 55% amino acid identity within the protease domain and are normally localized at the same tissue cell surfaces, they have very different substrate and peptide bond specificities indicating different functions. Homology models of the mouse meprin alpha and beta protease domains, based on the astacin crystal structure, revealed active site differences that can account for the marked differences in substrate specificity of the two subunits. 相似文献

The role of ciliated protozoa in pelagic freshwater ecosystems 总被引:11,自引:0,他引:11
The abundance and biomass of ciliates are both strongly related to lake trophic status as measured by chlorophylla concentrations. Taxonomic replacements occur with increasing eutrophication such that large-bodied forms (predominantly oligotrichs) are progressively replaced by smaller-bodied ciliates (mainly scuticociliates). Highly acidic lakes display a more pronounced dominance of large-bodied forms when contrasted with less acidic lakes of comparable trophy. Community structure of ciliate populations is determined largely by lake trophy with acidic oligotrophic systems being characterized by reduced diversity and species richness compared with hypereutrophic systems. The temporal and spatial distribution of small (< 100m) ciliate populations is ascribed to lake thermal regimes which provide localized concentrations of food resources. Likewise, in extremely productive lakes, very large (> 100m) meroplanktonic ciliates enter the water column during midsummer after the development of thermal stratification and associated profundal deoxygenation. Laboratory studies indicate that large zooplankton (crustaceans) are capable of utilizing ciliates as a food source, but there is little direct evidence from field studies documenting this trophic link. Ciliates can be voracious grazers of both bacterioplankton and phytoplankton, and each species has a distinct range of preferred particle size which is a function of both mouth size and morphology. Myxotrophic ciliates may be important components in some plankton communities, particularly during periods of nutrient limitation or after their displacement from the benthos of eutrophic lakes. Evidence regarding the importance of planktonic ciliated protozoa in nutrient regeneration and as intermediaries in energy flow is discussed. 相似文献

The application of high hydrostatic pressure is an effective tool to promote dissolution and refolding of protein from aggregates and inclusion bodies while minimizing reaggregation. In this study we explored the mechanism of high-pressure protein refolding by quantitatively assessing the magnitude of the protein-protein interactions both at atmospheric and elevated pressures for T4 lysozyme, in solutions containing various amounts of guanidinium hydrochloride. At atmospheric pressure, the protein- protein interactions are most attractive at moderate guanidinium hydrochloride concentrations (approximately 1-2 molar), as indicated by a minimum in B(22) values. In contrast, at a pressure of 1,000 bar no minimum in B(22) values is observed, indicating that high pressures colloidally stabilize protein against aggregation. Finally, experimental values of refractive index increments as a function of pressure indicate that at high pressures, wetting of the hydrophobic surfaces is favored, resulting in a reduction of the hydrophobic effect. This reduction in the hydrophobic effect reduces the driving force for aggregation of (partially) unfolded protein. 相似文献

Developments in digital imaging and fluorescent microscopy provide a new method and opportunities for quantification of protein expression in human tissue. Archived collections of paraffin-embedded tumors can be used to study the relationship between quantitative differences in protein expression in tumors and patient outcome. In this report we describe the use of a DeltaVision Restoration deconvolution microscope, combined with fluorescent immunohistochemistry, to obtain reproducible and quantitative estimates of protein expression in a formalin-fixed paraffin-embedded tissue. As proof of principle, we used antibodies to the estrogen and progesterone receptors in a hormone receptor-positive breast cancer specimen. We provide guidelines for control of day-to-day variability in camera and microscope performance to ensure that image acquisition leads to reproducible quantitative estimates of protein expression. We show that background autofluorescence related to formalin fixation can be controlled and that for proteins that are expressed in nearly every cell, multiplexing two primary antibodies on the same slide does not significantly affect the results obtained. We demonstrate that for proteins whose expression varies markedly from cell to cell, data reproducibility, as assessed by imaging successive tissue sections, is more difficult to determine. 相似文献

Utilizing a historical database to refine ground cover vegetation as indicators of wetland hydrology
Indicator species provide an easy and quick method of evaluating ecosystems. The species comprising the most useful indicators of wetlands should be distributed across a range of water depths and inundation durations, while each species is representative of a specific condition. Hydrophytic vegetation is commonly used to determine the existence and type of wetland; however, such indicator systems often depend on assigning species qualitatively to discrete categories based on assumptions about their distribution along a gradient of conditions. The current study proposes a wetland indicator system based on the quantitative responses of individual vegetation species to a gradient of water depths and periods of inundation. A long-term database was utilized to determine species responses to hydrological alterations in a series of wetlands. The hydrophytic plant species investigated (n = 29) displayed relatively narrow ranges of mean hydrologic values and were distributed linearly along multiple hydrologic gradients (hydroperiod, average water depth, and maximum water depth) ranging from Amphicarpum muhlenbergianum which was observed at the shallowest water depths and shortest hydroperiod to Pontederia cordata and Ludwigia repens which were characteristic of wetlands with the deepest water and longest hydroperiod. The species distribution and means along the hydrologic gradients tested indicates they are prime candidates for inclusion in a quantitative or continuum indicator system. The historical database utilized for this study provided valuable information for numerous species common to the Tampa Bay region for which little or no ecological information was previously available. The methodology utilized in this paper provides a cost and time effective method for obtaining the vast amounts of information required to refine plant indicator systems using a large number of species. 相似文献

Meprins are metalloendopeptidases expressed by leukocytes in the lamina propria of the human inflamed bowel, that degrade extracellular matrix proteins in vitro implicating them in leukocyte transmigration events. The aims of these studies were to 1) examine the expression of meprins in the mouse mesenteric lymph node, 2) determine whether macrophages express meprins, and 3) determine whether deletion of the meprin beta gene (Mep-1beta) mitigated the ability of leukocytes to disseminate through extracellular matrix in vitro. These studies show that meprin alpha and beta are expressed in leukocytes of the mouse mesenteric lymph node, and meprin alpha, but not beta, decreased during intestinal inflammation. Deletion of Mep-1beta gene decreased the ability of leukocytes to migrate through matrigel compared with wild-type leukocytes. Meprin beta, but not alpha, was detected in cortical and medullary macrophages of the lymph node. Thus overall, meprin beta is expressed by leukocytes in the draining lymph node of the intestine, regardless of the inflammatory status of the animal, and is likely to contribute to leukocyte transmigration events important to intestinal immune responses. Thus, the expression of meprins by leukocytes of the intestinal immune system may have important implications for diseases such as inflammatory bowel diseases, which are aggravated by leukocyte infiltration. 相似文献

Exocytosis is a vesicle fusion process driven by soluble N‐ethylmaleimide‐sensitive factor attachment protein receptors (SNAREs). A classic exocytic pathway is insulin‐stimulated translocation of the glucose transporter type 4 (GLUT4) from intracellular vesicles to the plasma membrane in adipocytes and skeletal muscles. The GLUT4 exocytic pathway plays a central role in maintaining blood glucose homeostasis and is compromised in insulin resistance and type 2 diabetes. A candidate regulator of GLUT4 exocytosis is tomosyn, a soluble protein expressed in adipocytes. Tomosyn directly binds to GLUT4 exocytic SNAREs in vitro but its role in GLUT4 exocytosis was unknown. In this work, we used CRISPR‐Cas9 genome editing to delete the two tomosyn‐encoding genes in adipocytes. We observed that both basal and insulin‐stimulated GLUT4 exocytosis was markedly elevated in the double knockout (DKO) cells. By contrast, adipocyte differentiation and insulin signaling remained intact in the DKO adipocytes. In a reconstituted liposome fusion assay, tomosyn inhibited all the SNARE complexes underlying GLUT4 exocytosis. The inhibitory activity of tomosyn was relieved by NSF and α‐SNAP, which act in concert to remove tomosyn from GLUT4 exocytic SNAREs. Together, these studies revealed an inhibitory role for tomosyn in insulin‐stimulated GLUT4 exocytosis in adipocytes. We suggest that tomosyn‐arrested SNAREs represent a reservoir of fusion capacity that could be harnessed to treat patients with insulin resistance and type 2 diabetes. 相似文献
