The two mammalian neuropeptides substance P (SP) and neurokinin A (NKA) have been demonstrated to stimulate DNA synthesis in connective tissue cells, suggesting that peripheral neurons may play a role in development and tissue regeneration. In this study we have tried to identify intracellular messengers required for SP- and NKA-induced DNA synthesis. SP and NKA, as well as platelet-derived growth factor (PDGF) stimulated formation of inositol phosphates in smooth muscle cells (SMC), whereas no effect on inositol phosphates formation occurred in response to nonmitogenic neuropeptides. Pretreatment of the cells with pertussis toxin markedly decreased DNA synthesis induced by NKA. This toxin inhibits formation of inositol phosphates by acting on a regulatory G-protein. Calcium and calmodulin antagonists also inhibited NKA-induced DNA synthesis. These results imply that the mitogenic signal(s) produced by activated neuropeptide receptors involves formation of inositol phosphate and activation of a calcium/calmodulin dependent process. We further report that other neuropeptides occurring in peripheral neurons, i.e., vasoactive intestinal polypeptide, calcitonin gene-related peptide, neuropeptide Y, somatostatin, or cholecystokinin, are without growth-stimulatory effect on cultured SMC.  相似文献

The recognition of a pathogen or a vaccine antigen formulation by cells in the innate immune system leads to production of proinflammatory cytokines, which will determine the ensuing acquired immune response quantitatively and qualitatively. Tumour necrosis factor (TNF)-alpha, interleukin (IL)-1 and IL-6 are the first set of cytokines produced upon such an encounter, which have roles both in protective immunity and immunopathogenesis evident with respiratory syncytial virus (RSV). RSV antigens in different physical adjuvant-vaccine formulations were analysed for their capacity to provoke cultured murine peritoneal cells to produce these three proinflammatory cytokines. RSV immunostimulating complex (ISCOM), i.e. both antigen and adjuvant are incorporated in the same particle, induced high levels of IL-1alpha being of the same magnitude or higher than those of live RSV and lipopolysaccharide (LPS). Live virus and LPS induced higher levels of IL-6 and TNF-alpha than ISCOM and so did non-adjuvanted UV-inactivated RSV but only at high doses. ISCOM-Matrix, i.e. ISCOM without antigens, admixed as a separate entity to inactivated RSV, downregulated or blocked the cytokine response to the inactivated RSV in contrast to ISCOM. Kinetic studies showed that ISCOM induced cytokine production first detected at hours 1, 2, 4 for TNF-alpha, IL-6 and IL-1alpha respectively, which was earlier than for the other antigen formulations containing corresponding doses of antigen and/or Quillaja adjuvant. Peak values for production of TNF-alpha and IL-6 were at 8 h and for IL-1alpha at 72 h following stimulation with ISCOM. The delayed appearance of IL-1alpha may reflect the cell-bound nature of this cytokine.  相似文献

Summary Resource utilization by cutthroat trout (CT) and Dolly Varden charr (DV) was studied 8 years after experimental transfers from sympatry had established reproducing allopatric populations in two nearby fishless lakes. Allopatric DV significantly increased their utilization of shallow-dwelling zoobenthos, and increased their vertical distribution in comparison to that in sympatry. In contrast, allopatric CT showed little change in the proportions of major prey types utilized, and, if anything, restricted their vertical distribution in comparison to that in sympatry. The results can be explained by the hypothesis that the resource use of DV is strongly influenced by interspecific competition from CT, whereas CT largely remains unaffected by this interaction. An alternative hypothesis, that lake differences can explain the differences in resource use between sympatry and allopatry, was evaluated by comparing food resource availability and other biotic and abiotic characteristics of the three study lakes. None of these could account for the shift in resource use by DV between sympatry and allopatry, but lake differences may explain why allopatric CT showed a restricted habitat use in comparison with their sympatric donor stock. The results of this whole-lake transfer experiment are consistent with earlier reported field and laboratory studies, and suggest that the aggressive dominance of CT is the most important mechanism by which DV are displaced from littoral and near-surface habitats in sympathy with CT.  相似文献

Labelled adenine, noradrenaline (NA), and gamma-aminobutyric acid (GABA) were taken up by the transversely cut hippocampal slice. [3H]NA and [14C]GABA were retained as such, [3H]- (or [14C]-) adenine mainly as adenine nucleotides. There was a spontaneous overflow of all three types of compounds ranging from 0.1 (GABA) to 0.21 (NA) %/min. The rate of [3H]NA overflow increased rapidly during electrical field stimulation. The release rate was well maintained over a 15-min period. The rate of [14C]GABA release also increased rapidly but it was not maintained over a 15-min period even if uptake and/or metabolism was inhibited by nipecotic acid (1 mM) and aminooxyacetic acid (AOAA, 0.1 mM). The bulk of the purines was released after the stimulation period. For all compounds the amounts released were frequency- and calcium-dependent. At a frequency of 3 Hz a 10 V stimulation was sufficient to cause a maximal [3H]NA release and 20 V to cause maximal [14C]GABA release, but 14C-purine release was increased further by increasing the voltage to 40 V. The evoked purine release was inhibited by a nucleoside uptake inhibitor (dipyridamole). On stimulation of [3H]NA-labelled slices the released radioactivity was composed of greater than 95% unchanged NA. The specific activities of NA in the slice and in the superfusate were practically identical. In [3H]adenine-labelled slices the released radioactivity was composed of adenosine, inosine, and hypoxanthine, but the activity in the slice of ATP, ADP, and AMP.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Summary Many fish populations have both resident and migratory individuals. Migrants usually grow larger and have higher reproductive potential but lower survival than resident conspecifics. The decision about migration versus residence probably depends on the individual growth rate, or a physiological process like metabolic rate which is correlated with growth rate. Fish usually mature as their somatic growth levels off, where energetic costs of maintenance approach energetic intake. After maturation, growth also stagnates because of resource allocation to reproduction. Instead of maturation, however, fish may move to an alternative feeding habitat and their fitness may thereby be increased. When doing so, maturity is usually delayed, either to the new asymptotic length, or sooner, if that gives higher expected fitness. Females often dominate among migrants and males among residents. The reason is probably that females maximize their fitness by growing larger, because their reproductive success generally increases exponentially with body size. Males, on the other hand, may maximize fitness by alternative life histories, e.g. fighting versus sneaking, as in many salmonid species where small residents are the sneakers and large migrants the fighters. Partial migration appears to be partly developmental, depending on environmental conditions, and partly genetic, inherited as a quantitative trait influenced by a number of genes.  相似文献

One dominating peptic phosphopeptide, Asx-Thr-Lys-Gly-Pro-Glx-Ile-Glx-Thr-Gly-Val-Leu-Arg-Arg-Ala-(32P)SerP-Val-Ala-Glx-Leu, was obtained from rat liver pyruvate kinase (type L) phosphorylated by cyclic 3′,5′-AMP-stimulated protein kinase from the same tissue. The sequence around the phosphorylated serine residue is similar to that of a corresponding but smaller peptic phosphopeptide previously isolated from pig liver (type L) pyruvate kinase, Leu-Arg-Arg-Ala-(32P)SerP-Leu.  相似文献
