The shoots developed from both the shoot tip and nodal explants of feathered amaranth (Celosia argentea var. plumosa—feathered cockscomb or plumed cockscomb) after 8 weeks of culture in the presence of either paclobutrazol or benzyladenine
(BA) were shorter than those developed on basal Murashige and Skoog (MS) medium (Physiol Plant, 15:473–497, 1962) alone. However, this retarding effect was more pronounced in the nodal explant culture. Shoot tip explants from 2-week-old
seedlings were more adversely affected by 0.85 or 1.7 μM paclobutrazol than those from older seedlings. In contrast, regardless
of preculture duration investigated nodal explants did not exhibit different response to three different concentrations of
paclobutazol. The response to 2.2 or 4.4 μM BA appeared to be largely independent of the age of the shoot tip explants or
preculture treatment of nodal explants. Shoots developed from nodal explants produced a higher number of terminal inflorescence
than those from shoot tip explants. Moreover, only lateral shoots from nodal explant culture formed inflorescence. Increased
preculture duration on basal MS medium could generally lessen the inhibitory effect of lower concentrations of paclobutazol
or BA on terminal or lateral inflorescence formation in nodal explant culture. 相似文献

Summary Plantlets of Capsicum annuum L. ev. Sweet Banana regenerated via somatic embryogenesis from immature zygotic embryos were capable of producing flower,
fruit, and seed when cultured in small tissue culture containers. In vitro floral buds were first formed on plantlets that grew on plantlet development medium [agar-gelled Murashige and Skoog (MS)
basal medium containing 1 mgl−1 (5.3 μM) α-naphthaleneacetic acid (NAA)] in a growth room at 22°C and continuous illumination. However, floral buds rarely developed
further into mature flowers. This problem was overcome using the vented autoclavable plant tissue culture containers. In vitro fruit formation and ripening was observed when liquid half-strength MS basal medium supplemented with 5 μg ml−1 silver thiosulfate, 1 mg l−1 (5.3 μM) NAA, and 3% sucrose was added to the surface of the plantlet development medium. Hand-pollination improved fruit set. Further
research in needed to determine why the pepper seeds formed in vitro failed to germinate. 相似文献

We used four cultivars ofCapsicum annuum L.—Sweet Banana, California Wonder, Yolo Wonder, and Ace—to reexamine the critical factors influencing somatic embryogenesis
from zygotic embryo explants, as reported in the literature. When we followed the protocol of Buyukalaca and Mavituna (1996),
which had induced somatic embryogenesis from mature zygotic embryos of cv. Ace, only callus was formed without embryogenesis
from our mature zygotic embryo expiants. Using the procedures of Harini and Lakshmi Sita (1993) and Binzel et al. (1996),
with some modifications, we were able to induce somatic embryogenesis in all four cultivars. Rates of conversion were significantly
reduced, from 75% and 65% to 40% and 28% in ’Sweet Banana’ and ’California Wonder’, respectively, when the immature zygotic
embryo expiants were held on the induction medium for longer than two weeks. Likewise, somatic embryogenesis of ’Yolo Wonder’
was not observed if the induction medium was supplemented with 10% glucose or fructose, or without 10% sucrose. For somatic
embryo induction and eventual plantlet conversion in Yolo Wonder’, maltose could adequately replace sucrose. In all four cultivars,
somatic embryos were initiated from immature zygotic explants on media with or without coconut water, under both light and
