Bacillus methylotrophicus M4-96 is a beneficial rhizobacterium that has been isolated from the rhizosphere of maize (Zea mays). In this study, we investigated its efficacy as a plant growth promoter for strawberry in vitro, as well as its ability to induce callose deposition in leaves to reduce the severity of Botrytis cinerea infection. Two methods of plant-bacterial interaction were used: inoculation near the root and emission of volatile compounds with no physical contact. Plant biomass increased under both treatments, but with developmental parameters of the plants differentially stimulated by each method. Root inoculation increased petiole number and root length, whereas bacterial volatiles increased petiole length and root number. A chemical analysis of the bacterial culture revealed the presence of indole acetic acid (0.21 μg L−1) and gibberellic acid (6.16 μg L−1). Acetoin was previously identified as the major volatile produced by the bacteria, and its application to strawberry explants increased their growth and development. Furthermore, when acetoin and both phytoregulators were added to the culture media, these positive effects were enhanced. The inoculation method also affected the size and quantity of callose deposits in the leaves. Treatment with volatiles increased callose deposition in the leaves by up to five-fold, which promoted a rapid defense reaction that inhibited the incidence of gray mold by reinforcing cell wall. Taken together, our results show that B. methylotrophicus M4-96 promotes growth and induces systemic resistance in strawberry plants.

GCC185 is a large coiled-coil protein at the trans Golgi network that is required for receipt of transport vesicles inbound from late endosomes and for anchoring noncentrosomal microtubules that emanate from the Golgi. Here, we demonstrate that recruitment of GCC185 to the Golgi is mediated by two Golgi-localized small GTPases of the Rab and Arl families. GCC185 binds Rab6, and mutation of residues needed for Rab binding abolishes Golgi localization. The crystal structure of Rab6 bound to the GCC185 Rab-binding domain reveals that Rab6 recognizes a two-fold symmetric surface on a coiled coil immediately adjacent to a C-terminal GRIP domain. Unexpectedly, Rab6 binding promotes association of Arl1 with the GRIP domain. We present a structure-derived model for dual GTPase membrane attachment that highlights the potential ability of Rab GTPases to reach binding partners at a significant distance from the membrane via their unstructured and membrane-anchored, hypervariable domains.  相似文献

Several authors have discussed whether Peromyscus furvus is a monotypic species rather than a polytypic entity, that it includes more than one species. Here, we analyze these questions by means of traditional morphometrics and by genetic analyses using ND3-ND4 mtDNA genes as markers. In spite of a generalized overlap of the measurable characters among populations, our analyses show that the northernmost populations, which was assignable to P. latirostris, consistently show larger dimensions overall. The amount of genetic differentiation revealed by our molecular data, support conclusive evidence to suggest this taxon is a valid species. Our results also disclose that morphometric and molecular segregation between P. furvus and P. angustirostris is still incomplete. Finally, the two populations from the state of Oaxaca showed more morphometric affinity with those attributable to P. furvus and revealed a discrete degree of genetic differentiation. Nevertheless, their systematic position is not clear yet.  相似文献

Little is known about the natural history of the Sphaerodactylus species endemic to the three islands located in the Mona Passage separating the Greater Antillean islands of Hispaniola and Puerto Rico. In this study, parts of two mitochondrial genes, 16S rRNA and 12S rRNA, were sequenced to determine the relationships between the sphaerodactylids that live in the Mona Passage and other Caribbean species from the same genus. While the main goal was to identify the biogeographical origin of these species, we also identified a genetically distinct type of dwarf gecko that warrants future evaluation as a possible new species. According to the reconstructed phylogenies, we propose a stepwise model of colonization wherein S. nicholsi from southwestern Puerto Rico or a very close ancestor gave rise through a founder event to Sphaerodactylus monensis on Mona Island. In a similar fashion, S. monensis or a very close ancestor on Mona Island gave rise to S. levinsi on Desecheo Island. This study also suggests that the most recent common ancestor between the species from the islands in the Mona Passage and Puerto Rico existed approximately 3 MYA.  相似文献

The importance of climatic conditions in shaping the geographic distribution of amphibian species is mainly associated to their high sensitivity to environmental conditions. How they cope with climate gradients through behavioral adaptations throughout their distribution is an important issue due to the ecological and evolutionary implications for population viability. Given their low dispersal abilities, the response to seasonal climate changes may not be migration, but behavioral and physiological adaptations. Here we tested whether shifts in climatic seasonality can predict the temporal variation of surface activity of the fossorial Lowland Burrowing Treefrog (Smilisca fodiens) across its geographical distribution. We employed Ecological Niche Modeling (ENM) to perform a monthly analysis of spatial variation of suitable climatic conditions (defined by the July conditions, the month of greatest activity), and then evaluated the geographical correspondence of monthly projections with the occurrence data per month. We found that the species activity, based on the species'' occurrence data, corresponds with the latitudinal variation of suitable climatic conditions. Due to the behavioral response of this fossorial frog to seasonal climate variation, we suggest that precipitation and temperature have played a major role in the definition of geographical and temporal distribution patterns, as well as in shaping behavioral adaptations to local climatic conditions. This highlights the influence of macroclimate on shaping activity patterns and the important role of fossorials habits to meet the environmental requirements necessary for survival.  相似文献

The Archaic Diet in Mesoamerica: Incentive for Milpa Development and Species Domestication. One of the central questions in the development of Mesoamerican civilization is how the alimentary, agronomic, and ecological complementarities were achieved within the milpa agroecosystem, which is one of its more important and distinctive cultural elements. In the Mesoamerican center of origin of agriculture and domestication of plants, located in western Mexico, we inquired among Náhuatl communities about the ancient dishes prepared with wild plants that are part of their ancient foodways, and the tools and technology used to prepare them. We found that the wild progenitors of Agave spp., Zea mays L, Cucurbita argyrosperma Hort. Ex L.H. Bayley, Phaseolus spp., Capsicum annum L., Solanum lycopersicum L., Physalis phyladelphica Lam, Spondias purpurea L., Persea americana Mill., and Hyptis suaveolens (L.) Poit are consumed in dishes that remain in the present food culture of the poor peasants, and are prepared with techniques and tools that were available in the Archaic period: Sun drying, roasting, toasting, baking, cracking, grinding, crushing, fermenting, and soaking in plain water or in water with ash, using three–stone fireplaces, stone toasters, crushers, grinders, rock pits, and three types of earth ovens. A remarkable finding was that beans could be incorporated into the diet without boiling, but just by toasting, stone grinding, and baking in corn dough tamales. Results obtained suggest that the basic Mesoamerican diet could have been shaped before the species involved were domesticated. Its nutritional complementarity since the Archaic period could have been one of the incentives for the development of the milpa system and the domestication of its species, achieving in this way also their ecological and agronomical complementarity.  相似文献
