Engineering of a multifunctional hemicellulase   总被引:1,自引:0,他引:1  
To engineer a multifunctional xylan-degrading enzyme, a chimera was created by fusing the xylanase domain of the Clostridium thermocellum xylanase (xynZ) and a dual functional arabinofuranosidase/xylosidase (DeAFc; from a compost starter mixture) through a flexible peptide linker. The xylanase domain of xynZ possesses previously unreported endoglucanase activity. The chimera, possessing the activities of xylanase, endoglucanase, arabinofuranosidase and xylosidase, was expressed in E. coli and purified. The chimera closely resembled the parental enzymes in pH, temperature optima and kinetics, and was more active than the parental enzyme mixture in the hydrolysis of natural xylans and corn stover. Summary and conclusions S.paracitrovorus does not readily dissimilate citric acid in the absence of sugar but does attack citric acid relatively vigorously in the presence of small quantities of glucose or lactose. The effect of glucose and lactose in initiating the dissimilation of citric acid is catalytic.The sugars which act catalytically are themselves fermented to approximately equimolar quantities of carbon dioxide, ethyl alcohol and lactic acid. The dissimilation of a combined substrate of citrate and glucose forms, in addition, acetic acid, acetylmethylcarbinol, 2,3-butylene glycol and under certain conditions, pyruvic acid which acts as an intermediate compound. Pyruvate is dissimilated to products similar to those from a fermentation of citrate plus glucose.The reactions ofKrebs' citric acid cycle apparently do not apply to the dissimilation of citric acid byS.paracitrovorus because the fermentation of citric acid proceeds anaerobically, consumes little oxygen aerobically and is not inhibited by arsenite.Inasmuch as milk contains lactose, the fermentation of citric acid in milk byS.paracitrovorus may be catalyzed as shown in these studies.Journal paper No.J711 of the Iowa Agricultural Experiment Station, Project 451.  相似文献

In Zygorhynchus moelleri, a homothallic Mucor, triglycendes are the main components of chloroform/methanol extractable lipids. The triglycerides accumulate in the aerial hyphae, particularly in the developing zygospores and in the lateral suspensors, but only after zygospore maturation. They are probably transported from the submerged mycelium to the aerial hyphae. Most of the fatty acid synthetase activity is found in the submerged mycelium. The fatty acid composition of the triglycerides does not change appreciably during sexual development. No influence of trisporic acids has been found on triglyceride synthesis or transport.  相似文献

Tryptic digestion followed by identification using mass spectrometry is an important step in many proteomic studies. Here, we describe the preparation of immobilized, acetylated trypsin for enhanced digestion efficacy in integrated protein analysis platforms. Complete digestion of cytochrome c was obtained with two types of modified-trypsin beads with a contact time of only 4 s, while corresponding unmodified-trypsin beads gave only incomplete digestion. The digestion rate of myoglobin, a protein known to be rather resistant to proteolysis, was not altered by acetylating trypsin and required a buffer containing 35% acetonitrile to obtain complete digestion. The use of acetylated-trypsin beads led to fewer interfering tryptic autolysis products, indicating an increased stability of this modified enzyme. Importantly, the modification did not affect trypsin's substrate specificity, as the peptide map of myoglobin was not altered upon acetylation of immobilized trypsin. Kinetic digestion experiments in solution with low-molecular-weight substrates and cytochrome c confirmed the increased catalytic efficiency (lower K(M) and higher k(cat)) and increased resistance to autolysis of trypsin upon acetylation. Enhancement of catalytic efficiency was correlated with the number of acetylations per molecule. The favorable properties of the new chemically modified trypsin reactor should make it a valuable tool in automated protein analysis systems.  相似文献

Near-isogenic lines (NILs) differing with regard to disease QTLs provide valuable material for a more detailed study into the genetic basis of quantitative resistance. Previously obtained information on QTLs that show an effect on leaf rust (Puccinia hordei) in barley was used in a marker-assisted backcross programme. The genome origin in backcross plants was controlled through AFLP marker analysis and graphical genotyping. Plants obtained after the third generation of backcrossing sufficiently resembled the recurrent parent. For one QTL, BC3S1 plants were evaluated in a disease test and genotyped. NILs containing the desired QTL in homozygous condition in a recipient background were finally obtained. A disease test and verification of the marker genotype confirmed the identity of the NILs. Simultaneous with the backcross programme a simulation study on efficiency of marker-assisted backcrossing was performed.  相似文献
