Ordered water structure in an A-DNA octamer at 1.7 A resolution   总被引:5,自引:0,他引:5  
The crystal structure of the deoxyoctamer d(G-G-Br U-A-BrU-A-C-C) was refined to a resolution of 1.7 A using combined diffractometer and synchrotron data. The analysis was carried out independently in two laboratories using different procedures. Although the final results are identical the comparison of the two approaches highlights potential problems in the refinement of oligonucleotides when only limited data are available. As part of the analysis the positions of 84 solvent molecules in the asymmetric unit were established. The DNA molecule is highly solvated, particularly the phosphate-sugar back-bone and the functional groups of the bases. The major groove contains, in the central BrU-A-BrU-A region, a ribbon of water molecules forming closed pentagons with shared edges. These water molecules are linked to the base O and N atoms and to the solvent chains connecting the O-1 phosphate oxygen atoms on each strand. The minor groove is also extensively hydrated with a continuous network in the central region and other networks at each end. The pattern of hydration is briefly compared with that observed in the structure of a B-dodecamer.  相似文献

Summary During locomotion, stick insectsCarausius morosus, place the tarsus of the rear leg near the tarsus of the ipsilateral middle leg, whatever the position of the latter. This adjustment by the hind leg requires that it receive information on the actual position of the middle leg tarsus. It is shown by ablation experiments that such information is contributed by the following proprioceptors of the middle leg: the ventral and dorsal coxal hairplates, the coxal hair rows, the trochanteral hairplate and the femoral chordotonal organ. Additional information comes from other, as yet unidentified, sense organs. Several alternatives are considered to explain how the signals from the diverse sense organs of the subcoxal joint might be combined in computing the target position for the protracting hind leg. In the experiments presented here adult stick insects (Carausius morosus) walk on a treadwheel with various legs standing on platforms fixed relative to the body of the insect. These standing legs produce large forees directed towards the rear which are modulated in the rhythm of the walking legs. Neighbouring legs which both stand on a platform often oscillate in phase. The Okavango River, in semi-arid northwestern Botswana, flows for over 400 km in a pristine wetland developed on a large (>22,000 km2) alluvial fan (Okavango Delta). An annual flood pulse inundates the floodplains of the wetlands and travels across the Delta in 4–6 months. In this study, we assess the effects of long hydraulic residence time, variable hydrologic interaction between river–floodplain–wetland and evapotranspiration on carbon cycling. We measured dissolved inorganic carbon (DIC) concentrations and stable carbon isotopes of DIC (δ13CDIC) from river water when the Delta was not flooded (low water) and during flooding (high water). During low water, the average DIC concentration was 31 % higher and the δ13CDIC 2.1 ‰ more enriched compared to high water. In the lower Delta with seasonally flooded wetlands, the average DIC concentration increased by 70 % during low water and by 331 % during high water compared to the Panhandle with permanently flooded wetlands. The increasing DIC concentration downriver is mostly due to evapoconcentration from transpiration and evaporation with increased transit time. The average δ13CDIC between low and high water decreased by 3.7 ‰ in the permanently flooded reaches compared to an increase of 1.6 ‰ in the seasonally flooded reaches. The lower δ13CDIC during high water in the permanently flooded reaches suggest that DIC influx from the floodplain-wetland affects river’s DIC cycling. In contrast, higher river channel elevations relative to the wetlands along seasonal flooded reaches limit hydrologic interaction and DIC cycling occurs mostly by water column processes and river-atmospheric exchange. We conclude that river-wetlands interaction and evapoconcentration are important factors controlling carbon cycling in the Okavango Delta.  相似文献

Lymphatic mapping and sentinel lymph node biopsy is a new technique used in the surgical treatment of patients with malignant melanoma. The purpose of this study was to evaluate the results of this approach for patients with melanoma of the lower extremity. Between May of 1994 and June of 1997 at the H. Lee Moffitt Cancer Center and Research Institute, 85 consecutive patients with clinical stage I and II melanoma of the lower extremity underwent lymphatic mapping and sentinel lymph node biopsy. These nodes were identified in all 85 patients by intraoperative lymphatic mapping with both radiolymphoscintigraphy and a vital blue dye injection. Eleven patients (12.9 percent) had histologically positive sentinel lymph nodes, and 10 patients underwent inguinal complete lymph node dissections. All 10 patients had no further histologically positive lymph nodes confirmed by subsequent complete dissection. Among 74 patients with histologically negative sentinel lymph nodes, only 2 patients (2.7 percent) developed inguinal nodal metastases during a mean follow-up period of 21.8 months (range, 13.5 to 58.3 months). The sensitivity of lymphatic mapping and sentinel lymph node biopsy in this series was 100 percent and the specificity was 97.3 percent. Therefore, we conclude that the use of lymphatic mapping and sentinel lymph node biopsy can accurately stage patients with melanoma of the lower extremity and provide a rational surgical approach for these patients.  相似文献

Walking in insects and most six-legged robots requires simultaneous control of up to 18 joints. Moreover, the number of joints that are mechanically coupled via body and ground varies from one moment to the next, and external conditions such as friction, compliance and slope of the substrate are often unpredictable. Thus, walking behaviour requires adaptive, context-dependent control of many degrees of freedom. As a consequence, modelling legged locomotion addresses many aspects of any motor behaviour in general. Based on results from behavioural experiments on arthropods, we describe a kinematic model of hexapod walking: the distributed artificial neural network controller walknet. Conceptually, the model addresses three basic problems in legged locomotion. (I) First, coordination of several legs requires coupling between the step cycles of adjacent legs, optimising synergistic propulsion, but ensuring stability through flexible adjustment to external disturbances. A set of behaviourally derived leg coordination rules can account for decentralised generation of different gaits, and allows stable walking of the insect model as well as of a number of legged robots. (II) Second, a wide range of different leg movements must be possible, e.g. to search for foothold, grasp for objects or groom the body surface. We present a simple neural network controller that can simulate targeted swing trajectories, obstacle avoidance reflexes and cyclic searching-movements. (III) Third, control of mechanically coupled joints of the legs in stance is achieved by exploiting the physical interactions between body, legs and substrate. A local positive displacement feedback, acting on individual leg joints, transforms passive displacement of a joint into active movement, generating synergistic assistance reflexes in all mechanically coupled joints.  相似文献

Stachybotrys chartarum has received much attention as a possible cause of sick-building syndrome. Because morphological species recognition in fungi can hide diversity, we applied a phylogenetic approach to search for cryptic species. We examined 23 isolates from the San Francisco Bay Area, and another seven from around the US. Using markers we developed for three polymorphic protein coding loci (chitin synthase 1, beta-tubulin 2, and trichodiene synthase 5), we infer that two distinct phylogenetic species exist within the single described morphological species. We have found no correlation between genetic isolation and geographic distance.  相似文献

Background and Objectives

Desmoplastic melanoma is a unique subtype of melanoma which typically affects older patients who often have comorbidities that can adversely affect survival. We sought to identify melanoma-specific factors influencing survival in patients with desmoplastic melanoma.

Methods

Retrospective review from 1993 to 2011 identified 316 patients with primary desmoplastic melanoma. Clinicopathologic characteristics were correlated with nodal status and outcome.

Results

Fifty-five patients (17.4%) had nodal disease: 33 had a positive sentinel lymph node biopsy and 22 developed nodal recurrences (no sentinel lymph node biopsy or false-negative sentinel lymph node biopsy). Nodal disease occurred more often in younger patients and in cases with mixed compared with pure histology (26.7% vs. 14.6%); both of these variables significantly predicted nodal status on multivariable analysis (p<0.05). After a median follow-up of 5.3 years, recurrence developed in 87 patients (27.5%), and 111 deaths occurred. The cause of death was known in 79 cases, with 47 deaths (59.5%) being melanoma-related. On multivariable analysis, Breslow thickness, mitotic rate ≥1/mm2 and nodal status significantly predicted melanoma-specific survival (p<0.05).

Conclusions

Nodal status predicts melanoma-specific survival in patients with desmoplastic melanoma. However, since patients with desmoplastic melanoma represent an older population, and a considerable proportion of deaths are not melanoma-related (40.5%), comorbidities should be carefully considered in making staging and treatment decisions in this population.  相似文献
