Fungal immunomodulatory proteins (FIPs) found in a wide variety of mushrooms hold significant therapeutic potential. Despite much research, the structural determinants for their immunomodulatory functions remain unknown. In this study, a DNA shuffling technique was used to create two shuffled FIP protein libraries: an intrageneric group containing products of shuffling between FIP-glu (FIP gene isolated from Ganoderma lucidum) and FIP-gsi (FIP gene isolated from Ganoderma sinense) genes and an intergeneric group containing the products of shuffling between FIP-glu, FIP-fve (FIP gene isolated from Flammulina velutipes), and FIP-vvo (FIP gene isolated from Volvariella volvacea) genes. The gene shuffling generated 426 and 412 recombinant clones, respectively. Using colony blot analysis, we selected clones that expressed relatively high levels of shuffled gene products recognized by specific polyclonal antibodies. We analyzed the DNA sequences of the selected shuffled genes, and testing of their protein products revealed that they maintained functional abilities to agglutinate blood cells and induce cytokine production by splenocytes from Kunming mice in vitro. Meanwhile, the relationships between protein structure and the hemagglutination activity and between the changed nucleotide sites and expression levels were explored by bioinformatic analysis. These combined analyses identified the nucleotide changes involved in regulating the expression levels and hemagglutination activities of the FIPs. Therefore, we were able to generate recombinant FIPs with improved biological activities and expression levels by using DNA shuffling, a powerful tool for the generation of novel therapeutic proteins and for their structural and functional studies.  相似文献

查尔酮合酶(chalcone synthase, CHS)是植物类黄酮化合物合成的关键酶,有关蕨类植物CHS基因的序列及功能信息尚不完善。本研究采用快速扩增cDNA末端(RACE)技术克隆获得了模式蕨类植物——水蕨(Ceratopteris thalictroides)CtCHS基因(GenBank登录号:JX027616.1),其cDNA序列全长为1616 bp,具有3个外显子和2个内含子,开放阅读框(ORF)为1215 bp,编码404个氨基酸。进化树分析表明,CtCHS与问荆(Equisetum arvense)、松叶蕨(Psilotum nudum)和3种薄囊蕨的查尔酮合成酶基因聚为一枝,说明这些蕨类植物亲缘关系较近且为单系起源。通过构建原核表达体系成功获得CtCHS蛋白的多克隆抗体并用于免疫印迹分析,结果表明CtCHS基因的表达明显受紫外光(UV)诱导。CtCHS基因的克隆与表达分析为进一步研究水蕨类黄酮化合物的合成及其调控机制提供了依据。  相似文献

Copper (Cu(II)) and nickel (Ni(II)) are often encountered in wastewaters. This study investigated the individual toxic effects of long-term addition of Cu(II) and Ni(II) on the biochemical properties of aerobic granules in sequencing batch reactors (SBRs). The biochemical properties of aerobic granules were characterized by extracellular polymeric substances (EPS) content, dehydrogenase activity, microbial community biodiversity, and SBR performance. One SBR was used as a control system, while another two received respective concentration of Cu(II) and Ni(II) equal to 5 mg/L initially and increased to 15 mg/L on day 27. Results showed that the addition of Cu(II) drastically reduced the biomass concentration, bioactivity, and biodiversity of aerobic granules, and certainly deteriorated the treatment performance. The toxic effect of Ni(II) on the biodiversity of aerobic granules was milder and the aerobic granular system elevated the level of Ni(II) toxicity tolerance. Even at a concentration of 15 mg/L, Ni(II) still stimulated the biomass yield and bioactivity of aerobic granules to some extent. The elevated tolerance seemed to be owed to the concentration gradient developed within granules, increased biomass concentration, and promoted EPS production in aerobic granular systems.  相似文献

We have performed systematic spectroscopic titrations to characterize the binding reaction of cationic meso-tetrakis(4-(N-methylpyridiumyl))porphyrin (TMPyP4) with the G-quadruplex (G4) of human telomeric single-strand oligonucleotide d[TAGGG(TTAGGG)3T] (S24), for which special effort was made to examine the TMPyP4-G4 binding stoichiometry, the binding modes, and the conformational conversion of the G4 structure under different potassium ion (K+) concentration. It is found that, in the presence of 0, 10 mM, and 100 mM K+, TMPyP4 forms a complex with the anti-parallel G4 in a TMPyP4-to-G4 molar ratio of 5, 5 and 3, respectively, and the increase of K+ concentration would reduce the binding affinity of TMPyP4 to G4. For the TMPyP4-G4 complex, the end-stacking mode and groove binding mode were presumed mainly by the results of time-resolved fluorescence spectroscopy in the three cases. Most importantly, it is found that TMPyP4 can directly induce the formation of the anti-parallel G4 structure from the single-strand oligonucleotide S24 in the absence of K+, and that it can preferentially induce the conformational conversion of the G4 structure from the hybrid-type to the anti-parallel one in the presence of K+.  相似文献

A water-soluble polysaccharide DNP-W2 composed of glucose, mannose, and galactose in the molar ratio of 6.1:2.9:2.0 had been isolated from the stems of Dendrobium nobile. Its molecular weight was 1.8 × 104 Da determined by HPGPC. Structural features of DNP-W2 were investigated by a combination of chemical and instrumental analysis, including FTIR, GC, GC-MS, periodate oxidation-Smith degradation, methylation analysis, partial acid hydrolysis, and NMR spectroscopy. The results showed that DNP-W2 is a 2-O-acetylgalactomannoglucan and has a backbone consisting of (1→4)-linked β-d-Glcp, (1→6)-linked β-d-Glcp, and (1→4)-linked β-d-Manp, with branches at O-6 of (1→4)-linked β-d-Glcp and β-d-Manp. The branches are composed of α-d-Galp. The acetyl groups are substituted at O-2 of (1→4)-linked Manp. Preliminary tests in vitro reveals that DNP-W2 can stimulate ConA- and LPS-induced T and B lymphocyte proliferation.  相似文献

This paper investigated the effects of ox-LDL on PCSK9, and the molecular mechanisms of PCSK9 siRNA-inhibited apoptosis induced by ox-LDL in human umbilical vein endothelial cells (HUVECs), to clarify the role of PCSK9 in atherosclerogenesis. HUVECs were incubated with ox-LDL for 24?h. The apoptosis was observed by Hoechst 33258 staining. The expression of PCSK9, LOX-1 mRNAs and proteins was detected by RT-PCR, western blot, respectively. The PCSK9 siRNAs labeled with fluorescence were transfected into HUVECs by Lipofectamine 2000. After transfection for 24?h, cells were treated with ox-LDL for 24?h, HUVECs apoptosis transfected siRNA was detected by Hoechst 33258 staining and flow cytometer. The expression of Bcl-2, Bax, caspase3, 8, 9 was detected by western blot. The activity of caspase3, 9 was detected by kits. Our results showed that apoptosis of HUVECs and the expressions of PCSK9 and LOX-1 were upregulated secondary to induction by ox-LDL in a concentration-dependent manner. However, ox-LDL-induced HUVEC apoptosis and PCSK9 expression, but not LOX-1 expression, were significantly reduced by PCSK9 siRNA. These results demonstrate a linkage between HUVEC apoptosis and PCSK9 expression. Furthermore, we detected the possible pathway involved in apoptotic regulation by PCSK9 siRNA; our results showed that the expression of Bcl-2 decreased, whereas that of Bax increased. In addition, ox-LDL enhanced the activity of caspase9 and then caspase3. Pretreatment of HUVECs with PCSK9 siRNA blocked these effects of ox-LDL. These findings suggest that ox-LDL-induced HUVECs apoptosis could be inhibited by PCSK9 siRNA, in which Bcl/Bax-caspase9-caspase3 pathway maybe was involved through reducing the Bcl-2/Bax ratio and inhibited the activation of both caspase9 and 3.  相似文献

喜马拉雅西部独特、丰富的自然资源对当地居民生计及生态服务等方面起着重要的作用。由于长期以来当地居民与山地生态系统的相互作用,特别是农业生产、畜牧业放牧、薪柴采集以及其他多种多样的资源利用方式,形成了一种特殊的山区文化景观。本文以印度北部的山地小流域PaliGad(共有25个村子)为例,主要研究当地的资源利用状况,利用卫星遥感数据对该地区可利用自然资源进行评估分析,通过从户到户的社会经济调查,对其提供的生态服务功能以及受威胁的程度进行估计,研究分析了村民对资源需求及获取的时空变化情况。结果显示,平均每人每天的薪柴采集量为1.12k,平均每人每天通过修剪枝叶获得饲料采集量为3.69kg,平均每人每天从森林中采集草料的量为3.25kgo对生态系统服务功能进行估测的结果显示,森林可提供更多的临时调节功能,而农业更多的是支撑服务功能,河流,水体给当地人提供了文化服务功能。以山区典型的人-地生态系统为例,这类生态系统中的自然资源破碎化程度很高。研究发现,该区域贫瘠土地上的自然资源需求还在不断增加。因此,从长远来看,人对资源的无止境获取将不利于整个流域的可持续发展。  相似文献
