This article presents an analysis of the microbial community growing on a stainless steel plate. A sample from this plate has been tested for the rpoC gene. Dominant biofilm species (clones) were identified with the help of molecular cloning and nucleotide sequencing of a 16S rRNA gene fragment. The activities of planktonic microorganisms and biofilm microorganisms attached to the plate were compared. Seven of nineteen sequences (37%) were found to belong to the Sphingobacteria group. Analysis of 16S rRNA gene sequences cloned from natural biofilms sampled from Lake Baikal showed a great variety of microorganisms in the biofilm. The following taxonomic groups were revealed among them: sphingobacteria, α-proteobacteria, cyanobacteria, verrucomicrobia, bacilli, γ-proteobacteria, flavobacteria, and β-proteobacteria. This kind of analysis was done in Lake Baikal for the first time.  相似文献

Gene expression, protein synthesis, and activities of alternative oxidase (AOX), uncoupling proteins (UCP), adenine nucleotide translocator (ANT), and non-coupled NAD(P)H dehydrogenases (NDex, NDPex, and NDin) were studied in shoots of etiolated winter wheat (Triticum aestivum L.) seedlings after exposure to hardening low positive (2°C for 7 days) and freezing (?2°C for 2 days) temperatures. The cold hardening efficiently increased frost-resistance of the seedlings and decreased the generation of reactive oxygen species (ROS) during further cold shock. Functioning of mitochondrial energy-dissipating systems can represent a mechanism responsible for the decrease in ROS under these conditions. These systems are different in their response to the action of the hardening low positive and freezing temperatures. The functioning of the first system causes induction of AOX and UCP synthesis associated with an increase in electron transfer via AOX in the mitochondrial respiratory chain and also with an increase in the sensitivity of mitochondrial non-phosphorylating respiration to linoleic and palmitic acids. The increase in electron transfer via AOX upon exposure of seedlings to hardening freezing temperature is associated with retention of a high activity of NDex. It seems that NDex but not the NDPex and NDin can play an important role in maintaining the functional state of mitochondria in heterotrophic tissues of plants under the influence of freezing temperatures. The involvement of the mitochondrial energy-dissipating systems and their possible physiological role in the adaptation of winter crops to cold and frost are discussed.  相似文献

Resistance of Penicillium piceumF-648 to hydrogen peroxide under short-term and prolonged oxidative stress was studied. An increase in the activity of intracellular catalase in fungal cells after short-term exposure to hydrogen peroxide was shown. Activation of fungal cells induced by H2O2 depends on the H2O2 concentration, time of exposure, and growth phase of the fungus. Variants of P. piceum F-648 that produced two forms of extracellular catalase with different catalytic properties were obtained due to prolonged adaptation to H2O2. Catalase with low affinity for substrate was produced predominantly by the parent culture and variant 3; however, a high substrate affinity of catalase was observed in variant 5. Variant 5 of P. piceum F-648 displayed a high catalytic activity and operational stability of catalase in the presence of phosphate ions and a concentration of substrate less than 30 mM at pH more than 7.  相似文献

Feature search for a light bar with one orientation (or color) embedded in an array of bars with a very different orientation (or color) is quick, easy and independent of the number of array elements. In contrast, search for a conjunction target has a linear response time dependence on the number of distractors. Training can improve performance of both these tasks. We report that these properties may not be valid for eccentric stimulus presentation. In general, the two hemifields are not equally suited to search, and training is most effective in the weaker hemifield. In addition, the feature-search independence of set-size may not always be valid for stimulus arrays that are presented peripherally. Subjects were tested on orientation and color feature tasks, and on orientation-color conjunction search with 3 array sizes presented at fixation or eccentrically in the right or left hemifield. During a second testing session, improvement was so much greater for the non-preferred hemifield that sometimes the preference was switched. Surprisingly, preferred hemifield performance actually declined for some subjects. Thus, the hemifield preference effect seems related to competition, and perhaps an automatic attention-directing mechanism. We confirmed the central presentation set-size independence for feature search but found a great difference between large and small arrays when presentation was lateral. There are two sources of this array size effect: 1. Target eccentricity, demonstrated by comparing performance for different target locations with the same array size. 2. Target location uncertainty, seen by comparing performance for different size arrays when the target elements appeared at the same locations. Training also affected the array-size dependence, changing search performance from set-size dependent to independent or vice versa at the point of greatest training effect.  相似文献

We studied the effects of treatments with adrenaline hydrochloride, obsidan (a -adrenoblocker), melipramine (an inhibitor of monoamine uptake by neurons), and reserpine (a sympatholytic drug) on tumor growth (Pliss' lymphosarcoma in rats) and on the antitumor activity of a novel cytostatic drug, chlofiden. We found that adrenaline and reserpine enhanced the antitumor effect of chlofiden. Isolated applications of adrenaline and melipramine exerted slight antitumor effects, while after reserpine treatment there was a trend toward stimulation of tumor growth. Under the conditions of the model used, obsidan demonstrated no noticeable antitumor activity and did not modify the antitumor effect of chlofiden. Possible mechanisms of the observed effects are discussed.  相似文献

Data on the feeding of larvae of mass Black Sea fish species of the families Blenniidae and Gobiidae with a length of 2.2–7.0 mm from coastal waters of the Crimea (Sevastopol) are presented. Despite the tendency for an improvement in the feeding of larval fish, Mnemiopsis leidyi, in the case of a prolonged staying in the plankton in mass amounts in the absence of Beroe ovata, exerts a negative effect on the food resources and the survival of larvae. This is evidenced by the indices of food consumption and the proportion of larvae with empty intestines.  相似文献

We studied the effect of a 14-day course of interval hypoxic hypoxia (IHH) on the concentration in the blood of healthy subjects of the vascular endothelial growth factor (vEGF) and the basic fibroblast growth factor (bFGF), which regulate endothelial cell proliferation and angiogenesis. The IHH course was carried out using a BIO-NOVA 204 plant for modeling hypoxic conditions. IHH resulted in a twofold increase in vEGF 24 h after its application; this was followed by a decrease in vEGF to the initial level 48 h after IHH, a subsequent increase by 40% 72 h after IHH, and normalization to the initial level by day 14. The concentration of bFGF increased by 21% 24 h after IHH and gradually decreased to the initial level by day 14. During the initial 24–72 h, changes in the concentration of both factors positively correlated with the dynamics of oxidative stress markers, such as total prooxidative activity and plasma concentration of thiobarbituric acid–reactive species. It is assumed that IHH increases the vEGF content owing to induction of its biosynthesis by reactive oxygen species. Therefore, it is promising to examine the effect of IHH on the morphofunctional parameters of endothelium.  相似文献

A comparative kinetic study of extracellular catalases produced by Penicillium piceum F-648 and their variants adapted to H2O2 was performed in culture liquid filtrates. The specific activity of catalase, the maximum rate of catalase-induced H2O2 degradation (V max), V max/K M ratio, and the catalase inactivation rate constant in the enzymatic reaction (k in, s–1) were estimated in phosphate buffer (pH 7.4) at 30°C. The effective constant representing the rate of catalase thermal inactivation (k in *, s–1) was determined at 45°C. In all samples, the specific activity and K M for catalase were maximum at a protein concentration in culture liquid filtrates of (2.5–3.5) × 10–4 mg/ml. The effective constants describing the rate of H2O2 degradation (k, s–1) were similar to that observed in the initial culture. These values reflected a twofold decrease in catalase activity in culture liquid filtrates. We hypothesized that culture liquid filtrates contain two isoforms of extracellular catalase characterized by different activities and affinities for H2O2. Catalases from variants 5 and 3 with high and low affinities for H2O2, respectively, had a greater operational stability than the enzyme from the initial culture. The method of adaptive selection for H2O2 can be used to obtain fungal variants producing extracellular catalases with improved properties.  相似文献

Growth-stimulating effects of water-soluble nanoparticles of N-substituted monoamino-acid derivatives of fullerene C60 (L- and D-alanine, L- and D-valine, L- and D-aspartic acid, β-alanine, and γ-aminobutyric and ε-aminocaproic acids in potassium salt form) were investigated. It was found that the nanoparticle size and relative antiradical activity of fullerene derivatives were factors that influence such physiological parameters of field peas as seed germination rate, germination energy, and root growth capacity. It was shown that the relative antiradical activity of nanoparticles in the selected group of compounds was determined by the total surface area of the nanoparticles regardless of the structure of the amino-acid substituent. The possibility of using amino-acid derivatives of fullerene as effective growth stimulating substances has been demonstrated. A dose-dependent effect of N-(monohydrofullerenyl)-D-alanine potassium salt in a concentration range of 10–9–10–11 M on the seed-germination rate and germination energy of field peas has been demonstrated.
