In this paper, a most sensitive electrochemical biosensor for detection of prostate‐specific antigen (PSA) was designed. To reach the goal, a sandwich type electrode composed of reduced graphene oxide/ gold nanoparticles (GO/AuNPs), Anti‐Total PSA monoclonal antibody, and anti‐Free PSA antibody was assembled. The functionalized materials were thoroughly characterized by atomic force microscope spectroscopy, transmission electron microscopy, and X‐ray diffraction techniques. The electrochemical properties of each of the modification step were evaluated by cyclic voltammetry and electrochemical impedance spectroscopy. The results presented that the proposed biosensor possesses high sensitivity toward total and free PSA. Furthermore, the fabricated biosensor revealed an excellent selectivity for PSA in comparison to the other tumor markers such as BHCG, Alb, CEA, CA125, and CA19‐9. The limit of detection for the proposed electrochemical biosensor was estimated to be around 0.2 and 0.07 ng/mL for total and free PSA antigen, respectively.  相似文献

Voice impairments, attention to increased unhealthy social behavior and voice abuse, have been increasing dramatically. Therefore, diagnosis of voice diseases has an important role in the opportune treatment of pathologic voices. This paper presents an extensive study in identification of different voice disorders which their origin is in the vocal folds. Firstly, a qualitative study is applied based on short-time Fourier transform (STFT) and continuous wavelet transform (CWT) in order to investigate their aptitude in the presentation of discriminative features to identify disordered voices from normal ones. Therefore, wavelet packet transform (WPT) for their ability to analyze scrutinizingly a signal at several levels of resolution is chosen as strong speech signal parameterization method. The ability of energy and entropy features, obtained from the coefficients in the output nodes of the optimum wavelet packet tree, is investigated. Linear discriminant analysis (LDA) and principal component analysis (PCA) are evaluated as feature dimension reduction methods in order to optimize recognition algorithm. The performance of each structure is evaluated in terms of the accuracy, sensitivity, specificity, and area under the receiver operating curve (AUC). Eventually, entropy features in the sixth level of WPT decomposition along with feature dimension reduction by LDA and a support vector machine-based classification method is the most optimum algorithm that leads to the recognition rate of 100% and AUC of 100%. Proposed system clearly outperforms previous works in both respect of accuracy and reduction of residues; which may lead in full accuracy and high speed diagnosis procedure.  相似文献

Mahshahr has a strategic position and is considered as industrial hub of Iran. Selected heavy metals and polycyclic aromatic hydrocarbons (PAHs) contamination and health risk, and the mineralogical composition of street dust from Mahshahr were investigated. Results indicated that geology is the main determinant of the dust mineralogical composition. Calculated enrichment factor (EF) and principal component analysis (PCA) showed that Pb, Hg, Zn, and Cu accumulations are greatly influenced by anthropogenic sources including traffic and industry. High heavy metals content poses great ecological risk in the study area and exposure doses revealed that ingestion is the main exposure route to street dust, especially for children in residential/commercial areas. It was found that the total amount of PAHs (∑PAHs) varies from 161 to 1996 µg/kg, dominated by four-ring PAHs. Diagnostic ratios and PCA showed that both petrogenic and pyrogenic sources of PAHs in Mahshahr street dust and traffic play important roles in this respect. Furthermore, toxic equivalents and incremental lifetime cancer risk of PAHs in street dust indicated a high potential carcinogenic risk for inhabitants mainly via dermal contact and ingestion pathways particularly for outdoor workers in industrial use scenario. Finally, distribution maps of total hazard index of heavy metals and cancer risk of PAHs indicated the most impacted zones for different groups and use scenarios.  相似文献

Several recent studies have reported a decline in virus retention during virus challenge filtration experiments, although the mechanism(s) governing this phenomenon for different filters remains uncertain. Experiments were performed to evaluate the retention of PP7 and PR772 bacteriophage through Ultipor VF Grade DV20 virus filters during constant pressure filtration. While the larger PR772 phage was fully retained under all conditions, a 2‐log decline in retention of the small PP7 phage was observed at high throughputs, even under conditions where there was no decline in filtrate flux. In addition, prefouling the membrane with an immunoglobulin G solution had no effect on phage retention. An internal polarization model was developed to describe the decline in phage retention arising from the accumulation of phage in the upper (reservoir) layer within the filter which increases the challenge to the lower (rejection) layer. Independent support for this internal polarization phenomenon was provided by confocal microscopy of fluorescently labeled phage within the membrane. The model was in good agreement with phage retention data over a wide range of phage titers, confirming that virus retention is throughput dependent and supporting current recommendations for virus retention validation studies. These results provide important insights into the factors governing virus retention by membrane filters and their dependence on the underlying structure of the virus filter membrane. © 2014 American Institute of Chemical Engineers Biotechnol. Recently, bacterial cellulose (BC) based wound dressing have raised significant interests in medical fields. However, to our best knowledge, it is apparent that the BC itself has no antibacterial activity. In this study, we optimized graphene oxide‐silver (GO‐Ag) nanohybrid synthesis using Response Surface Methodology and impregnate it to BC and carefully investigate their antibacterial activities against both the Gram‐negative bacteria Escherichia coli and the Gram‐positive bacteria Staphylococcus aureus. We discover that, compared to silver nanoparticles, GO‐Ag nanohybrid with an optimal GO suspension's pH and ratio is much more effective and shows synergistically enhanced, strong antibacterial activities at rather low dose. The GO‐Ag nanohybrid is more toxic to E. coli than that to S. aureus. The antibacterial and mechanical properties of BC/GO‐Ag composite are further investigated.  相似文献

Downregulation of microRNA-590-3p (miR-590-3p) is a frequently occurring, nonphysiological event which is observed in several human cancers, especially breast cancer. However, the significance of miR-590-3p still remain unclear in the progression of this disease. This study explored the role of miR-590-3p in apoptosis of breast cancer cells. Gene expression of miR-590-3p, Sirtuin-1 (SIRT1), Bcl-2 associated X protein (BAX), and p21 was evaluated with real-time polymerase chain reaction (PCR) and SIRT1 protein expression was assessed by Western blot analysis in breast cancer cell lines. Bioinformatics analysis and luciferase reporter assay were used to evaluate targeting of SIRT1 messenger RNA (mRNA) by miR-590-3p. Cells were transfected with miR-590-3p mimic and inhibitor and their effects on the expression and activity of SIRT1 were evaluated. The effects of miR-590-3p upregulation on the acetylation of p53 as well as cell viability and apoptosis were assessed by Western blot analysis, WST-1 assay, and flow cytometry, respectively. miR-590-3p expression was considerably downregulated in breast cancer cells which was accompanied by upregulation of SIRT1 expression. SIRT1 was recognized as a direct target for miR-590-3p in breast cancer cells and its protein expression and activity was dramatically inhibited by the miR-590-3p. In addition, there was an increase in p53 and its acetylated form that ultimately led to upregulation of BAX and p21 expression, suppression of cell survival, and considerable induction of apoptosis in breast cancer cells. These findings suggest that miR-590-3p exerts tumor-suppressing effects through targeting SIRT1 in breast cancer cells, which makes it a potential therapeutic target for developing more efficient treatments for breast cancer.  相似文献

Product quality analyses are critical for developing cell line and bioprocess producing therapeutic proteins with desired critical product quality attributes. To facilitate these analyses, a high‐throughput small‐scale protein purification (SSP) is required to quickly purify many samples in parallel. Here we develop an SSP using ion exchange resins to purify a positively charged recombinant growth factor P1 in the presence of negatively charged dextran sulfate supplemented to improve the cell culture performance. The major challenge in this work is that the strong ionic interaction between P1 and dextran sulfate disrupts interaction between P1 and chromatography resins. To solve this problem, we develop a two‐step SSP using Q Sepharose Fast Flow (QFF) and SP Sepharose XL (SPXL) resins to purify P1. The overall yield of this two‐step SSP is 78%. Moreover, the SSP does not affect the critical product quality attributes.
