Little is known about deep soil heterogeneity, or its relationship with fine root distribution. Beneath a mature, closed-canopy forest of eastern Amazonia, and the pastures and secondary forests that are derived from this forest, soil soft spots and hollow chambers occur to at least 9 meters depth. We measured the vertical distribution of these soil patches, and compared chemical characteristics, mycorrhizal infection, and root density of soil soft spots with the surrounding matrix of more homogeneous soil. Soil soft spots and chambers varied little with depth, but occupied the greatest soil volume (0.8 to 1.2%) from 4 to 6 m depth in the mature forest. Soft spots had lower pH, P availability and arbuscular mycorrhizal infection, and higher K availability than surrounding soil. Root length density was 2 to 15 times higher in soft spots than in surrounding soil. In the pastures, roots were found only in soil soft spots at depths of >3 m. Pastures and secondary forest had more soil chambers in the upper meter of soil than mature forest, but were otherwise indistinguishable in their patterns of deep soil heterogeneity. Soil soft spots may be vestiges of cutter ant nest chambers, while hollow chambers are cutter ant chambers and root channels. This article describes the phytochemical study of Cannabis sativa roots from northeastern Brazil. The dried plant material was pulverized and subjected to exhaustive maceration with ethanol at room temperature, obtaining the crude ethanolic extract (Cs-EEBR). The volatile compounds were analyzed by gas chromatography coupled with mass spectrometry (GC/MS), which allowed to identify 22 compounds by comparing the linear retention index (LRI), the similarity index (SI) and the fragmentation pattern of the constituents with the literature. By this technique the major compounds identified were: friedelan-3-one and β-sitosterol. In addition, two fractions were obtained from Cs-EEBR by classical column chromatography and preparative thin layer chromatography. These fractions were analyzed by NMR and IR and together with the mass spectrometry data allowed to identify the compounds: epifriedelanol, friedelan-3-one, β-sitosterol and stigmasterol. The study contributed to the phytochemical knowledge of Cannabis sativa, specifically the roots, as there are few reports on the chemical constituents of this part of the plant.  相似文献

Summary— Multidrug-resistant (MDR) variants of a human osteosarcoma cell line (U-2 OS) have been recently obtained by continuous exposure to doxorubicin (DX). The growth and phenotypic characteristics of these cell lines have been demonstrated to be related to the level of expression of P-glycoprotein. In this work, the morphological changes associated with MDR have been evaluated by quantitative image analysis and transmission electron microscopy. Resistant cells present morphological changes with respect to sensitive cells at both cytoplasmic and nuclear level. Some of these changes appear to be related to the degree of resistance but not to the direct presence of DX, since deprived cells maintain some modified characters, while others are partly lost. These findings suggest that DX exposure affects cell metabolism causing progressive changes of the cell morphotype.  相似文献

A mosaic of intact native and human-modified vegetation use can provide important habitat for top predators such as the puma (Puma concolor), avoiding negative effects on other species and ecological processes due to cascade trophic interactions. This study investigates the effects of restoration scenarios on the puma’s habitat suitability in the most developed Brazilian region (São Paulo State). Species Distribution Models incorporating restoration scenarios were developed using the species’ occurrence information to (1) map habitat suitability of pumas in São Paulo State, Southeast, Brazil; (2) test the relative contribution of environmental variables ecologically relevant to the species habitat suitability and (3) project the predicted habitat suitability to future native vegetation restoration scenarios. The Maximum Entropy algorithm was used (Test AUC of 0.84 ± 0.0228) based on seven environmental non-correlated variables and non-autocorrelated presence-only records (n = 342). The percentage of native vegetation (positive influence), elevation (positive influence) and density of roads (negative influence) were considered the most important environmental variables to the model. Model projections to restoration scenarios reflected the high positive relationship between pumas and native vegetation. These projections identified new high suitability areas for pumas (probability of presence >0.5) in highly deforested regions. High suitability areas were increased from 5.3% to 8.5% of the total State extension when the landscapes were restored for ≥ the minimum native vegetation cover rule (20%) established by the Brazilian Forest Code in private lands. This study highlights the importance of a landscape planning approach to improve the conservation outlook for pumas and other species, including not only the establishment and management of protected areas, but also the habitat restoration on private lands. Importantly, the results may inform environmental policies and land use planning in São Paulo State, Brazil.  相似文献

Zusammenfassung Der Charakter der Polyphenoloxydase-Aktivität als Erbmerkmal wurde aus folgenden Tatsachen geschlossen: Der Unterschied in der Aktivität dieses Fermentes zwischen 2 Sorten blieb über die ganze Vegetationszeit erhalten und wiederholte sich in ihren Nachkommen. In der F1 zeigte sich deutliche Dominanz der geringeren Aktivität.Es besteht kein Zusammenhang zwischen Polyphenoloxydase-Aktivität und Anfälligkeit fürPeronospora tabacina, denn beim Vergleich je einerPeronospora-resistenten und einer anfälligen Sorte war einmal die anfällige und einmal die resistente die fermentaktivere. Die Unterschiede in der Aktivität der Polyphenoloxydase ließen also kein Prinzip erkennen.Zwischen der Polyphenoloxydase-Aktivität und der Y-Virus-Anfälligkeit dagegen konnte ein deutlicher Zusammenhang festgestellt werden. Die Unterschiede in der Aktivität der Polyphenoloxydase je einer Y-Virus-resistenten und einer anfälligen Sorte verhielten sich gleichsinnig: Die resistenten hatten eine höhere Polyphenoloxydase-Aktivität als die anfälligen Sorten.Die Frage nach der Art des Zusammenhangs zwischen Y-Virus-Disposition und Polyphenoloxydase-Aktivität wurde diskutiert: Es kann kein ursächlicher sein in der Weise, daß Y-Virus-Anfälligkeit von der Polyphenoloxydase-Aktivität abhänge, sondern er muß nach den vorliegenden Versuchsergebnissen als indirekter gedeutet werden derart, daß beide Erbmerkmale, Y-Virus-Anfälligkeit und Polyphenoloxydase-Aktivität von einer gemeinsamen, gengesteuerten dritten Größe abhängen. Die Polyphenoloxydase-Aktivität wird damit als bloßes Anzeichen der physiologischen Aktivität der betreffenden Sorte betrachtet, ohne selbst einen direkten Einfluß auf die Krankheitsdisposition zu haben.
Summary consequent on the following facts polyphenoloxidase activity proved to be a hereditary quality: The difference of activity between two varieties endured during the whole vegetation time and replicated in their descendants. In F1 generation dominance of lower activity appeared.There is no relation between polyphenoloxidase activity and susceptibility toPeronospora tabacina, for comparing everyPeronospora susceptible variety with a resistent one, no regularity could be observed: Once the susceptible variety and another time the resistant possessed the higher enzyme activity.Between polyphenoloxidase activity and Y Virus disposition a clear relation became evident: The resistant varieties possessed a higher activity of this enzyme than the susceptible ones.The question is discussed how to explain this relation between polyphenoloxidase activity and Y Virus disposition. It can not be a causal one in this way that Y Virus susceptibility should depend on polyphenoloxidase activity. Relation must be an indirect one in this way that both hereditary qualities: Y Virus and oxidase activity depend on the same third gene-dependant but unknown quantity. Thus polyphenoloxidase activity is considered to act only as an indicator of physiological processes in the plant without itself having a direct influence on the disposition to Y Virus disease.

Bacteria in nature often encounter non-antibiotic antibacterials (NAAs), such as disinfectants and heavy metals, and they can evolve resistance via mechanisms that are also involved in antibiotic resistance. Understanding whether susceptibility to different types of antibacterials is non-randomly associated across natural and clinical bacteria is therefore important for predicting the spread of resistance, yet there is no consensus about the extent of such associations or underlying mechanisms. We tested for associations between susceptibility phenotypes of 93 natural and clinical Escherichia coli isolates to various NAAs and antibiotics. Across all compound combinations, we detected a small number of non-random associations, including a trio of positive associations among chloramphenicol, triclosan and benzalkonium chloride. We investigated genetic mechanisms that can explain such associations using genomic information, genetic knockouts and experimental evolution. This revealed some mutations that are selected for by experimental exposure to one compound and confer cross-resistance to other compounds. Surprisingly, these interactions were asymmetric: selection for chloramphenicol resistance conferred cross-resistance to triclosan and benzalkonium chloride, but selection for triclosan resistance did not confer cross-resistance to other compounds. These results identify genetic changes involved in variable cross-resistance across antibiotics and NAAs, potentially contributing to associations in natural and clinical bacteria.  相似文献

Background aimsA medium supplemented with fetal bovine serum (FBS) is of common use for the expansion of human mesenchymal stromal cells (MSCs). However, its use is discouraged by regulatory authorities because of the risk of zoonoses and immune reactions. Human platelet lysate (PL) obtained by freezing/thawing disruption of platelets has been proposed as a possible substitute of FBS. The process is time-consuming and not well standardized. A new method for obtaining PL that is based on the use of ultrasound is proposed.MethodsPlatelet sonication was performed by submerging platelet-containing plastic bags in an ultrasonic bath. To evaluate platelet lysis we measured platelet-derived growth factor-AB release. PL efficiency was tested by expanding bone marrow (BM)-MSCs, measuring population doubling time, differentiation capacity and immunogenic properties. Safety was evaluated by karyotyping expanded cells.ResultsAfter 30 minutes of sonication, 74% of platelet derived growth factor-AB was released. PL enhanced BM-MSC proliferation rate compared with FBS. The mean cumulative population doubling (cPD) of cells growth in PL at 10%, 7.5% and 5% was better compared with cPD obtained with 10% FBS. PD time (hours) of MSCs with PL obtained by sonication was shorter than for cPD with PL obtained by freezing/thawing (18.9 versus 17.4, P < 0.01). BM mononucleated cells expressed MSC markers and were able to differentiate into adipogenic, osteogenic and chondrogenic lineages. When BM-MSCs and T cells were co-cultured in close contact, immunosuppressive activity of BM-MSCs was maintained. Cell karyotype showed no genetic alterations.ConclusionsThe proposed method for the production of PL by sonication could be a safe, efficient and fast substitute of FBS, without the potential risks of FBS.  相似文献
