Using Greene's melanoma transplanted into Syrian (golden) hamsters, we determined the relative biological effectiveness (RBE) of thermal neutron capture therapy (TNCT) using 10B-paraboronophenylalanine (10B-BPA) in comparison with a 9-MeV electron beam. We also obtained the RBE of the 10B(n,α)7Li reaction by calculation based on summed dose data from TNCT. Throughout this study, the Kyoto University Research Reactor was used as the source for thermal neutrons; the reactor was specially altered to attain a low contamination level both for gamma-rays and fast neutrons. 10B-BPA was administered 8 hours before thermal neutron irradiation to the hamsters with melanoma. The tumor was then irradiated at 5 MW for 90 minutes. The absorbed dose from this TNCT was calculated by the method of Fairchild and Goodman (Phys. Med. Biol. 1966; 2:15–30). The RBEs of the TNCT and the 10B(n,α)7Li reaction obtained by the tumor growth delay time (TGDT) method were 2.22 and 2.51, respectively, at 10.5 days of TGDT. These RBE values varied with TGDT and the absorbed dose. The RBE value of TNCT had a peak at 7.0 days of TGDT; that of the 10B(n,α)7Li reaction was higher at a low absorbed dose level and lower at a high absorbed dose level.  相似文献

Specific and powerful cancer killing effect for melanoma by boron neutron capture therapy (BNCT) using DOPA analogue, 10B-p-boronophenylalanine (10B-BPA), has been established, but amelanotic melanoma is insufficiently responsive to 10B-BPA BNCT in comparison with actively melanin-producing melanoma. Although the accumulation mechanism of 10B-BPA within melanoma was not established, we have recently obtained findings suggesting that melanin monomers, key intermediates for melanin polymer formation, play a critical role in 10B-BPA accumulation. In addition, there are some kinds of human amelanotic melanomas, such as MEL2A, in which expression of tyrosinase is repressed or lacking though tyrosinase-related protein (TRP)-l and TRP-2 are well expressed. Thus, by using a similarly tyrosinase-lacking mouse amelanotic melanoma cell line, A1059, we constructed TA1059 cells by transfecting human tyrosinase-cDNA into these cells. TA1059 cells acquired higher DOPA-oxidase and DOPAchrome tautomerase activity as well as eumelanin content at even higher levels than those of B16F10 cells. TA1059 cells showed about 2.5 times higher p-boronophenylalanine (BPA) uptake than A1059 cells in culture. In animal experiments, by using these cell lines, tumor growth of TA1059 was significantly suppressed by 10B-BPA BNCT as compared with A1059. These findings indicate that the induction of active melanin biosynthesis by melanogenic gene-transfer effectively improves the treatment of amelanotic melanoma by BNCT.  相似文献

提取梨 (PyrusserotinaRehd .)自交不亲和品种“二十世纪”(基因型为S2 S4 )、自交亲和的突变品种“奥嗄二十世纪”(S2 SSM4 ,SM =Stylar_partmutant;花柱部分突变 )及其亲和后代花柱的可溶性蛋白。经等电聚焦电泳 (IEF_PAGE)分析表明 ,“奥嗄二十世纪”及其后代花柱仍存在SSM4 蛋白 ,但其含量逐代减少 ,同时发现“奥嗄二十世纪”的SSM4 基因仅在柱头表达 ,而“二十世纪”的S4 基因表达的部位除了柱头外 ,还包括花柱上部及花柱下部 ,且表达量呈现从柱头到花柱下部下降的趋势。S蛋白经等电聚焦电泳的凝胶板进行RNase活性染色处理 ,也得到相同的结果。从花柱 (包括柱头 )中纯化出的S蛋白经SDS_PAGE电泳后进行RNase活性染色的结果表明 ,S4 与SSM4 蛋白的分子量相近 (约 30kD) ,并且均具有RNase活性。进一步以酵母RNA为基质测定的比活性也基本相等 ,约为 2 75U·min-1·mg-1蛋白。在离体条件下 ,上述两种S蛋白 (S_RNase)也以相同的程度抑制S4 或SSM4 花粉发芽及花粉管伸长。研究证明 ,自交亲和突变品种“奥嗄二十世纪”的SSM4 基因也具有原始自交不亲和品种“二十世纪”S4 基因的功能。因此 ,其自交亲和的原因可归结为SSM4 基因的表达量较少及SSM4 基因仅在柱头中表达的缘故。  相似文献

Stylar soluble proteins in self-incompatible “Nijisseiki” (S2S4), self-compatible “Osa-Nijisseiki” ( S2SSM4, SM means stylar-part mutant) and its progeny were analyzed by isoelectric focusing polyacrylamide gel electrophoresis (IEF-PAGE).SSM4-allele associated protein, SSM4-protein, existed in the style of “Osa-Nijisseiki” and its progeny. The SSM4-protein expressed only in the stigma of “Osa-Nijisseiki”, whereas in its original variety “Nijisseiki”, S4-protein expressed in the upper and lower parts of the style as well as in the stigma, and its expression amount decreased from the upper part to the lower part. The protein bands analyzed by IEF-PAGE were subjected to RNase activity staining. The results showed that the S4- and the SSM4-proteins have the similar molecular weights (approximately 30 kD) and RNase activity. The specific-activities measured with yeast RNA were similar, equivalent to approximately 275 U·min-1·mg-1 protein. The S SM4-protein showed almost the same inhibitory effects as the S4-protein on the pollen germination and the pollen tube growth with S4- and SSM4-alleles in vitro . From the above results, the reasons of the self-compatibility of “Osa-Nijisseiki” are considered as (1) low expression of the SSM4-gene and (2) the SSM4-gene expression only in stigma.  相似文献

Azolla, a small water fern, abscises its roots and branches within 30 min upon treatment with various stresses. This study was conducted to test whether, in the rapid abscission that occurs in Azolla, breakdown of wall components of abscission zone cells by OH is involved. Experimentally generated OH caused the rapid separation of abscission zone cells from detached roots and the rapid shedding of roots from whole plants. Electron microscopic observations revealed that OH rapidly and selectively dissolved a well‐developed middle lamella between abscission zone cells and resultantly caused rapid cell separation and shedding. Treatment of abscission zones of Impatiens leaf petiole with OH also accelerated the separation of abscission zone cells. However, compared with that of Azolla roots, accelerative effects in Impatiens were weak. A large amount of OH was cytochemically detected in abscission zone cells both of Azolla roots and of Impatiens leaf petioles. These results suggest that OH is involved in the cell separation process not only in the rapid abscission in Azolla but also in the abscission of Impatiens. However, for rapid abscission to occur, a well‐developed middle lamella, a unique structure, which is sensitive to the attack of OH, might be needed.  相似文献

Left-handed Helical Polynucleotides with D-Sugar Phosphodiester Backbones   总被引:3,自引:0,他引:3  
Naturally occurring polynucleotides have right-handed helical confrontations in the solid state1 and in solution2. Poly(dI-dC)poly(dI-dC) was found to form a left-handed helix in spite of the D-sugar backbone. Also, L-adenylyl-(3′–5′)-L-adenosine synthesized by Tazawa et al4. takes up the left-handed stacked conformation. We had synthesized a dinucleoside monophosphate, 8,2′-anhydro-8-mercapto-9-β-D-arabinofuranosyladenine phosphoryl-(3′–5′)-8,2′-anhydro-8-mercapto-9-β-D-arabinofuranosyladenine (AspAs) (molecular structure Ia; see also ref. 5) and this compound has a left-handed stacked conformation. The two bases in Ia, having the D-sugar backbone, stacked along the left-handed helical axis; these bases are fixed at ?CN = ?108° (syn-anti region) by the anhydro linkages.  相似文献

The plants of Kadsura longipedunculata (Schisandraceae) are monoecious and possess either red or yellow male flowers (the androecium), with yellow tepals, and yellow female flowers. All flower types simultaneously produce heat and floral odours (dominated by methyl butyrate) throughout a 4–5-h nocturnal period. The flowers are pollinated only by female, pollen-eating Megommata sp. (Cecidomyiidae). Pollen is the only reward, and female flowers use the same attractants as male flowers but offer no food (pollination by deceit). Open pollinated flowers in nature varied in fruit set from 8 to 92%. Megommata (subfamily Cecidomyiinae, supertribe Cecidomyiidi), consists of six described species, which feed on Coccoidea (scale insects) and are distributed worldwide.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society , 2008, 93 , 523–536.  相似文献

In theory, one factor determining the rate and nature of the assembly of island biotas is the presence or absence of stepping stone islands, yet no field studies have demonstrated stepping stone function in practice. Krakatau, in Sunda Strait, is about equidistant from Java and Sumatra. Sebesi lies about half way between Krakatau and Sumatra, but no island intervenes between Krakatau and the nearest coast of Java. We assess the evidence that Sebesi has acted as an important stepping stone for Krakatau's recolonization since the devastating 1883 volcanic eruption. About a quarter of Krakatau's resident land birds, two-fifths of its reptiles, bats and land molluscs, and about two-thirds of its termites, pteridophytes, butterflies and spermatophytes are unknown on Sebesi, evidently having colonized without stepping stone involvement. Identifiable Sumatran taxa do not outnumber identifiable Javan ones on Krakatau, nor do historical distribution records indicate movement from Sebesi to Krakatau in animal groups. Krakatau's biota is not a subset of Sebesi's in predominantly anemochorous or thallassochorous plant groups, butterflies, reptiles or bats, and is only marginally so in termites. It is a subset in predominantly zoochorous spermatophyte groups, except Ficus species, and in birds and land molluscs. Comparison with a weaker stepping stone candidate, Panaitan, provides no evidence for a stepping stone role for Sebesi in butterflies or termites. We discuss the dispersal and establishment constraints on colonization by the groups involved, and conclude that, overall, Sebesi had little impact as a stepping stone. 1. Population dynamics of Asphondylia sphaera Monzen (Diptera: Cecidomyiidae), a species that induces fruit galls on Ligustrum japonicum (Oleaceae), was studied from 1970 to 1996 in broad‐leaved evergreen forests in Kagoshima, southern Japan. The numbers of fruit galls and emerging adults fluctuated greatly from year to year along with alternate year flowering of the host plant. 2. To detect density‐dependent and independent forces operating on the A. sphaera population and to assess the relative strength of top‐down and bottom‐up effects, we used the method of Key‐factor/key‐stage analysis, which allowed us to avoid various problems of the conventional key‐factor analysis. 3. Five factors and seven stages were distinguished in the life tables. Key‐factor/key‐stage analysis indicated that the number of flower buds made the largest contribution to the annual changes in total survival rate by operating through the proportion of eggs and first instars that survived abortion of host flower buds and flowers, which contributed most to density‐dependency. 4. The population dynamics of A. sphaera has been strongly influenced by the bottom‐up effects of both the abundance of flower buds and the abortion of flowers of L. japonicum. 5. A top‐down effect of Bracon asphondyliae Maeto (Hymenoptera: Braconidae) was weakly density‐dependent and contributed less to the total survival rate. Abiotic factors, such as summer and winter temperatures and the effect of typhoons were negligible.  相似文献
