Continuous measurements of cytoplasmic pH (pHc) in Sinapis roothairs have been carried out with double-barrelled pH-micro-electrodesin order to gain information on translocation of protons acrossthe plasmalemma and cytoplasmic pH control. (i) The cytoplasmicpH of Sinapis (7–33 ? 0–12, standard conditions)changes no more than 0.1 pHc, per pHo-unit, regardless of whethercyanide is present or not. (ii) Weak acids rapidly acidify pHcand hyperpolarize, while weak bases alkalize pHc and depolarizethe cells, (iii) 1.0 mol M,3 NaCN acidifies the cytoplasm by0.4 to 0.7 pH-units, but alkalizes the vacuole. (iv) 20 mmolm–3 CCCP has no significant effect on pHc, if added atpH 9.6 or 7.2, but acidifies pHc by 1.3 units at pH 4.3. Inthe presence of CCCP, cyanide acidifies the cytoplasm, (v) Chloridetransiently acidifies pHc, while K+, Na+, and have no significant effects, (vi) Cytoplasmic buffer capacityforms a bell-shaped curve versus pHc with an optimum of about50 mol m–3 H+pHc-unit. The modes of proton re-entry and the effects of active and passiveproton transport on cellular pH control are critically discussed.It is suggested that the proton leak, consisting of H+-cotransport(e.g. H+/Cl) rather than H+-uniport, is no threat topHc. The proton export pump, although itself reacting to changesin pHc, influences pHc only to a minor extent. It is concludedthat buffer capacity and membrane transport play moderate rolesin pHc control in Sinapis, while the interlocked H+-producingand -consuming reactions of cellular metabolism are the mainregulating factors. Bodily preserved, secondarily phosphatized arthropods discovered in drill cores on He***l Peninsula, northern Poland, and in its vicinity date from the Upper Cambrian. Comparisons between a group of arthropods of the Upper Cambrian of Sweden recognized as stem-lineage crustaceans indicate that one of these new forms, Cambrocaris baltica n. gen. n. sp., also represents a derivative of the early phase of crustacean evolution prior to the crown-group level. The material also yielded a specimen identified as Skara minuta Müller & Walossek, 1985, hitherto known only from Västergotland, Sweden, and two limb fragments which cannot be assigned to species. □ Crustacea, stem-lineage derivatives. The hydrogen ion-sensitive liquid-membrane micro-electrode,as described by Amman, Lanter, Steiner, Schulthess, Shijo, andSimon (1981) has been developed further and made applicablefor turgescent plant cells even with tough cell walls, by treatmentwith polyvinylchloride (PVC). Such an electrode is slower (t=5–10s) than the untreated electrode (t=2–6 s), but displays55–59 mV/pH-unit between pH 4·3 and 9·0,and is almost insensitive towards different buffers and K+.The electrodes are usable on more than one cell and have stillgood recalibration properties. Testing the electrodes on 11different cell types of Riccia fluitans, Sinapis alba, Zea mays,Avena sativa, Kalanchöe daigremontiana, Lemna gibba andChara corallina, we find the internal pH slightly alkaline (7·1–7·6)in ten cases (exception: old rhizoids of Riccia, pH1=4·8).From that we conclude that the pH-electrode measures in thecytosol. According to the different plant material, severalprocedures for internal pH-measurements are presented and supportedby data: Application of cyanide and acetic acid causes internalacidification. Light-off transiently alkalinazes, light-on transientlyacidifies the internal pH. The advantages and limitations ofthe method are critically discussed, and it is concluded thatthis electrode is a powerful tool in plant physiology. The influence of ferricyanide on transmembrane electron transfer,proton secretion, membrane potential, and cytoplasmic pH ofLimnobium stoloniferum (G.F. Mey) Griseb. root cells was investigatedat different external pH HCF III reduction by the roots was accompanied by membrane depolarization,an increase in proton secretion and by alkalinization of thecytoplasm. In the next century, the boreal ecosystems are projected to experience greater rates of warming than most other regions of the world. As the boreal forest constitutes a reservoir of trees of huge ecological importance and only partially known economic potential, any possible climate‐related change in plant growth and dynamics has to be promptly predicted and evaluated. A model for assessing xylem phenology in black spruce [Picea mariana (Mill.) B.S.P.] using daily temperatures and thermal thresholds was defined and applied to predict changes in onset, ending and duration of xylem growth under different warming scenarios with temperatures rising by up to 3 °C. This was achieved by collecting and analyzing a dataset obtained from a 7‐year monitoring of cambium phenology and wood formation on a weekly time‐scale in trees growing in four sites at different latitudes and altitudes in the Saguenay‐Lac‐Saint‐Jean region (Quebec, Canada). The onset of xylem growth occurred between mid‐May and early June while the end ranged between mid‐September and early October, resulting in a growing season of 101–141 days. The model predicted longer duration of xylem growth at higher temperatures, with an increase of 8–11 days/ °C, because of an earlier onset and later ending of growth. With an increase of 3 °C in the mean temperature during the year, the duration of xylem growth changed on average from 125 to 160 days. The predicted changes in cambial phenology could significantly affect future wood production of the boreal ecosystems.  相似文献

ABSTRACT State wildlife agencies commonly offer private landowners cost-share and technical assistance to improve habitat, but the cost-effectiveness and long-term outcomes of these efforts are rarely evaluated. In 1998, we began a 3-part, statewide evaluation of the Illinois Department of Natural Resources’ (IDNR; USA) Private Land Wildlife Habitat Management Program (hereafter the program) as it functioned from 1986 to 1996. We sent a mail-back questionnaire to IDNR biologists to collect information on their perceptions of the program. We sent a separate mail-back questionnaire to private landholders who participated in the program to ascertain their demographic profile, motivations for participation, and attitudes regarding the program. We also conducted on-site evaluations of private properties that were managed under the guidance of the program. We conducted our study to 1) assess if the program effectively assisted participants in establishing and maintaining wildlife habitat, and 2) determine factors associated with optimal management of wildlife habitat on private lands to refine the program and improve effectiveness. We found significant differences between participants in their land use priorities and motivations for managing wildlife and in the resources available to participants to establish and maintain habitat. Our results indicate that although financial incentives may increase participation in private lands initiatives, improving technical and material assistance to landholders is essential for maintaining quality wildlife habitat over the long-term.  相似文献

In the aquatic liverwort Riccia fluitans the regulation of theplasma membrane H+/amino acid symport has been investigated.Cytosolic pH (pHc), membrane potential (Em) and membrane conductancehave been measured and related to transport data, (i) The releaseof [14C]amino acids is strongly stimulated by cytosolic acidification,induced by the external addition of acetic acid, a decreasein external K+, and in the change from light to dark. On average,a decrease in pHc of 0.5 to 0.6 units corresponded with a 4-foldstimulation in amino acid efflux. (ii) External pH changes havefar less effect on substrate transport than the cytosolic pHshifts of the same order. (iii) The inwardly directed positivecurrent, induced by amino acids, is severely inhibited by cytosolicacidification. (iv) Fusicoccin (FC) stimulates amino acid uptakewithout considerable change in proton motive force. (v) Whenthe proton motive force is kept constant, the uptake of aminoacids into Riccia thalli is much lower than when the pump isdeactivated. It is suggested that both the proton pump activityand cytosolic pH are the dominant factors in the regulationof the H+/amino acid symport across the plasma membrane of Ricciafluitans, and it is concluded that the proton motive force isnot a reliable quantity to predict and interpret transport kinetics. The leaf, stem, root, tuber and dropper anatomy of the orchid tribe Diseae (including the subtribes Satyriinae, Disinae, Brownlecinac, Huttonaeinae and Coryciinae) is reviewed. The study is largely based on investigations of 123 species, and data from several previous publications have also been incorporated. Two characters were identified as being taxonomically valuable: (1) the presence of sclerenchyma caps associated with leaf vascular bundles, and (2) the degree of dissection of the siphonostele of the tuber ('polystelic' or 'monostelic'). The phylogenetic analysis shows that anatomical characters do not change the basic structure of a cladogram that is based on morphological characters. The taxa of Diseae are discussed on the basis of anatomical data. Subtribes Satyriinae (excluding the anatomically unusual genus Pachites), Brownleeinae, Huttonaeinae, and Coryciinae are uniform in. critical anatomical characters. However, subtribe Disinae is rather diverse in vegetative anatomy. Disa sect. Micranthae differs from the rest of the genus in its leaf anatomy. The occurrence of foliar sclerenchyma bundle caps and 'polystelic' tubers supports the incorporation of Herschelianthe in Disa sect. Stenocarpa.  相似文献
